Thermal analysis of interaction of epidermal lipids V Petra Kaloušová, degree work , June 2009 Abstract Natural function of human skin is to protect the body from adverse influences of the surrounding. In 1950's and 70's it was discovered, that the skin barrier is situated in stratum corneum and is determined by lipids in intercellular space of that layer. Transdermal application of medicaments has many advantages compared to peroral one. On the other hand stratum corneum is a certain barrier for penetration of the medicaments. Thus we have focused on this lipid layer and its interaction with water. We have used samples prepared in vitro consisting of cholesterol, pseudoceramid and modulators of transdermal penetration. We have tried to hydrate the samples and tracked their characteristics and interactions by differential scanning calorimetry. At the same time we have evaluated the individual groups of drugs and proper experimental conditions for that particular measurement.

Skin equivalents in basic and applied research: Development of human hair equivalents

In this study, three novel organotypic human folliculoid assays for basic and applied hair research were developed. Two organotypic "sandwich" systems consist of a pseudodermis (collagen I mixed with and contracted by human interfollicular dermal fibroblasts) on which one of two upper layers is placed: either a mixture of Matrigel™ and follicular dermal papilla fibroblasts (DPC), with outer root sheath keratinocytes (ORSK) layered on the top ("layered sandwich" system), or a mixture of Matrigel™, DPC and ORSK ("mixed sandwich" system). Third, the patented new technique generating human folliculoid microspheres (HFM), consisting of human DPC and ORSK within an extracellular matrix was established. Studying a number of different markers (e.g. proliferation, apoptosis, cytokeratin- 6, versican), it was shown that these folliculoid systems, cultured under well-defined conditions retain several essential epithelial-mesenchymal interactions characteristic for human scalp hair follicle. Selected, recognized hair growthmodulatory agents modulate these parameters in a manner that suggests that all developed organotypic systems allow the standardized pre-clinical assessment of test agents on relevant human hair growth markers under substantially simplified in vitro conditions that approximate the in vivo situation....

Form and function of nouns in Czech: relation between nominal case and syntactic function. Based on a synchronic written corpus of Czech (SYN2005)

The case in Czech is the basic morphological means by which nouns express their function in a sentence. The objective of this thesis is to describe, from a frequency point of view, the relation between form and function of nouns, or, more precisely, how frequently cases (both simple and prepositional) are used to realise syntactic functions in sentences. The thesis is based on one of the largest corpora of written synchronic Czech: 100-million-token corpus SYN2005. In order to obtain data on frequencies of syntactic functions of nouns in relation to their cases, we annotated the corpus SYN2005 with a dependency syntactic annotation. For this annotation, we adopted the format of the analytical layer of the Prague Dependency Treebank. The syntactic annotation has been performed by a stochastic parser: the MST parser. Since the reliability of this annotation was not high enough, we have built an automatic correction module, which identifies errors of syntactic annotation in the output of the stochastic parser and corrects these errors by means of linguistic rules. We have implemented 26 different rules, but annotation errors have been reduced by merely 6-8%. However, this correction module can be further developed. Transport of charged and neutral particles across the model biomembranes

This work was focused on the preparation of model stabilized phospholipid membranes formed on porous polycarbonate carrier. 1,2-dipalmitoyl-sn-glycero-3-phosphocholin was used for their formation in hydrophilic pores of polycarbonate carrier. For characterization of the formation of phospholipid layers, their changes and a study of transport processes, electrochemical impedance spectroscopy and voltammetry were used. Transport of cadmium and copper ions was studied in the presence and in the absence of ionophore calcimycin which was incorporated into the formed of phospholipid membrane. Because these ions are often bound in complexes with various substances, such as low molecular weight organic acids (LMWOAs), this work was also focused on the transport of copper and cadmium ions across the model phospholipid membranes in the presence of malic acid, citric acid and oxalic acid at different pH. Besides the use of ionophore, some pilot experiments were performed to realize the transfer of copper ions using two peptides, nisin and transportan 10. Formation of phospholipid membranes and the transport processes were characterized by two proposed electric equivalent circuits which correspond to the covered and to the uncovered polycarbobate carrier. Unified Editor for CEGUI

This thesis presents a free software GUI application called the CEGUI Unified Editor. The application is mainly written in Python and is licensed under GPLv3. Its purpose is to create and modify assets of graphical interfaces made with the CEGUI library. Features include project management, imageset editing and layout editing. Data for older versions of CEGUI are transparently converted using compatibility layers. Big emphasis is put on ease of use, collaboration between multiple content authors and portability.

