Impact of NKR-P1 polymorphism on Ly49 receptors expression in hybrid mouse strains (C57BL/6 x Balb/c, F10-12) Abstract Natural killer (NK) cells constitute the subpopulation of large granular lymphocytes which mediate spontaneous immune response against infected, transformed or allogeneic cells and thus represent an important component of the innate immunity. NK cells express a wide repertoir of surface receptors which can be either activating or inhibitory and which mediate NK cell recognition and regulation of cytolytic activity. NKR-P1 and Ly49 receptor families belong to the most important murine NK receptors. Both NKR-P1 and Ly49 families are members of C-type lectin-like superfamily of receptors encoded by natural killer gene complex (NKC) on chromosome 6 and include both activating and inhibitory members. The aim of this diploma thesis was to elucidate the impact of Nkr-p1c gene divergence on Ly49 receptors expression and to find out whether the Ly49 and Nkr-p1 gene clusters (which are localized on opposite ends of NKC) are inherited independently or whether the NKC domain is inherited as a complex. The second research interest was to illustrate the influence of the above mentioned divergence on cytotoxic activity of NK cells and tumor growth. Prognosis and diagnostics in acute myeloid leukemia

The main aim of the dissertation was in patients with acute myeloid leukemia to investigate the impact of favorable prognostic chromosomal aberrations and increased expression of a family of multiple drug resistance genes to achieve a complete remission and overall survival, and identify the potential prognostic factors. Experiments were focused on the introduction of routine diagnostics of two prognostically favorable fusion genes, AML1/ETO and CBFbeta/MYH11 and multiple drug resistance gene MDR1. This testing was performed by reverse transcription, PCR and subsequent electrophoretic analysis of its products.

Domestic Violence from the Perspective of the Aggressor

Domestic violence is a complex and widely spread phenomenon. Although this phenomenon is discussed, an unanimous and generally accepted definition of domestic violence has not been established so far. Domestic violence is violence among close relatives in families and households. There are various forms physical, psychological, sexual, economic, and others the combination of different forms is the most frequent. Typical features of domestic violence are: growing intensity, long termism, repeating, dividing roles and nonpublicity. It is gradually becoming a part of every day life and the cycle of domestic abuse is getting shorter. Participants in domestic violence are endangered people, violent people and children, or other members of household. Not only men are perpetrators of domestic abuse, but also women. In the Czech Republic there exist helping schemes especially for victims, but it is necessary to have a complex approach, because the whole family is concerned. Therefore therapeutic services for violent people, who do not manage their emotions and aggression in domestic context, are starting to appear. Thanks to these schemes, a change in their behaviour is possible; either the family may stay together, or it is help on letting go which is beneficial to all involved. The main goal of this bachelor thesis is to introduce the life tracks of individuals, who have become aggressors in domestic context, with an emphasis on subjectively perceived moments which caused this socially pathological behaviour. The research question was whether domestic violence appeared also in the primary family of perpetrator. Qualitative research strategy was used, data were gathered by half-structured or free interviews. The representative group consisted of domestic abuse perpetrators, who were chosen for this purpose. In the research five communication partners took part. Data were collected in crisis ambulatory service SOS Centre Diakonie CCE-SKP in Prague. The research results have shown that domestic aggressors´ primary families were in most cases not harmonious, and these people are unbalanced in a way. If domestic violence perpetrators are able to confess themselves they do not know how to deal with their emotions, and they voluntarily choose professional help and therapy, the chance to eliminate the primary reason of violence is bigger. This bachelor thesis could be beneficial for social workers involved in this issue. Outputs could be used as a study material for intervention centers and could be an impulse for more attention to domestic abuse perpetrators it would naturally help to other participants, mainly to victims.

The role of TRPV1 receptors in nociceptive signalling at spinal cord level

Modulation of nociceptive synaptic transmission in the spinal cord dorsal horn plays a key role in the development and maintenance of pathological pain states and chronic pain diseases. Important role in this process play Transient receptor potential Vanilloid 1 receptors (TRPV1), present on presynaptic endings of primary afferents in the superficial spinal cord dorsal horn. Changes in TRPV1 activity have significant impact on nociceptive transmission. There are number of processes that influence the function of spinal TRPV1 receptors. This work is focused on the role of protease-activated receptors type 2 (PAR2), C-C motif chemokine ligand 2 (CCL2) and the effect of chemotherapeutic drug paclitaxel in modulation of synaptic nociceptive transmission and activation of TRPV1 receptors. PAR2 receptors belong to a family of four G-protein-coupled receptors activated by proteases. The role of PAR2 receptors in pain perception is closely related to their presence in a population of dorsal root ganglion neurons, where they are also co-expressed with TRPV1. Activation of PAR2 may lead to peripheral and central sensitization. Title: Image of Austrian Society in the time of National Socialism: a Thematic Analysis of the Novel All our Games by Erika Mitterer Abstract: This diploma thesis deals with the work of the Austrian author Erika Mitterer and her experience with the ideology of the Third Reich. In the first part, I present the life and activities of the author, while putting a special emphasis on the period of National Socialism. The second part deals with the general characteristics of Mitterer's literary production. The main part of the thesis is the analysis of the novel All our Games, in which Mitterer tries to achieve a qualitative image of Austrian society during the Nazi regime. This picture is drawn by the Author using the example of a life of a fictive family. A general picture of the novel's time is constructed on the basis of the motives that are found in the book. Routes planning in Úslava microregion

