The development of Leishmania in the insect vector of the genus Phlebotomus

The work was focused on the study of various aspects of development of Leishmania in vectors of genus Phlebotomus and can be divided into two main parts. In the first part, we studied the competition of Leishmania major and L. turanica during their development in the sand flies who are natural vectors of L. major using the experimental co-infections of fluorescently marked promastigots of these two species. While both leishmania species developed similar in the intestine of Phlebotomus papatasi, L. turanica prevailed in P. duboscqi in the late stages of infection. The fluorescent marking of Leishmania should allow us also to study possible genetic exchange between species at different stages of Leishmania infection (2nd, 9th and 14th day after the infective feeding). Using the flow cytometry (FACS) we have repeatedly identified dozens of objects emitting red and green signals in the intestinal homogenates of co-infected sand flies, however further analysis with confocal microscope disproved these objects as the hybrid promastigots of Leishmania. Freedom of Expression and Censorship on the Internet and Social Networks

This thesis is focused on freedom of expression and censorship on the internet and social networks. Its goal is to evaluate current situation of censorship of the internet, to describe the most often used methods of censorship and suppressing free speech, as well as methods of its circumvention. The thesis is focused on the progress of internet censorship in its shift from a tool of governmental domestic network control into a tool in the hands of multinational organizations. Some focus is also given to the importance of anonymity as a way to freedom of speech, and to the analysis of current situation and possible future outcomes. This thesis is divided into five chapters. The first one serves as an introduction into the problematics. Second chapter describes methods of internet censorship, the third one describes its circumvention. The fourth chapter looks into the current situation regarding the role of social media in terms of internet censorship and possible results of censorship as well as future evolvement. Fifth and last chapter is dedicated to research of anonymous parts of internet community that could otherwise face censorship.

Strategy of Corporate Social Responsibility in Hilton Worlwide company and its application in local conditions

This bachelor thesis focuses on strategy of corporate social responsibility (CSR) in company Hilton Worldwide. The bachelor thesis is divided into two parts. The theoretical part deals with concept of corporate social responsibility, with the philosophy of CSR, its history and the present. This part also defines key words related to corporate social responsibility like CSR, stakeholders, corporate philanthropy, sustainable development, business ethics and corporate citizenship. It also focuses on the important role of communication in general and also in CSR. The practical part introduces the concept and strategy of CSR in the company Hilton Worldwide. This part also presents hotels Hilton Prague and Hilton Prague Old Town and their activities within the CSR. SWOT analysis as strategy tool was used to evaluate the concept and communication of CSR in both hotels. The final results of analyses showed problems in internal and external communication. Based on results of SWOT analysis are suggested solutions and recommendations that should improve understanding and perception of CSR concept in both hotels, improve internal communication for employees, and use all modern communication tools and channels for communicating information about CSR activities. These measures will lead to increased loyalty and strengthening the company's competitiveness.

Analysis, design and implementation of CRM solutions for selected company

Bachelor thesis focuses on the area of information systems, which are used to support customer relationship management in companies. The main objective of this work is to provide a sample of solution for CRM system implementation for real customer from the banking sector. The first part defines basic functional areas of any CRM application. Each of these areas is described in more detail in terms of its primary use and tasks that within the CRM as a whole performs. Further is described the analytical area of CRM application (or Customer Intelligence), which is gradually playing a key role in the entire CRM system in past few years. It includes its potential uses in various fields. At the end of the theoretical part, the author compares the fundamental differences between cloud model of deployment and traditional on-premise solution. The practical part focuses on the main objective of the thesis and description of the supplied solution in the form of a module, which extends the existing CRM application with the option for business partners registration of Hypoteční banka, a. s. and ČSOB Pojišťovna, centralized in one system. The requirement is build in Microsoft Dynamics CRM 2011 and as the source system it is used CRM system of Hypoteční banka, a. s..

The civil elites at the local level

The main objective of this thesis is the identification and understanding of the mechanisms of recruiting local political elites and their role in the process of democratization of local politics, and in the process of strengthening local civil society on concrete example selected municipality. The case study concerns with the municipality Semily, which was chosen to work as a community with exemplary transparent and open manner of governance. In 2006 there was a significant change in political representation when recruiting new political elites was a top down process led by a particular person according to predetermined priorities.

Methods and ways of implementation of changes and innovations in companies through project management are in today's society very well established. There exists methodologies, techniques and tools for the management of individual projects. However in a role of project manager, I very often faced the fact that companies are performing the project portfolio management in very intuitive way. This in itself leads to failed and prematurely terminated projects, initiation of the wrong projects or realisation of correct projects, but at the wrong time. Very often I have also recognized, that there is lack of awareness of fact, that there are also Open Source applications that can help with the organization's portfolio. This thesis deals with the definition of requirements, search and selection of proper OSS application and subsequent implementation. In the first part of this thesis I prepare a theoretical framework about PPM, and on that basis then identify and verify a set of requirements for selection of right OSS PPM application. The next section of thesis describes searching for suitable applications from a variety of sources, assessment against the requirements and the final selection. In the last part the implementation project is drafted, which aims to serve other colleagues in project management as one of the possible implementation paths. The outputs are then continuously confronted with experts in matter of projects and project management in the field of IT in order to maximize symbiosis between theory and real life experiences. Virtually every project management methodology defines the collection of already-proven techniques, best practices or lessons learned from the previous similar implementations. The main contribution of this thesis I see in fact that it contains not only the design of the project implementation, but also describes a logical path, what leads to the result. Therefore this thesis could be used as a base or discussed best practice, when a project of implementation of PPM application in place.

