Interaction of proteins Clock and Cycle, Preparation of constructs for subsequent expression in S2 cells

The aim of this thesis was to create constructs containing actin or Hsp70 promoter, complete coding DNA sequence of Clock and cycle genes from the linden bug (Pyrrhocoris apterus) and tag, green or red fluorescent protein. Such prepared constructs were designed to be used to study interactions of Clock and Cycle proteins in vivo in Schneider S2 cell line. In addition, effectiveness, reliability and applicability of the used method Gateway cloning was examined.
Clock genes in mammalian circadian pacemaker

The behavior of mammals and their physiological processes are dependent on the daily rhythms. These rhythms are controlled by an endogenous circadian clock that responds to light/dark cycles of environment. In mammals, the circadian clocks, are arranged hierarchically, and work in almost all cells and tissues. Suprachiasmatic nuclei (SCN) in the hypothalamus are at the top of the hierarchy and work as a major circadian pacemaker. This work presents the clock genes that have been discovered in the last twenty years in the SCN of mammals. The first chapter focuses on the general mechanism of circadian rhythms and structure of the SCN. The second chapter describes the transcriptional-translational feedback loops that are an essential part of the proper function of the circadian clock, and function of individual genes is briefly introduced in this chapter. The following chapters already include a chronological characterization of Clock, Bmal1, Period and Cryptochrome genes, as they were discovered. If the clock gene expresses different function from the clockwork mechanism, it is briefly introduced in the final paragraph on the chapter. Circadian clock genes in insects

This thesis focuses on molecular characterization of circadian clock genes in insects. It explores genetic diversity of circadian clock genes by molecular characterization of several insect species including two dipteran flies (Musca domestica and Drosophila melanogaster), two cockroach species representing ancestral insects, and the linden bug, Pyrrhocoris apterus. Furthermore it considers various roles of circadian clock genes in insect physiology. Application of molecular-biology methods in Pyrrhocoris apterus, non-model insect species, enable us to investigate involvement of circadian clock genes in photoperiod induced physiological responses. Application of molecular-biology methods in Periplaneta americana and Blattella germanica was used to explore involvement of circadian clock genes in magnetoreception.

Bachelor thesis entitled "Microcontroller-controlledtimer" is focused on design and practical application created a device that will be capaje of depending on external dating of simple tasks. This paperdescribes the features and capabilities of the microcontroller ATmega162, the Operations and program services. There is also described several other components used such as RTC, LCD display, matrix keyboard, temperature and othersensors. The first part is devoted to the oretical introduction to microcontroller and used parts. In another part ofthe PCB design - suitable layout of components, connectors, location, solving problems in PCB design. Fortesting functions designed and manufactured equipment is made simple test PCB. The practical part ended custom programming microcontroller programming language C.
Modernization of the master clock for slave clocks controlling

This diploma thesis describes modernization of the master clock HN 50 created by company ELEKON, s.r.o. Master clock can be used for controlling of analog or digital slave clock or for switching school bells. Exact differences between old and new version of the clock are described and ways of synchronization and controlling of slave clock are explained. Main goal of modernization is applying the SMT technology for automation of production. Another problems and shortcomings of previous version are developed, such as new power supply structure, selection of a new microcontroller and usage of several different displays. Every problem is developed and new solutions are chosen. Last part of the document describes the software which is clarified on several flowcharts. Outputs of the thesis are complete schematics, printed circuit boards and 3D models of the modernized device.

This bachelor thesis describes the development of mobile application for Windows Phone 7 focused on sleep and awakening the user. The application is very easy to use and uses built-in modules of advanced mobile phones, such as high resolution camera and the accelerometer. This thesis describes the main features of Windows Phone 7 platform, analysis of competing solutions, application concept and its implementation. Furthermore, this work describes the solution of various problems in the development and also provides examples of methods and tools. Finally, there is also a list of used libraries.

The purpose of this thesis is to describe the operating system Contiki for embedded systems, NTP time synchronisation protocol and to design and implement an NTP client for the Contiki operating system.
Experimental Evaluation of Clock Synchronization for Networked Control Systems

Within this topic, a testing environment is built, where different clock synchronization methods are evaluated in order to establish a maximal achievable clock precision in a distributed SCADA system based on the QNX operating system. Furthermore a measurement of parameters of communication for different technologies and circumstances is taken. For this purpose, porting of the PTP protocol to QNX OS is necessary.
