Modal analysis of tall slender structures using software GMAST
Hračov, Stanislav ; Pospíšil, Stanislav ; Brůna, Tomáš
The paper is concerned with a verification of modal properties of tall slender structures computed by recently developed software GMAST. Software GMAST determines eigen-frequencies, eigen-modes and corresponding modal quantities of stacks, towers, guyed masts and similar civil engineering structures using slope-deflection method. A numerical model is based on an idealization of the structure as a continuous 2D beam with continuously distributed parameters on elastic supports. Shaft of structure is divided into sections which could be considered as prismatic i.e. with constant unit mass, bending stiffness and static axial force. The effect of guy ropes is introduced in a form of elastic supports with a frequency dependent stiffness. The applicability of the software in praxis is presented on an example of existing structures, for which the numerically and experimentally identified modal properties are compared.

Synthesis of novel cardioprotective iron chelators
Hrušková, Kateřina ; Roh, Jaroslav (referee) ; Vávrová, Kateřina (advisor)
Oxidative stress participates in patophysiology of many serious cardiovascular diseases. Free intracellular iron occurs as a catalyst of Haber-Weiss and Fenton reaction between superoxides and peroxides increasing the formation of highly toxic hydroxyl radicals, which cause cell damage. Iron chelators diminish the pool of free iron and thus become perspective agents in therapy of various diseases, e.g. anthracycline-induced cardiotoxicity. Aroylhydrazones group, such as salicylaldehyde-isonicotinoylhydrazone (SIH), appear to be highly efficient chelators. Unfortunately, pharmacokinetic studies focused on these compounds revealed their low stability in plasma. Therefore, I synthesised a series of SIH analogs in order to increase their stability together with preserving the ability to chelate free intracellular iron and to define their structure-activity relationships. A basic hypothesis in design of the novel chelators was using substituted ketone instead of aldehyde, leading potentially to an enhanced stability of hydrazone bond. SIH Two different methods were used during the reactions, a conventional heating in an oil bath and heating in a microwave reactor. The latter caused a significant shortening of the reaction time. In vitro studies of novel compounds showed their higher stability in plasma,...

Determination of vitamine C and dehydroascorbic acid using UHPLC-MS method
Proroková, Zuzana ; Chocholoušová Havlíková, Lucie (referee) ; Nováková, Lucie (advisor)
A coupling of Ultra High Performance liquid chromatography with mass spectrometry provides a technique, which is rapid and sensitive. This thesis is focused on the use of UHPLC-MS for the determination of ascorbic acid (AA) and dehydroascorbic acid (DHA). AA is a small polar molecule that acts as an antioxidant. After oxidation AA creates DHA. AA/DHA ratio is an indicator of a redox state of organism. For the determination of AA and DHA several methods have been developed, which usually do not allow the simultaneous analysis, but require multistep subtraction procedure. The optimization of UHPLC-MS method for the determination of AA and DHA include the choice of mobile and stationary phase and mass spectrometry detector set- up. The choice of appropriate conditions depended mainly on retention time and the detector response. The effect of stationary phase, concentration, pH, composition of mobile phase on retention of AA and DHA was observed. Effect of mobile phase on stability of AA and DHA was observed as well. BEH Shield RP C18, BEH HILIC and BEH Amide column were compared. The best results were achieved on column BEH Shield RP C18. Measurements with 0.1%, 0.05%, 0.01% formic acid, 0.1%, 0.05%, 0.01% acetic acid, ammonium formate at pH 3.5 and amonium acetate at pH 4.4 and 6.8 as a water...

Addressing the mass of animal diseases within the scope of municipalities with extended powers and the district.
PLACEK, Jiří
Mass infections in animals are presently a very common occurrence, as the society faces up to these extraordinary situations more and more frequently. Direct hazards to life, health and property can take place in the case of animal mass infections, mostly however secondary impacts upon the management, economy and psyche of the stock farmers, who can even lose large numbers of animals in a relatively short period, temporarily taking the stock-farming equipment out of service. My thesis deals with solving the issue of the disposal of animal mass infections within the area of municipalities with extended scope of powers. Such activities for particular areas have already been processed in the emergency and crisis plans of the region. Furthermore, I look into various possibilities of animal disposal, as well as the activities of the Integrated Rescue System{\crq}s individual bodies, federal agencies, self-government bodies and legal entities when dealing with such events. One of the possibilities to dispose of infected animals is using the unit for production of non-combustible gas. First designed to put out fires of hay stacks, this unit can be also used to dispose of infected animals, e.g. breeding poultry and piglets. The principle of this method lies in forcing out the oxygen from a closed area by the use of carbon dioxide, or possibly developing carbon monoxide, resulting in the death of the animals. As far as the economic point of view is concerned, this procedure is less demanding, it however requires a thorough sealing off of the area.