Supported Emplayment from the Perspective of Emplayers

The philosophy of supported employment in the Czech Republic has been developing for over than twenty years. Nevertheless this service is still neglected and only a few non-profit organizations put it into practice. The service is not legislatively based and also the society does not have a clear opinion on employment of people with disabilities. Current labour market situation is not advantageous at all. Therefore some support of disabled people looking for a job is becoming more necessary and required. The employers are an important part of the whole problems, because their attitude to the idea of supported employment plays a significant role. The bachelor's thesis "Supported Employment from the Perspective of Employers" has pretentions to look into supported employment from the employers' point of view. They are not active initiators of the service, but they are important participants for its successful realization. The theoretical part of the thesis tries to describe the basic problems. It covers the service of supported employment from historical development to its importance. It focuses on special characteristics of disabilities and their social impact on human beings. The closing part is dedicated to working opportunities for people with disabilities in the circumstances of current legislative and economic situation. The practical part is written in the form of a qualitative research. The method of questioning and semi-structured interviews were used for the purposes of the research. Eight people were chosen intentionally for the survey. The data gained from the interviews were analysed with the method of open coding. The main aim of the thesis was to map out empoyers' experience with the service of supported employment. Motivation of employers to put this service into practice was the next part of the research. The question was: "What reasons for realization of supported employment do predominate by employers?" The research showed the most frequent facts, which predestine the integration possibility of disabled people in open labour market fundamentally. After the interviews and answering the questions of the research, it was possible to describe the employers' experience with the supported employment service and also their main incentives for its realization. It was found out that employers' experience with the supported employment service is quite limited. Employers often get to know it through the agency for supported employment, which is not only the initiator of cooperation, but it also plays an important role when a disabled person is being employed. An important point of successful employment is often how the emploeyer is satisfied with the agency. The employment of disabled people often brings certain risks connected with the limits of disabilities. Employers, who enable disabled people to work, are aware of that. The impulse for employment can vary from financial advantages, company philantrophy to personal solidarity of the employer. Social responsibility of the company is becoming a trend, that many employers start to know. But it can be presumed to influence the employment of more people with disabilities and at the same time the looking up the supported employment service by the employers. This bachelor's thesis can be contributive to understanding the importance of employment for disabled people's living. It brings information about the employers' experience with the realization of supported employment and motivation to the realization. It can also help to uncover integration obstacles of people with disabilities in open labour market.

The Oriental Institute in Prague: Cultural Department and the Orient

A substantial contribution to Oriental studies was founded with a new institution, the Oriental Institute. The idea was promoted by the Czechoslovak president T. G. Masaryk and by his advisor in Oriental politics, Alois Musil. The institute was financially supported by the ministry of Foreign trade, the main role played Minister Rudolf Hotowetz (first director of the Oriental Institute). The Oriental Institute was established in 1922 and financially and functionally stabilised in 1927/1928. There were no permanent salaried posts; however, the cultural section of the Institute financed publications and travel by means of fellowships and grants.

Health care professional as a patient

The bachelor's thesis deals with the topic of a health care professional dealing with personal experience in a patient's role. The theoretical part presents facts associated with health and ilness psychology, as well as pieces of knowledge about human personality and a man's role in human society. It describes the special demands of general nurse's profession on a person. Special attention is paid to a situation when a nurse becomes a patient. The research part analyses the influnce of nurse's profession over her personal health and ilness perception. It describes difficulties that are experienced when a nurse is coping with her own hospital admission. The main goal of the thesis is to find out whether personal experience in a patient's role influences nurse's future life in any way.

The role of p38-Mapk14/11 as an enabler of primitive endoderm (PrE) maturation and as a sensor of metabolism during mouse preimplantation embryo development

ANNOTATION This thesis describes the role of p38 mitogen activated kinases alpha/beta, also referred to as Mapk14/11 (p38-Mapk14/11), in regulating primitive endoderm (PrE) cell-fate and its sensitivity towards exogenously supplemented amino acids during mouse preimplantation embryo development. This study utilizing various pharmacological inhibitors, gene transcript over-expression and immunofluorescence assisted confocal microscopy imaging, has revealed a novel and pivotal role p38-Mapk14/11 in promoting germane maturation of the blastocyst stage inner cell mass (ICM)into the defined epiblast (EPI) and PrE and its potential requirement under amino acid deprived conditions.
