Expression of different forms of actine in some non-muscular and muscular tissues and tumors

Actins are ubiquitous eucaryotic proteins. Actin filaments are involved in diverse functions which include cell contraction, motility, adhesion, division, cell shape maintenance and muscle contraction. Four actin isoforms are tissue specific (α-smooth muscle actin, α-cardiac actin, α-skeletal actin and γ-smooth muscle actin). Two other actin isoforms (cytoplasmic β- and γ-actins) are ubiquitous. Alpha-smooth muscle actin (α-SMA) expression is typically found in vascular and enteric muscle tissue, in myoepithelial cells, myofibroblasts and pericytes. However, expression of this actin isoform could be detected in a variety of other cells and tumors with a preexisting different phenotype. Our study focused on the expression of actin isoforms in some muscular and non-muscular tissues and tumors. The goals of our study were: 1 Analysis of actin isoforms expression in normal, osteoarthrotic, posttraumatic and transplanted cartilage. 2 Analysis of actin isoform expression in some non-muscle tumors. 3 Analysis of actin isoform expression in uterine leiomyomas after therapy and in leiomyomas with inclusion bodies of the gastrointestinal tract and uterus and the detailed analysis of inclusion bodies. Classification of marked cells migration in tissue

This diploma thesis deals with analysing of modern methods for cell detection, visualization and quantification in 3D space. The first section deals with optical methods for cells detection. There is detailed discussion about cell labeling and detection on confocal microscopy. There is also description about developed algorithm for whole cell volume quantification from microscopy images. This could made a comparsion of fluorescence signal according to time of cell labeling and according to cell shapes. There was also optimalization of handmade tissue phantoms visualization. It could be compared a possibilities of cell detections in these phantoms by confocal microscopy and OCT. It was also implemented algorithm for quantification of cells from OCT images. Besides confocal microscopy and OCT cells are also analyzed by other methods. The last part is the Conclusion of results and comparison of used methods.

The knowledge society and open economy: impacts on the competitiveness of the Czech republic in the global world

This diploma thesis is focused on the competitive advantage of the Czech republic in the global world of nowadays. First, it is important to explain the expression a "knowledge economy" or a "knowledge society". In my thesis, I will introduce the theories of human capital, knowledge economy, innovation, economic growth and divers sources of appropriate informations. After the introduction of these theories, I will follow up with the introduction of the Lisbon strategy and its analysis. Here, I will do a comparison of the EU, Czech Republic and the global economy. The reason of this thesis is to show a possible ways of development and economic growth of the Czech Republic. The Lisbon strategy reflects the complexity of this issue. Thus, the next chapters will be about the analysis of Finland's economic transformation and its current state. Following with the short introduction and analysis of the american and french education, science and research systém and shape. Thus, those acquired knowledge will serve me to make an outline of the possible solutions for the Czech Republic because our current situation enables us to inspire ourselves in the whole diversity and uniqueness of their economic and social systems and to apply their know-how in the broader context of the Czech republic.

Application of monosubstituted cyclodextrins in chiral separation of omeprazole by capillary electrophoresis

Chiral separation of omeprazole was the subject of this work. Five diferent monosubstituted derivatives of α- and β-cyclodextrins, two of them cationic and three anionics, were used as chiral selectors in various buffers of different pH. The influence of CD and buffer concentrations on chiral separation was studied. Voltage of 25 kV, UV detection in the range from 202 to 303 nm and constant temperature of 20 řC was used for all measurements. Concentration of CDs between 0 and 7 mmol.l-1 was used. Fused silica capillary of 50 µm ID and 375 µm OD, 48,5 cm total and 40 cm effective lenght was used as separation capillary. Dimethylformamide was used in stock solution of omeprazole as solvent, and was simultaneously applied as EOF marker. Succesfull chiral separation of omeprazole was achieved under the application of 2'-O-carboxymethyl-β-cyclodextrin (2'-CM-β-CD) and mono-6-deoxy-6-N,N,N',N',N'-pentamethylethylendiammonio- cyclomaltoheptaose dichloride (PEMEDA-β-CD) where the resolution of 1,42 (pH = 5,1) and 4,29 (pH = 11,1) was measured, respectively.

Analysis, characterization and separation of 2,6-diaminopimelic acid derivatives by capillary zone electrophoresis and micellar electrokinetic chromatography

In this work, capillary zone electrophoresis (CZE) and micellar electrokinetic chromatography (MEKC) with UV-photometric detection were used for the determination of degree of chemical purity, limit of detection (LOD) and limit of quantitation (LOQ) of N-acyl derivatives of 2,6-diaminopimelic acid (DAP). The characterization of DAP derivatives was performed by determination of their effective electroforetic mobilities in several background electrolytes in acidic and alkalic pH range. The mobilities were corrected to reference temperature 25 řC. Separation of the mixture of DAP derivatives were achieved by MEKC in acid background elektrolyte (500 mmol·dm-3 acetic acid, pH 2.54) using anionic surfactant 60 mmol·dm-3 sodium dodecylsulfate (SDS) as a constituent of the micellar pseudostationary phase. Application of Diversity Management in Selected Company

This diploma thesis is focused on the application of diversity management in selected technically oriented company. A theoretical part gives and overview of essential terms regarding given topic and general findings about areas of diversity and gender management and its implementation in corporate sphere. The practical part deals with analysis of internal environment of the company and a perception of diversity and gender balance by employees. The aim of this thesis is to evaluate a concrete application of diversity management in selected company and to prepare proposals of solutions of potential issues detected.

