Evaluation of applicability of indirect photometric detection for chiral separations of selected carboxylic acids in dairy products

In this diploma work is check possibility application indirect photometric detection for enantiomers separation lactic acids and beta-hydroxybutyric acid with capillary electrophoresis. As chiral selector is used vankomycin. Verifyed his ability partial separation lactic acid on antipode with using picric acid as vizualition anion in background electrolyte. Used was coated capillary. Application indirect photometric detection succeed. Chiral separation beta-hydroxybutyric acid wasn't successful. Hip roll by selection fit vizualization anions for purposes indirect photometric detection. Are measured their electrophoretic and spectrophotometric characteristics, for inquest compatibility use in capillary electrophoresis in relation to indirect photometric detection. From timed datums were to be you - nap optimal vizualizatin anions for others experimentation. According to ascertained and measured feature reflect be optimal eosine soluble in alcohol and bromophenol blue.
Possibilities of reprocessing of the glycol and glycerol waste into suitable materials

At the beginning of the thesis is summarized biodiesel production and its world production. There is more developed biodiesel production in the Czech Republic and in the European Union and some European legal regulations and standards for biodiesel. Also described herein is a composition of biodiesel and the description of the technology of its production. In the theoretical section summarizes the physical and chemical properties of glycerol and there is also summarized the traditional use of glycerol as food, explosives, etc. There are also new procedures that were performed in the experimental part of the thesis. Then, here are the proposed new procedures for processing waste glycerol and glycol, which were conducted in the experimental part of the thesis. There are described the ion exchange properties of the cation exchangers and also properties of salts generated by neutralization of NaOH in the waste glycerol with organic acids and CO2. There is also described wood protection, properties of ethylene glycol as coolant and properties of boroglycerols and boroglycols as protection for wood and mechanism of their preparation. Then there is described a mechanism of dehydration of glycerol to acrolein and options which can perform the synthesis. In the experimental part, the pH of waste glycerol was measured and then was measured the amount of NaOH in the waste glycerol. There is also desribed cleaning of waste glycerol from NaOH using acidic cation exchange resins on columns, or by neutralization with oleic acid, lactic acid and CO2. Further syntheses were performed glycerol borate and ethylene glycol borate. Water amount in ethylene glycol and methanol with water amount in glycerol were found by distillation. Then were prepared boroglycol and boroglycerol. By the amount resulting from the amount of reaction water were observed reaction conditions H3BO3, Na2B4O7•10 H2O and glycerol. At the end was carried out experimental dilution resulting boroglycerol and boroglycol by ethanol, methanol, TMB. Finally, the diluted boroglycerol was deposited on wood. Dehydration of glycerol to acrolein under the catalytic action of KHSO4 was performed. The results are given all the results and discussed the possibility of using the knowledge gained in this work in the industry. In conclusion, there are listed the economic comparison using waste glycerol and crude oil as a raw material and also charts the development of oil prices in 40 years since 1970, and graph of the price of waste glycerol. There is also discussed technological applicability of the methods used in practice.

The thesis entitled „Quality parameters of mayonnaise with additives“ aimed to perform model production of mayonnaise samples according to different recipes (combination of additives – lactic acid, citric acid, sodium sulphate) with different levels of additives and to determine the basic quality characteristic (pH, dry matter content, titratable acidity content) and sensory properties. The pH of model samples ranged from 3.52 (sample with citric acid and lactic acid combination) to 4.03 (sample with lactic acid). The dry matter content ranged from 53.59 % (sample with sodium sulphate and lactic acid) to 59.86 % (sample with citric acid). The titratable acidity content of the model samples ranged from 42 mmol·l−1 (sample with lactic acid) to 56 mmol·l−1 (sample with citric acid). In the sensory analysis, the sample with combination of citric acid and lactic acid got the best results. The results obtained confirm that the additives in the mayonnaise did not have a negative effect on the quality and sensory characteristics.
Electrochemical biosensors with spatially separated enzymatic and detection parts for selective analysis in flow-through arrangement

This dissertation thesis presents the newly developed four highly reusable, stable as well as simple, and cost-effective electrochemical (bi)enzymatic biosensors for the selective and reliable determination of choline, acetylcholine, uric acid, and L-lactic acid in flow injection analysis. All biosensors are based on the concept of the spatial separation of the biorecognition part from detection one and amperometric monitoring of the enzymatically consumed oxygen via its four-electron reduction at the highly negative detection potential. In this way, the design of the biosensors includes an easily replaceable enzymatic mini-reactor(s) connected upstream to the flow cell that contains the appropriate silver amalgam-based transducer. The enzymatic mini-reactor based on choline oxidase, uricase, or lactate oxidase was used for choline, uric acid, or L-lactic acid biosensors, respectively. The acetylcholine bienzymatic biosensor includes the consequently connected choline oxidase- and acetylcholinesterase-based mini-reactors. Active preparatives based on koji fermentation and medical mushrooms extracts

This bachelor's thesis deals with the optimization of the growth of the rare species fungus of the hedgehog mushroom on cereal substrates for the purpose of the subsequent to cultivation of lactic acid bacteria. The theoretical part is focused on the description of the rare fungus species and the use of the cereals as substrate and the characterization of their active substances. It also deals with lactic fermentation and its use. In the experimental part, the preparation of the cereal substrate for the cultivation of the hedgehog mushroom was optimized. The method of inoculation of the cereals substrate with liquid mycelium in combination with plate mycelium was chosen as the most applicable. In this combination, the substrate does not dry out and grows evenly from both sides of the substrate. The composition of the cereal substrate with grown mycelium was characterized (antioxidant activity, carbohydrates, proteins). Finally, the substrate was inoculated with lactic bacteria. The lactic bacteria added nutritional value to the product. Fermentation products (antioxidant activity, carbohydrates, proteins) were characterized, and bacterial viability was analysed.
Production and application of antimicrobial substances produced by lactic acid bacteria