Splitting user interface layer to client and server part

Web applications can be divided into two basic groups by the approach to the data transfer. Applications from the first group have their entire logic at the server side. That implies large data transfers between clients and the server. Applications from the second group duplicate part of their logic at the client side. That simplifies the communication but on the contrary it then requires more complicated development and maintenance. The goal of the thesis is to develop an application model combining the advantages of both approaches for the most common application scenarios.

Measurement of the asynchronous motor

The work deals with the methods of measurement of various types and characteristics of induction engines is divided into three main chapters. The first chapter introduces us to the history superficially induction engines and the direction of work. The second chapter is devoted to different types of tests on this induction engines. The third chapter lists the results of measurements on two engines also known as type AOM090L02-016 (which is equipped with single layer winding) and type AOM090L02-017 (which is equipped with a double layer winding). In the end this work describing measurement of torque characteristics from run-up characteristic by using PC and freewheel.

Development of DMS module for digital advertising agency

This work documents the whole process of developing an information system for a small advertising agency. Its main sphere of business is the internet, hence the company is classified as a digital or interactive agency. My objectives are: analyzing business processes, assembling the functional specification and developing the core of the application and the module for a document management. I start with gathering some common parameters of DMS along with requirements for a web application framework, which is intended to use for a base of the intranet. Then I move to presenting specifics of our agency and I analyze its processes in order to capture all content and desired functionality of the developed system. Next phase is dedicated to design. First, I set the intranet in the context of global Enterprise architecture. I continue with modeling all architectural layers (conceptual, technological and implemental). The application is modular and benefits from using MVC architectural pattern. DMS module implements workflow functionality and I have used principles of Workflow Management Coalition to design that features. The last part of my work describes developed modules in more detail. My objectives were accomplished, core of the intranet is developed and currently in process of testing.

Characterisation of Rat Alveolar Cell Line R3/1

The rat cell line R3/1 displays several phenotypical features of alveolar epithelial type I cells. In order to characterise the cell line as an in vitro model for drug disposition studies, R3/1 cells were cultured on Transwell filters and the transepithelial electrical resistance (TEER) was measured as a parameter for the integrity of cell layers. Presence of cell junctional proteins (i.e., E-cadherin, occludin, ZO-1 and ZO-2) was studied on mRNA (by reverse transcriptase-polymerase chain reaction, RT-PCR) and antigen level (by immunofluorescence microscopy, IFM). Moreover, the expression pattern of catabolic peptidases (carboxypeptidase M, aminopeptidases A, B, N and P, γ-glutamyltransferase, dipeptidylpeptidase IV, angiotensin-converting enzyme, and endopeptidases 24.11 and 24.15) was analysed in R3/1 cells and compared to rat alveolar epithelial I-like cells in primary culture. TEER values were peaking at ~99±17 Ω cm2 after 5 days in culture. Addition of 0.1 µM dexamethasone together with foetal bovine serum at 20% increased TEER by 65%. However, none of culture conditions used in our study yielded monolayers with TEER values comparable to those of primary cultures of rat pneumocytes. Celá léta mi zní v hlavě dvě melodie, zpívám si je, každý den si na ně vzpomenu. Až když jsem viděla video z roku 1990, tedy 18 let zpátky, zjistila jsem, že toho roku (tedy v mém prvním pelhřimovském roce) jsme tyto dvě písně nacvičovali. A asi by mi v hlavě zůstaly, i kdybych byla na kurzu jen jednou.... V Pelhřimově se schází sociálně rozmanitá skupina lidí - zhruba půl napůl zastoupena středoškoláky a vysokoškoláky; jsou to lidé s hudebním vzděláním i bez; lidé majetní i nemajetní; někteří mají pobyt v kurzu namísto dovolené a šetři na to celý rok. Děti se naučí komunikovat s dětmi chytrými, hloupějšími, problémovými, neproblémovými...prostě, je tam průřez všemi možnými vrstvami. Kurz "Rodina zpívá, hraje a tancuje" je nepřebernou studnicí nápadů a inspirace pro učitele. Ti zde mohou vyzkoušet sami na sobě a na dětech vše v praxi, vidí, jak se ta která věc dětem líbí a pro koho se spíš hodí, včetně úskalí, na která mohou narazit. Mnozí učitelé mají pelhřimovský zpěvníček jako náhradu za oficiální materiály a pomáhají jim v návratu lidové písni. Také tematická utříděnost ušetří hodně práce.