Amperometric detection of aminoderivatives of biphenyl and naphthalene at screen printed carbon electrodes

According to International Agency for Research on Cancer (IARC), some specific isomers of amino derivatives of polycyclic aromatic hydrocarbons (APAHs) are classified as carcinogenic to humans (Group 1). Several APAHs are employed in the manufacturing of widely-used azodyes, therefore, sensitive and selective analytical methods for their determinations are still needed. This study aims at the possibilities of detection of 2-aminobiphenyl (2-AB), 4-aminobiphenyl (4-AB), 1-aminonaphthalene (1-AN) and 2-aminonaphthalene (2-AN) using bare screen-printed carbon electrodes (SPCE). These were employed as working electrodes in amperometric detection flow cell for repeated use in liquid flow methods - reverse phase high performance liquid chromatography (RP-HPLC) and flow injection analysis. Sufficient stability and repeatability was obtained for mobile phases containing the maximum of 70% of acetonitrile. Limits of detection for 4-AB given by FIA in the aqueous solution of 0,01mol.l-1 phosphate buffer pH 3,0; or in the solution of acetonitrile and 0,01mol.l-1 phosphate buffer pH 3,0 (40:60) (V/V) were 1,0 mol·l-1 and 2,0 mol·l-1 , respectively. The above thesis topic I chose for two reasons. The first reason was that I had as a secondary specialization in Business Valuation chosen course of study in this specialization. I met with the subject in Practical Business Valuation. In this course, I participated in valuation process Consumer Unity in Jindrichuv Hradec. Because at my residence located Consumer Unity in Nymburk, whose stores I visit from time to time, I wonder if it is appropriate to evaluate the company using the income approach (or under what conditions) and what is the value of the company. Therefore, the objective of this thesis I have provided to determine the market value of unity, consumer cooperative in Nymburk. The second reason why I chose the topic of "valuation" as a whole is the fact that my favorite literary genres include detective novels. With a little imagination it can be said that the search for business value is similar or even identical to the above literary genre. When first we see business, we know about him very little, but we suspect that it certainly has some value. In the case of murder, we see the body, but do not know the offender. Over time composing pieces together and connect us with each connection, and finally ideally we will see a clear picture and find out who is the perpetrator, even if the culprit was the victim. If we are looking for enterprise value, this is a similar case. Gradually, as we proceed through financial, strategic analysis, the value of generators, the financial plan to the final valuation, we find that the company is healthy, if it is a competitive, what is the prognosis for the future, when we are at the end of the quest unable to determine its value. Proper "thriller" should consist of five components. From the introduction plot, climax, and denouement of the conclusion. Therefore, the thesis is divided into five parts. The first part consists of basic information about the company, where he briefly introduce the valuated company. Followed by a financial analysis that tells us what threats or vice versa opportunities deriving from the financial statements of the company and answer the question of whether the company is financially able to continue to operate. Another part of the work is strategic analysis to assess the position on a particular market and answers the question of how they will develop sales and market share of the business in the future. Atlantes and caryatids in Prague

Diploma work "Atlantes and Caryatids in Prague" deals with tradition, transition, specific development a shape varieties of elements taken from antique architecture - atlantes and caryatids. For deep comprehension of the whole development, first it's necessary show and understand origins and significance of these elements in antiquity itself. It's also shown a first age of copying and imitating of old models. Original caryatids were dancers of Artemis cult in a village called Karyai (Peloponnésos). Placing human figures in architecture is of older date than story of treachery of Karyai's citizens (Vitruvius), from which is derived placing them under an entablature as an expression of a punishment (in place of columns). Further, both stories are merging. E.g. in case of Erechtheionkorai we can speak of placing them for honor. Mothological origins of Atlant is well-known and placing him into architecture is logical. In hellenistic times were made o lot of reminiscences of old statues (copies or rather mixing of different sources) which can be considered as a first large wave of historism in development of figural supports. Insect repellent and feeding deterrent activity of natural sesquiterpene lactones and their derivatives

Thirty compounds were investigated in order to detect their repellent and antifeedant activity. The substances were tested on several insect test-objects: Sitophilus granarius, Tribolium confusum and Trogoderma granarium. Five sesquiterpene alpha-exometylene-gamma-lactones and one structurally transformed dimethylamino derivative showed high antifeedant and repellent activities.