Czech Republic has one of the densest and highest quality marked hiking trails in Europe, covering completely the whole country. But these trails are not planned with respect to the minority of the population with some kind of handicap. This is a group of families with small children who prefer hiking or biking, for families with strollers and disabled persons in wheelchairs. As the author of this thesis is a councilor of the village Jarov and also a councilor of this village in the Mikroregion Úslava organization he decided to plan a short circular hiking trails with respect to these neglected tourist groups in order to expand tourism in the area of Mikroregion Úslava. These routes may also partly overlap with marked hiking trails (due to their density) and are planned with practical experiences of the author. The results of the thesis will be incorporated into the information brochures of Mikroregion Úslava. The thesis is divided into theoretical and practical part. Theoretical part describes the methods for solving the travelling salesman problem and provides general information on the concept of micro-regions - voluntary unions of the municipalities. The practical part describes Mikroregion Úslava, the points of interest database creation procedure and planning sightseeing routes for families with children on foot, by bicycle or in strollers. There is also planned a route for disabled persons in wheelchairs in Blovice, which the author of the thesis eventually eliminated from the reasons described.

Fulfilling of bio-psycho-social need in clients with independent disorder in home care

Over and over again we are making sure how much is psychic support needed, to motivate patient (client) to regain health while hospitalized. We are finding out that family background cannot be replaced for any person (patient), not even by best medical and nursing care possible while hospitalized. Home care can (perfectly) unite expertise with support of family. Hospitals and home cares should be try satisfying all bio-psycho-social needs of clients. But it doesnt always work perfectly. Thats why i choose topic of bachelor work: satisfying of bps needs among clients with indep.dis. I collected informations from clients by asking them with questionaires. Goal of this work is to compare saticfacton of bio-psycho-social needs among clients with independent disorder in home care and those in hospital. Goal was build based on conditions, that biological, psychical a social needs are better saturated in home care than in hospitals. This work should inform about advantages of home care for clients and I wish it can help both medics and ordinary people who take care of people with independent disorder.

Redistribution aspects of benefit

In my bachelor work I deal with social security benefits especially from the point of redistributing. In the same time I pay attention mainly to families with children. Each of us has to realize that receiving and paying of particular benefits is an integral part and also a principle of our living standart. The first part of my work delimits theoretical terms and explains the construction of various benefits and the conditions for receiving them. Further practical part is dedicated to specific analysis of some section of social security. In the last part I mention practical examples for calculaton of particular benefits and their payments.

Post-synthesis synthesis: Czech kinetic art of the 60's and the following creative techniques of its protagonists

The study analyses situation on Czech, formerly Czechoslovak, art scene during 70's and 80's of the twentieth century in context of the progress of kinetism. It addresses the question whether and how the kinetism as a synthesis of visual arts, architecture, theater, film and music but also science and technology has been promoted after the demise of the group of the same name. The study is divided into the three basic parts. After introducing the problem by attempt to delimit the kinetism on conceptual and temporal basis the first part follows the initial, rather sporadic, attempts of capturing the motion in the visual arts in a period of the first half of twentieth century. The substantial focus of this part is on the work of Zdenek Pesanek. The second part provides a brief insight by comparison and references into the aggradizement period of international kinetism that occurred in the 50s and through to 70s. In parallel with movement in the West it follows two particular groupments of kinetists in the Eastern block - the Muscovite group called Dvizhenie and the Prague's group Synthesis. Amount of inluence of the kind of leisure time activities of fifteen year old children on manifestation of agressiveness

The bachelory thesis brings the analysis of spending 6oustř time of 6oustř year old children with relation to the 6oustřednou agression and bullying, which is a topical 6ous nowadays. The goal of the thesis is to determine whether the way of spending 6oustř time, namely the fact that 6oustř year old children spend 6oust free time playing 6oustřed games or watching television programmes including 6oustřednou violence, inluences 6oust agressiveness. The thesis deals with the 6ous of spending 6oustř time of 6oustř year old children, the inluence of family on 6oustř year old children and also the development and the reasons of agression in the life of a child and bullying in its 6oustř forms and stages.
