Value orientation of sporting and non-sporting seniors

Problem: The problems of values have been in the forefront of human interest since ancient philosophy. Due to the serious nature of this topic it is not a surprise that values are presently the focus of nearly all social sciences (psychology, economics, sociology, anthropology, ethnology etc.). However, value orientation in seniors still represents a relatively not very explored area. In this respect, the value orientation of seniors in relation to sport and the barriers inhibiting their participation in locomotion activities in particular is a topical issue in the light of the present-day demographical development in the developed countries. Hypothesis: We presume that we find relationships between the value orientation of a selected senior population and their degree of participation in locomotion activities, the type of locomotion activities and socio-demographic characteristics. We also presume that we find relationships between the value orientation of a selected senior population and their subjective perception of barriers limiting or inhibiting seniors' involvement in locomotion activities. History of Znojmo's brewery (1363-2010) - abstract The content of this Bachelor Degree Thesis is the complete history and building development of the only remaining Znojmo brewery in the period of 1720 to 2010, a brewery that is no longer operating. At the same time, this thesis describes the history of brewing in Znojmo begining from the first sings in the 13th century (emphasising 1363 which was a legendary year for Znojmo's brewing) till 2010 when the last brewery in Znojmo was completely closed and the tradition of brewing in Znojmo lasting for almost 700 years was interrupted. The following chapters describe for example the origin of the name "Hostan", the present name of Znojmo's beer, concentrating to all products, once produced in Znojmo brewery, and awards that once famously delicious Znojmo's beer Hostan received all round the Czech Republic. The thesis completely summarizes all the years of Beer Festival - a modern tradition that arose thanks to the existence of brewery and ceased, when the brewery was closed. Applications of real-time PCR for characterization particles suitable for DNA isolation

The theoretical part of the diploma thesis was focused on core-shell type magnetic carriers, used mainly in medical, molecular-biological and biochemical applications. Encapsulation of the core is essential for these applications due to the decrease od non-specific protein adsorbtion, increase of biocompatibility and the possible functionalization of magnetic carriers. In the experimental part, the DNA (E. coli) was amplified by real-time PCR in the presence of poly(hydroxymethacrylate-co-glycidylmethacrylate) (P(HEMA-co-GMA)) magnetic carriers with/without carboxyl groups. The inhibitory effect of different concentrations of magnetic carriers in the PCR mixture was evaluated from the calibration curve parameter values obtained by regression analysis. The presence of a specific PCR product was verified by agarose gel electrophoresis. Most of magnetic carriers without carboxyl groups extinguished the fluorescence in the concentration range of 2,0 – 4,0 g.l-1 in the PCR mixture, without inhibition of DNA amplification - the carriers were biocompatible. Magnetic carriers with carboxyl groups extinguished the fluorescence in the lower concentration range (0,4 – 4,0 g.l-1 in the PCR mixture). Their inhibition of amplification was in the concentration range of 2,0 – 4,0 g.l-1 in the PCR mixture, from the concentration 0,8 g.l-1 in the PCR mixture, the inhibition did not occur and the carriers were biocompatible. The results do not depend on the characteristic properties of the magnetic carriers but on the presence of the carboxyl groups on the surface of the carrier and the degree of coverage of the magnetic core by the polymer. Real-time PCR has become an effective tool for studying magnetic core encapsulation and the influence of functional groups on the surface of the polymeric layer.

This thesis - Contrastive Topics in Czech - describes some of syntactic and semantic characteristics of the experessions, called Contrastive Topics. Following theory of Functional Generative Description (p. Sgall, E. Hajičová and J. Panevová), author is trying to Bnd a ways to explain and verify several hypotheses of FGD by searching data of Prague Dependency Treebank. Prague Dependency Treebank is collection of czech sentence s, annotated on three levels of depth, namely the morphemic layer, the surface shape of sentences and tectogrammatical layer. The deep syntactic structure annotations in the Prague Dependency Treebank, the so-called tectogrammatical tree structures, capture three basic aspects of the underlying structure of sentences: the dependency tree structure, the kinds of dependency syntactic relations, and the basic characteristics of the topicfocus articulation. This Bachelor's degree thesis focuses on drug aproval process in the Czech Republic and researches consumer's awareness about this process. Before active substance becomes a drug and gets to a consumer who needs its effects it has to go through a long process which begins with research and which continues through clinical studies, drug aproval and various logistical chanels. There is a process of marketing authorisation on nationally authorised medicinal products described at this thesis. Practical part of this thesis deals with a research done. The purpose of research was to find out the level of awareness of consumers about the process of drug aproval and other matters that are conected to marketing authorisaton process as package leaflet, labelling and pharmacovigilance.

Influence of Al ionts in nickel hydroxide on behaviour of electrodes

This master's thesis is concerned with the influence of aluminate ions added in various concentrations into the structure of nicel hydroxide that creates Ni-Cd accumulators positive electrode active mass basis nowadays. Nicel hydroxide occurs in two basic modifications ? and ß. Nowadays used ß modification reaches low capacity in comparison with ? modification. The disadvantage of ? modification is its instability and gradual degradation to ß modification. The aim of this diploma paper is to select the optimal amount of aluminate additives into the structure of nicel hydroxide with regard to electrochemical efficiency, stability, and degradation degree of the accumulated layer.

