Resumé / English summary Title of the thesis: Optimal capital structure of an enterprise Summary: The purpose of my thesis is to analyse optimal capital structure of an enterprise and important factors in the capital structure decision making, because the enterprise's capital structure is one of the most important decisions management has to make. The capital structure is how a firm finances its overall operations and growth by using different sources of funds and the capital structure optimisation is relatively a complex issue. The thesis is composed of eight chapters. Chapter One is introductory and explains my choice for the purpose of this thesis and describes content of following chapters. Chapters Two a Three define basic terminology used in the thesis, particularly the basic items of the property and capital structure of an enterprise. The aim of Chapter Four is to survey the most important capital structure theories, especially the Modigliani-Miller theorem (of Franco Modigliani, Merton Miller) that forms the basis for modern thinking on capital structure. Chapter Five describes the most significant determinants of optimal capital structure of an enterprise and how those factors might affect optimal capital structure. Preparation of the 14-3-3 Protein Binding Partners for Structural Studies.

Tyrosine hydroxylase belongs to the group of hydroxylases of aromatic acids and catalyzes a key step in the biosynthesis of catecholamine neurotransmitters. The tyrosine hydroxylase possesses the homotetrameric structure and contains three structural domains: the N-terminal regulatory domain, the catalytic domain and the C-terminal tetramerization domain. The activity of tyrosine hydroxylase is regulated by phosphorylation and through the regulation of its expression. Phosphorylation at Ser-19 induces binding of the 14-3-3 protein, which affects the structure of the regulatory domain and protects it against both dephosphorylation and degradation. Since the structure of the regulatory domain is still unknown, we decided to perform its structural characterization using NMR techniques. First, the expression and purification protocol of the regulatory domain of tyrosine hydroxylase was optimized. The protein was expressed as a His-tag fusion protein and its purification is composed from two steps: the chelating chromatography and the size-exclusion chromatography. The dynamic light scattering and the 1 H nuclear magnetic resonance were used to verify its monodispersity, and hence its suitability for further experiments.

Structure and function of chloroplasts in selected woody plants grown under increased CO2 concentration

The effect of elevated CO2 concentration (EC CO2) on photosynthesis has been observed on many hierarchical levels. There was a significant increase in the rate of photosynthesis of examined trees observed in previous studies thus I hypothesised these changes are accompanied by changes of chloroplast ultrastructure and photosystem content and function and the main aim of this study is to evaluate these adjustments. In this study 13 - 14 years old seedlings of Norway spruce (Picea abies L. Karst.) grown in glass domes with adjustable windows - one with ambient CO2 concentration as a control, the other one with simulated EC CO2 (700 ppm) - during the vegetative season were examined. Pigment content, fluorescence and reflectance indexes, activity of photosystem 1 (PS1) and 2 (PS 2) of isolated chloroplasts, size of cross-sectional area of chloroplast and proportion of stromal to granal thylakoids under EC CO2 treatment were investigated. Although there was a significant increase in the maximum rate of photosynthetic assimilation of trees from EC CO2 (observed by other researchers of our team), decreased chlorophyll and carotenoid content as well as the activity of both photosystems were observed, which is usually atributed to photosynthetic acclimation. Mechanical behaviour of variably saturated clay fills VLADISLAVA KOSTKANOVÁ Ph.D. Thesis - English Abstract The excavated overburden from open-cast coal mines is end-dumped in fills. Clay type soil prevails among the filled soils in North-western Bohemia. As a consequence of the character and state of the soil, together with the mining and dumping technology, a typical lumpy structure of the clay fill is formed. The filled lumps range in size and vary in mechanical properties, but stiff to solid lumps up to 0.5 m in size are prevalent. Large and open macrovoids between the single lumps are a characteristic feature of the clay fills. Clay fills exhibit a double porosity structure, consisting of intergranular porosity between the lumps and intragranular porosity inside them. The total porosity of the freshly filled soils can reach 70%. This type of clay fill structure is metastable. This thesis focuses on the problematic behaviour of clay fill soils due to their metastable structure and their partly saturated state. Long-term monitoring of matric suction at depths up to 2 metres in two reclaimed clay fills revealed two different trends. Seasonally-dependent suction oscillations were recorded in the moderate slope of the clay fill in the Bílina mine, while no suction pointing to fully saturated conditions was...

Evolution of the Czech labour market in 2004-2014

This diploma thesis aims to analyze and evaluate the structure of the labour market in different economic cycles in the Czech Republic during 2004-2014 and compare reciprocity with German labour market, which is closest to the Czech. The theoretical part deals with general issues in the labour market, the economic cycle and the development of the global economic crisis. The work confirms the hypothesis that nowadays in 2014, four years after the economic crisis that hit the Czech economy in 2008-2010, the structure of the labour market in the Czech Republic has changed compared to the years before crisis. It also partially confirms the minor hypothesis that labour market indicators still haven´t reached the level which Czech Republic had before the crisis. The unemployment rate is still not at pre-crisis levels, but employment improved compared to 2008

DSS for energy saving of potential clients

This diploma thesis focuses on a model-oriented system for decision support. The main contribution of the thesis consists in creating a system simulating energy saving which would result from the purchase of plastic windows and/or of a heat pump. The basis of the simulator is a functional model consisting of individual variables and connections between them and created in compliance with relevant notation. The thesis describes and illustrates not only the model structure itself, but, in the same time, also its logic on which basis the corresponding energetic saving is calculated after entry of necessary initial values. The diploma thesis includes, in addition to the model, also the description and the graphical representation of the created user interface which provides the user with easy entry of parameters and clear display of results. The introductory part of this work introduces the basic theoretical terms and approaches concerning the system dynamics together with its principles. The following part is dedicated to the description of the enterprise for which the system for decision support was created. The final part contains the above mentioned model, its structure and description, as well as the created user interface.

Content and structure analysis of Czech material science journals abstracts : Hutnické listy and Ceramics-Silikáty

An abstract is a reduced scholarly document crucial for relevant scientific articles and full text documents selection. Therefore abstracts should briefly and clearly represent the full text content. The aim of this research is to construct proper structure, which would be internationally as well as interdisciplinary applied, primarily within the European Union. The structure used in the research was supposed to be evaluated as the most suitable one by the experts. There were 2 material science journals (100 abstracts from each journal) selected. Those samples were analysed in freeware Weft QDA, designed for content analysis of text documents. Sample abstracts were based on the content analysis resulting data. The samples were evaluated by material science experts in the survey. The most common combination of categories in original abstracts were background, methodology and results. The repsonses of surveys pointed out, that the recommended abstract include all necessary information and exclude the redundant ones.

Sport Manager Simulator

The first part of presented work contains a description of the technological structure of the web application - a system for virtual simulation of a sport world controlled by user's active management of a team. The structure is divided into multiple layers - presentation, appliction and database. Use and utilization of particular frameworks on these layers are described in the work. In the second part we examine the implementation of the model used to generate the course of the match taking into account the parameters influenced by the managing abilities of users owning their competing teams. We introduce Markov models, their use in our model, connection between coincidence and observation of the situations and we perform statistical analysis of the results.
