Chování savců a jejich fyziologické procesy jsou závislé na denních rytmech. Ty jsou řízeny endogenními cirkadiánními hodinami, které reagují na cykly světla a tmy prostředí. U savců jsou cirkadiánní hodiny uspořádány hierarchicky a pracují téměř ve všech buňkách a tkáních. Suprachiasmatické jádra (SCN) v hypotalamu jsou na vrcholu této hierarchie a pracují jako hlavní cirkadiánní pacemaker. Tato práce představuje jednotlivé geny, které byly za posledních dvacet let v SCN savců objeveny. První kapitola se zaměřuje na obecný mechanismus cirkadiánních rytmů a strukturu SCN. V druhé kapitole jsou popsány transkripčně-translační zpětnovazebné smyčky, které jsou nezbytnou součástí správné funkce cirkadiánních hodin, a stručně je v této kapitole představena funkce jednotlivých genů. Následující kapitoly patří již chronologické charakterizaci Clock, Bmal1, Period a Cryptochrom genů, tak jak byly objeveny. Pokud je známá i jiná funkce hodinových genů než v molekulárním mechanismu cirkadiánních oscilací, je uvedena v závěrečném odstavci každé kapitoly. Závěrečné kapitoly se zabývají dalšími molekulami, kasein kinázami, ROR a REV- ERB receptory, které svojí funkcí ovlivňují expresi a degradaci jednotlivých cirkadiánních genů v SCN. These rhythms are controlled by an endogenous circadian clock that responds to light/dark cycles of environment. In mammals, the circadian clocks, are arranged hierarchically, and work in almost all cells and tissues. Suprachiasmatic nuclei (SCN) in the hypothalamus are at the top of the hierarchy and work as a major circadian pacemaker. This work presents the clock genes that have been discovered in the last twenty years in the SCN of mammals. The first chapter focuses on the general mechanism of circadian rhythms and structure of the SCN. The second chapter describes the transcriptional-translational feedback loops that are an essential part of the proper function of the circadian clock, and function of individual genes is briefly introduced in this chapter. The following chapters already include a chronological characterization of Clock, Bmal1, Period and Cryptochrome genes, as they were discovered. If the clock gene expresses different function from the clockwork mechanism, it is briefly introduced in the final paragraph on the chapter.
