Modeling the movement of dangerous gases above the central part of Pardubice town
Dittrt, František ; Brych, Karel ; Jaňour, Zbyněk
The study was focused on the transport of the dangerous gases above the urban area of Pardubice town. The gaseous dispersion was caused after an accident with an outflow of chlorine in the chemical factory. Diffusion fields were simulated using a model of landscape surrounds the chemical plant at scale 1:3000. The results indicate fast decreasing of the surface concentrations due the intensive turbulent mixing inside the urban surface. Measurements were engaged in the tunnel by Infrared Analyzer IREX and results were presented as map layers in GIS.

Dynamika impaktních proudů zkoumaná v příčných řezech proudem
Tesař, Václav ; Něnička, Václav
Authors investigated experimentally air jets impinging on a flat plate. The object of interest were vortical instability structures in the mixing layer of the jet. Their formation was triggered by azimuthal waves generated in the nozzle exit. The structures were visualised by planar laser-light sheet illuminating a jet cross section where it scattered on by water mist particles added into air. The cross section was, photographed by a high-frequency camera located behind transparent impingement plate.

Land cover changes in Bohemian Switzerland using remote sensing data
Lihanová, Kristýna ; Štych, Přemysl (advisor) ; Součková, Jana (referee)
Land cover changes in Bohemian Switzerland using remote sensing data Abstract The purpose of this project was to classify land cover in The National Park Bohemian Schwitzerland from LAMDSAT data (1999, 2002, 2009), FORMOSAT data (2006) and QuickBird data (2009). Data used in this paper are described in details. The method used for images classification was method of unsupervised classification in Geomatica program. Created classes were merged into the final five classes. An Assessment of accuracy of classification showed that results don't reflect reality. Mixed forests are the biggest problem because they can be mistaken for coniferous or deciduous forests. Classification results were smoothed low pass filer and converted into a vector formats in ArcGIS program. For the years 2006 and 2009 was counted the vegetation indexes NDVI and LAI. Results showed that the densest vegetation is in deciduous forests. Changes in land cover were assessed in two ways. Shares of each class were calculated for all years. Overlays layers are created in ArcGIS. Overlap was conducted between 1999 and 2002 and between 1999 and 2009. Most changes were found between the coniferous and mixed forests. These changes are caused by the inaccuracies in classification. Key words: land cover, Bohemian Switzerland, forest, Landsat,...

Mixed-use building in Bruntál
Ondrušková, Veronika ; Šteffek, Libor (referee) ; Brzoň, Roman (advisor)
This diploma thesis deals with design documentation of New mixed-use building in Bruntál. The building is located in the centre of the Bruntál on the plot number 83/1. Regulatory plan defines this plot for construction. It is marked as SC – mixed-use residental plot. The building contains the main conditions of use – living, shopping, restaurant and administration. The building has 3 above-ground floors and 1 basement. At ground floor there is café and shop. At first floor there are offices and conference room. At second floor there are flats. In basement there is utility room, cellars, garage and storages. The café and shop is separated and has its own entrances. Horizontal and vertical load-bearing and non-load bearing structures are bulit of permanent formwork VELOX – perimeter load-bearing walls are sandwiches, internal load-bearing walls are of cast-in-place reinforced concrete, which is covered by permanent formwork. Non-load bearing walls are bulit of 2 boards of permanent formwork with inserted acoustic insulation. Vertical load-bearing structures are made of cast-in-place reinforced concrete, which is covered by permanent formwork. The building has concrete foundation strips and cast-in-place reinforced concrete stairs. The roof has got classical order of layers.

Transfection of stem cells and other testicular cells of Xenopus tropicalis
Šídová, Monika ; Tlapáková, Tereza (advisor) ; Petr, Jaroslav (referee)
Xenopus tropicalis represents one of the most important model organism used in developmental and cellular biology. Laboratory of Developmental Biology at the Faculty of Science, Charles University in Prague has successfully established a long-term culture of X. tropicalis juvenile testicular cells. Based on expression profiling analysis of selected specific markers (Sox9, WT1, etc) it was proven that the major cell type in this culture is pre-Sertoli cells. Furthermore these pre-Sertoli cells allow a longterm cultivation of the germinal stem cells. By performing a histochemical test for the presence of alkaline phosphatase in the colony of these cells were proven the features of stem cells. In this diploma thesis we focused on optimization of work with the mixed cell culture. In particular we define conditions of dissociation and subsequent separation of a feeder-layer formed by the pre-Sertoli cells. We also attempted to develop suitable conditions for transfection of the germinal cells. With these techniques we will to investigate the functional properties of the germinal stem cells. Moreover, it provides us a powerful tool for performing another experiments focused on transgenesis and/or different gene inactivation. Powered by TCPDF (www.tcpdf.org)

Linzerová, Petra.: Biological Effect of Plant Metabolites III. Izolation of Alkaloids from Eschscholzia californica Cham. A Diploma Work, Charles University in Praque, Faculty Of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany And Ecology, Hradec Králové 2009, 54 pages Aims of this diploma thesis were to isolate alkaloids from J1 extract obtained from a herb with roots from a plant Eschscholzia californica Cham., to perform the identification of gained alkaloid and to participate in determination of activity of isolated alkaloid against the erythrocytic acetylcholinesterase. After finding suitable mix of reagents we divided the extract on a chromatografic column filled with silica gel by system of chloroform : ethanol with gradual increasing of concentration ethanol. With obtained eluents we made a orientation thin layer chromatography with developing system acetic acid : ethanol : water (60 : 30 : 10). Alkaloids were detected by UV radiation and spray with Dragendorff reagent. After decantation of fractions with similar spectrum of matters we cleared selected mixture. We obtained two alkaloids; NMR spectroscopy will be used to determine the structures of these alkaloids. Ševčíková Jonáková, Jana ; Tlapáková, Tereza (advisor) ; Drda Morávková, Alena (referee)
5 Abstract Xenopus tropicalis is an important model organism used in developmental and cell biology. Long term primary mixed cell culture, derived from juvenile testis of Xenopus tropicalis, is the first tissue culture of this specie. Characterization of this tissue culture is important for its further handling and subsequent experiments. The culture is composed of Sertoli cell precursors. Feeder layer of this cells enables long term germinal stem cell cultivation. Influence of condicioned medium plays important role during the cell culture cultivation. Result of this task approved effect of condicioned medium for cell culture growth. Cell culture and testis gene expression analyse revealed, that in process of Sertoli cell differentiation many growth and differentiation factors are involved. Presence of marker genes has been approved by RT-PCR and in situ hybridization. This in vitro system can contribute to clarification of different roles of pre-Sertoli and Sertoli cells in testis differentiation, and therefore it establishes a new spermatogenesis model offering the possibility to distinguish parts of the pre-Sertoli and Sertoli cells differentiation, which is difficult in mammal models.