Designing mass modification of client contact records and theirs consolidation within financial institution
Převrátil, Martin ; Kunstová, Renáta (advisor) ; Jeřábek, František (referee)
This master thesis deals with management request towards informatics department within front company that provides wide range of financial services. Demand refers to client contact data, particularly correspondence addresses and calls for possibility to mass change such records. There are several relatively separated tasks which need to be discussed in order to fulfill the request. First part introduces the company and analyze current situation. Then continue by describing correspondence addresses issue and supports with reasons for the inevitable change. Company's information system architecture is outlined with emphasis on central database that is being formed. This part also further describes options how data can be stored. Middle part contains reason--based choice where correspondence records should be maintained in company's data architecture. Implementation plan with chosen storage pattern is formed here and connected changes in system are highlighted. As a last processes connected to correspondence addresses mass modification are described and drawn with selected notation. After that user graphic interface is designed as a place where the self request will be served.

DSLR Video and its Impact on the Conventions in Filmmaking
Nuska, Petr ; Získal, Bohuš (advisor) ; Šlerka, Josef (referee)
The thesis deals with the involvement of digital single-lens reflex cameras (DSLR) with video-capturing capacity in documentary and ethnographic film, more particularly, it examines whether and to what extent these genres were influenced by a trend called DSLR revolution. The main contribution of the work is a qualitative analysis of the testimonies of ten filmmakers, who used DSLR cameras while working on their films during the years 2010-2014. The synthesis of this analysis is conceptualised as a reconstruction of the process of decision-making whether to use or not to use a DSLR camera, considering its benefits (affordability, technical and aesthetic qualities, compactness and simplicity) and its limits (ergonomics, visual artifacts, workflow limits). The reconstruction of this decision-making process is considered in the context of specific experiences of the participants and it focuses on the impact of DSLRs on filming workflow and final product. The final chapter deals with the DSLR revolution as a social trend which was stimulated by the influence of online communities of DSLR filmmakers who actively participated in the course of the revolution. Appendix of the thesis contains a brief description of the film projects of all participants and the model of the structured questionnaire used for data...

NATO and WMD: Evolution of the threat construction and Alliance's response
Hrdinová, Lucie ; Rychnovská, Dagmar (advisor) ; Ludvík, Jan (referee)
This Diploma Thesis called "NATO and WMD: Evolution of the threat construction and Alliance's response" deals with the evolution of understanding of Weapons of Mass Destruction (WMD) within the North Atlantic Treaty Organization (NATO). WMD, especially the nuclear ones, have preoccupied NATO since its establishment. However, it was only after the end of the Cold War that NATO started to focus on WMD as a specific overarching category, consisting of chemical, biological and nuclear (since 9/11 also radiological) weapons. Despite the conspicuousness of the concept and increased use of the WMD term, the definition of weapons of mass destruction is not clear-cut. However, the way the actors understand WMD is very important. For this reason, NATO's official discourse is studied as an indicator of how is WMD understood in the Alliance, which will allow to fulfill objectives of this thesis. First, the work intends to analyze how has the WMD threat been constructed and interpreted within the Alliance since the year 1991 until this day. Secondly, this thesis aims to find out what is NATO's response to the dangers posed by the identified WMD threats (i.e. proliferation and terrorism) and identify the concrete measures the Allies undertook to tackle the evolving WMD threat. This thesis is based on...

HOUSE ON THE EDGE – Valašské Meziříčí, Sokolská Street
Zabadalová, Kateřina ; Mutina, Petr (referee) ; Jura, Pavel (advisor)
The designed building is based on the original historical buildings and the scale of adjacent historic center. The plot, on which originally were several independent row houses, by its size greatly exceeds the scale of surrounding buildings. Land is transversely divided into seven parts, which are based on the original allotment of the land. The mass of the object creates a complete town house, whose scale corresponds to the surrounding buildings. From the height perspective, the house is divided into three and four storey. Individual parts by its dynamic shifting give rise to large area of glass shop windows in the parter and also allow to illuminate the object from the west and east side. The existing car parking enables a pedestrian passage from the center to the adjacent residential buildings on the south side. For this reason, at the busiest pedestrian routes there are free passages through the parter. They are also used as a sheltered entrance to the individual parts of the apartment house. The entrance to the underground garage from the street Polášková is due to good infrastructure. Supplying of the neighboring object is preserved in its current form, as well as the shop windows. Parterre, thanks to the overall glazing, relieves the house, creating a clear division between public and private part and become part of the street. Areas created by the division of the object extend the street and form a diverse corners for people passing. The individual flats themselves are defined by the shape of the mass itself. They differ both in size, shape and orientation. They offer from the smallest studios to maisonettes with access to the walkway roof of the lower mass.