Liveness Detection of a Finger Based on Changes of Papillary Lines

There are several frauds against biometric systems (BSs) and several techniques exist to secure BSs against these frauds. One of the techniques is liveness detection. To fool fingerprint sensors, latent fingerprints, dummy fingers and wafer-thin layer attached to the finger are being used. Liveness detection is being used also when scanning fingerprints. Several different characteristics of the live finger can be used to detect liveness, for example sweat, conductivity etc. In this thesis, new approach is examined. It is based on the expandability of the finger as an effect of heartbeats/pulsation. As the skin is expanding, also the distances between papillary lines are expanding. Whole finger expands approximately in range of 4,5 ľm , the distance between two neighbor papillary lines in 0,454 ľm . This value collides with wavelength of blue and green light. The result from this work is following. The resolution of the capturing device is not high enough to capture the expandability on distance between two neighbor papillary lines. Also, because of collision with wavelength, the diffraction effect is presented and the result images are influenced by this error.

Advanced Heterostructured Ceramic Materials

The doctoral thesis is orientated to preparation of advanced ceramic materials by electrophoretic deposition (EPD). The vertical mode of EPD in constant current regime was used in this study. The morphology of prepared materials (image structural analysis), physical properties (EPD yields, relative density, roughness of deposit surface) and mechanical properties (hardness, fracture toughness, elastic module) were characterized on the prepared deposits from alumina (Al2O3), zirconia (ZrO2) or their composites. It was found that the surface charge of alumina or zirconia particles was opposite (inversion charge) in stabilized isopropanolic dispersions than in case of water dispersions. The model of alumina or zirconia particles “inverse” stabilization was proposed. This type of EPD was further studied in presence of different acidic stabilizers. The obtained results from the experimental work (-potential and electric conductivity of dispersions, EPD yields, relative density of deposits, roughness of deposit surface etc.) showed the monochloracetic acid as the optimal type of acidic stabilizer. The kinetic of EPD process from dispersions containing isopropanol, monochloracetic acid and Al2O3 or ZrO2 particles and its influence on the final properties of prepared deposits was studied. The new method of kinetic measurement was developed. Due to good knowledge of EPD kinetic the ceramic laminates (Al2O3/ZrO2) with optimized layer thickness were prepared. The internal stresses and their influence on crack trajectory in ceramic laminates contained 100-150 strongly bonded layers were studied. It was found that the dominant role of crack deflection played the internal compressive stresses, i.e. with increasing of amount of internal compressive stresses the crack deflection was also increased. This work brought a new knowledge in the area of non-aqueous ceramic dispersion stabilization and preparation of heterostructured ceramic materials.

Multimodal Registration of Fundus Camera and OCT Retinal Images

Tato práce se zabývá multimodální registrací snímků sítnice z různých skenovacích zařízení. Multimodální registrace umožňuje zvýraznit prvky na snímcích sítnice, které jsou důležité pro detekci různých typů onemocnění oka (jako je glaukom, degradace nervových vláken, degradace cév, atd.). Teoretická část tvoří zhruba první půlku práce a je následována praktickou částí, která popisuje postupy při různých typech registrací snímků z fundus kamery, SLO a OCT. Registrace fundus a SLO snímků je provedena pomocí prostorové transformace. Tato práce popisuje tři různé metody registrace SLO snímků se snímky z fundus kamery. První a zároveň nejjednodušší je manuální registrace. Druhou je automatická registrace založená na metodě korelace. Výsledky, včetně porovnání obou metod, jsou uvedeny v závěru. Třetím typem je poloautomatická registrace, která využívá výhod obou předchozích metod a tím pádem je kompromisem mezi rychlostí a přesností registrace. Registrace fundus snímků a B-scanů z OCT je realizována dvěma různými metodami. První je opět založená na korelaci a druhá na prostorové transformaci. Všechny tyto registrační metody jsou realizovány také prakticky v programovém prostředí Matlab.

Transfection of stem cells and other testicular cells of Xenopus tropicalis

Xenopus tropicalis represents one of the most important model organism used in developmental and cellular biology. Laboratory of Developmental Biology at the Faculty of Science, Charles University in Prague has successfully established a long-term culture of X. tropicalis juvenile testicular cells. Based on expression profiling analysis of selected specific markers (Sox9, WT1, etc) it was proven that the major cell type in this culture is pre-Sertoli cells. Furthermore these pre-Sertoli cells allow a longterm cultivation of the germinal stem cells. By performing a histochemical test for the presence of alkaline phosphatase in the colony of these cells were proven the features of stem cells. In this diploma thesis we focused on optimization of work with the mixed cell culture. In particular we define conditions of dissociation and subsequent separation of a feeder-layer formed by the pre-Sertoli cells. We also attempted to develop suitable conditions for transfection of the germinal cells. With these techniques we will to investigate the functional properties of the germinal stem cells.