The bachelor’s thesis deals with the production of antimicrobial substances by selected lactic acid bacteria of the genera Lactobacillus, Pediococcus and Bifidobacterium, in which the production of bacteriocins was assumed based on the conducted research. Bacteriocins are peptide-based antimicrobial substances with the potential to be widely used primarily as biopreservatives in the food industry. The theoretical part of the bachelor’s thesis focuses on antimicrobial substances produced by probiotic bacteria with a further concentration on bacteriocins produced by selected bacterial species and their current research and application in the industry. In the experimental part, the antimicrobial activity of prepared isolates from probiotic bacteria was tested on selected gram-negative and gram-positive bacteria. Specifically, this was done by the agar diffusion method, the broth dilution method and the cell viability test using resazurin. The amount of lactic acid and acetic acid in the individual isolates was determined using high performance reverse-phase liquid chromatography, which made it possible to further discuss their influence on the antimicrobial activity of the isolates. Strong antimicrobial activity against indicator bacteria was found in all isolates. For some isolates, it was higher than the antimicrobial activity of lactic acid itself together with its concentration which is found in the isolate according to the measurements. That is a significant stimulus for further research of these probiotic isolates, especially the L. acidophilus and L. plantarum isolates. For example, it was found that the tested isolate from L. acidophilus showed an inhibitory activity against S. epidermidis of 90,76 % and 81,62 % against E. coli at a concentration of 20,47 mg/ml. The isolate from L. acidophilus was subsequently applied to plant packaging with the potential of usage in the food industry, and the inhibitory activity of this isolate against contaminating microorganisms was observed.
Quality characteristics of beef during storage

The topic of this work is to verify the effect of different treatments of beef after slaughter on quality characteristics, aerobic plate count on the surface of meat during maturation and sensory properties of matured meat before and after heat treatment. The literature review describes the quality characteristics of meat, the effects on quality, the maturing process and the methods of treatment of meat after slaughter. The practical part of the work consisted of treating meat after slaughter with lactic acid, lactic acid + onion peel extract and pure onion peel extract. The next part consisted of measuring the pH and the aerobic plate count during the dry maturation of beef quarters and after the wet maturation in the vacuum bags, determine the composition of the meat, the dripping of the water, the color and the sensory evaluation of the meat. The obtained values were statistically evaluated.
Effect of molecular structure on liquid crystalline behaviour for chiral lactic acid derivatives

Liquid crystals are self-organizing materials that have great potential for practical applications. The relationship <molecular structure - mesomorphic properties> has not been fully established and understood despite numerous studies. We discuss a relatively broad subclass of calamitic chiral lactic acid derivatives and provide specific examples of effective tuning of their mesomorphic and electro-optical behaviour using various types of lateral substitution placed on the molecular core.
Preparation and characterization of unsaturated bio-polyester resins

The theoretical part of this work is devoted to the selection of suitable components for synthesis of a branched oligoester and their structural arrangement. Further, oligomers of lactic acid esterified with crotonic acid branched by erythritol or pentaerythritol are discussed. Crosslinking of prepared oligoesters is accomplished by free radical mechanism using vinyl acetate as reactive diluent and 2,2'-azo-bis-isobutyronitrile as initiator. Resin was cured by vinyl acetate in different mass ratios. Results show that there in an optimal ratio for curing between oligoester and reactive diluent. Using less vinyl acetate results in insufficient crosslink density or, on the contrary, using considerable excess of vinyl acetate causes rapid polymerization, that also results in lower density of crosslinking. The crosslink density was determined indirectly by extraction of the soluble part to acetone after curing and drying of the product.
UTILIZATION OF FOOD PROCESSING WASTE FOR LACTIC ACID AND ETHANOL PRODUCTION

The doctoral thesis is focused on the microbial production of lactic acid and ethanol using food processing waste as substrate. Coffee processing waste (spent coffee grounds), wine production waste (grape pomace) and orange processing waste (orange peel) were chosen as substrates for experiments. The theoretical part is dedicated to summarizing current knowledge about waste from food production and possibilities of its processing. It also deals with selected metabolites (lactic acid, ethanol) to which these wastes can be used. Part of the experiments was focused on the characterization and optimization of hydrolysis to maximize the amount of fermentable saccharides. Different combinations of chemical, physical and enzymatic hydrolysis of selected substrates have been tested. Subsequently, a suitable strain for lactic acid and ethanol production was searched for. In the case of lactic acid production, 7 bacterial strains were selected (Lactobacillus casei CCM 4798, Bacillus coagulans CCM 2013, Bacillus coagulans CCM 2658, Lactobacillus rhamnosus CCM 1825T, Lactobacillus delbruckii subsp. bulgaricus CCM 7190, Lactobacillus plantarum CCM 7039T, Streptococcus thermophilus CCM 4757). These strains were first cultivated on the synthetic media containing different kind of saccharides. Afterward, the cultivation on the waste biomass hydrolysates were tested. In the case of ethanol production, 2 yeast strains kmeny (S. cerevisiae CNCTC 6646 a S. cerevisiae CNCTC 6651) were cultivated on hydrolysates of individual waste substrates. Subsequently, the experiments focused on the production of lactic acid and ethanol on hydrolysates of waste biomass in bioreactor were done. The last part of this doctoral thesis deals with the microaerobic pretreatment of lignocellulosic biomass to increase the production of organic acids during the acetogenic phase of anaerobic digestion.
