Structure and properties of bacterial biofilms

This thesis deals with the study of basic properties of cells of bacterial biofilm produced by PHA producing bacteria, specifically Burkholderia cepacia and Burkholderia sacchari. Two cultivation approaches were used to compare different types of cells: at first, the cells were cultivated in bioreactor providing both planktonic and biofilm attached cells, secondly, cells grown on Petri dishes were also included in the study. In experimental part the affinity of each type of cells to hydrophobic phase was studied using BATH method. It was found that sessile cells, either biofilm associated cells as well as cells taken from Petri dishes, prefer hydrophobic phases more than planktonic cells. Also size and Zeta potential were measured using Dynamic Light Scattering (DLS). It was found for studied samples that, cells in these samples tend to aggregate and the difference of size between both types of cells occurs only for biofilm of the strain Burkholderia cepacia. Finally the impact of presence various bacterial cells types on surface tension of water as well as amount of PHA cumulated in different types of cells were observed.

The main objective of bachelor thesis was evaluation of quality of spa and wellness services in spa city Karlovy Vary. The practical part of thesis focuses on evaluation of data acquired by field survey which were shown in graphs. Practical point of view of business companies is represented by guided interview with manager of tour operator Čedok and sales and marketing deputy of Alžbětiny lázně. Bachelor thesis also contains new package for clients in retirement age.

This thesis deals with a design of a circular draining machine. The aim is to design a machine that complies with the respective requirements. The machine is assembled in a line for the preparation of paper stock, which is further processed for final paper production.
Phosphate conversion coatings

The main objective of the master’s thesis is to study the influence of phosphate coating structure on the mechanical properties of screw connection. The theoretical part describes the conventional phosphating process and it is followed by an analysis of current knowledge of phosphating technology. Experiments are carried out on screw valves mounted to a high pressure diesel pump by Bosch Diesel s.r.o., made of unalloyed carbon steel. To investigate the effect of the structure on the connection sealing, twelve kinds of phosphate coatings varying in one process parameter were prepared. The monitored parameters determining the phosphate structure are – type of phosphate and activation bath, its temperature and concentration and time of immersion. The sealing connection is examined during simulated assembly process by angle controlled tightening while measuring torque and permanent material deformation. Prepared layers are characterized by determination of weight per unit area and their structure observed by electron microscope.

Yverdon-les-Bains Thermal Spa is a group of elements searching for their identity since the end of the 20th century. The spa complex consists od several buildings built in different eras. Each one of the has its own strong formal identity whether expressed by facades, materials, volumes or spaces. All the historic buildings are to be preserved and new spa is to be designed to complement them. Potential of the new spa development is one of the most important values of the diploma project.

Designed construction of a spa with the wellness center programs (relaxation type), for 200-250 people (at one moment) at Brno city, near Voronez Hotel and Exhibition Centre. The five-floor spa building is designed as a puzzle in the shape originating from a dice. Cycles of water in its various forms and states of matter or properties fade into one another here. Each floor is designed to have its own theme (water world, the Asian world, aroma floor, salt floor, sunny and airy bath) and offers various types of premises and procedures: swimming, relaxing in the water, baths, showers, steam rooms, saunas, infrared saunas, wraps and massages etc. The spa program is accompanied by various types of refreshments: cafe, bar, tearoom, fruit bar. Rooftop pool and bar offers a unique view on the city. The spa and hotel Voronež parking is carried out within the ground floor.
Using Persteril\copyright{} in practice for the prevention of fungal infections of eggs and fry baths sturgeon compared to commonly used products

Using Persteril36 (Acidum peraceticum) in practice for the prevention of fungal infections of eggs and fry baths sturgeon in comparison with the used products deals with the disinfection efficiency. In the experimental part were used three concentrations Persteril36, whose active ingredient is peracetic acid (36%), and the other were evaluated ToruMin, Acriflavin, TeraP and AquahumTM. Persteril36 in high concentrations appear to be very effective biocide, that leaves no residue and is not harmful to the environment. Other disinfectants have been evaluate detail in the results. Some of them appear to be very effective and they results exceeded expectations.
