Characterization of nut oils

This bachelor thesis deals with the characterization of selected nut oils, almond and pistachio virgin oils. The teoretical part concentrates on the description of nut fruits, the characterization of aroma-active compounds, fatty acids and their content in the nut oils. After that, the fatty characteristics, which determines the oil quality, and instrumental methods selected for individual assignment are described. In the experimental part there were 42 volatile compounds identified in the nut oils by the headspace solid-phase microextraction coupled to gas chromatography-mass spectrometry, 35 were found in almond oil and 24 were found in pistachio oil. Free and bound fatty acids were converted into methyl esters using acid-catalysed esterification with boron trifluoride-methanol solution as a catalyst and then they were determined by gas chromatography with flame ionization detection. There were 12 fatty acids identified in almond oil and 14 fatty acids in pistachio oil. Oleic acid and linoleic acid predominated in both samples. The quality and properties of nut oils were valued by determining the fatty characteristics (acid value, saponification value, ester value, iodine value by Hanus method and peroxid value). The increased value of peroxide value was found in both oils which correlates with the trait and storage time of samples.
The assessment of aroma active compounds in elderberry juice

This work deals with the assessment of aroma active compounds in elderberry juice. In the theoretical part, the characteristic of elderberry, including species, varieties of elderberries, content compounds and use is described. The production of fruit juices and SPME-GC method, which has been chosen for assessment, is shortly summarized here. In the experimental part, identification and quantification of aroma active compounds in 11 samples with various content of elderberry juice combined with other type of fruit juice, diluted or not diluted with water, was made. The fiber CARTM/PDMS 85 µm was used for extraction of aroma compounds. 43 aromatic compounds were identified in total 5 aldehydes, 7 ketones, 18 alcohols, 5 esters and 8 fatty acids. The most of all aroma active compounds were determined in sample containing 10% of elderberry juice with 50% of apple juice and 40% of water, but some of them were not identified. The most of all compounds identified were determined in commercially acquired concentrated elderberry juice sample from Austria.
The optimization and validation of the GC-MS method for assessment of volatile aroma compounds in cheese

This work deals with the introducing of the method for the determination of volatile aroma active compounds in different types of natural and processed cheeses. The theoretical part describes the basic groups of aroma compounds. Than the methods used for their determination with focusing on solid phase microextraction and gas chromatography coupled with mass spectrometry are described. The experimental part deals with the optimization of the selected method parameters. For extracting volatile substances from the sample matrix the fiber DVB/CAR/PDMS 50/30 m was selected, these compounds were then determined by gas chromatography coupled with mass spectrometry. In the frame of validating proces the repeatability of the method was verified, RSDs of retention times
Assessment of aroma active compounds in elderberries

This diploma thesis deals with the determination of aroma volatile compounds in elderberry. The theoretical part describes the botanical characteristics of elderberry, the incidence and the other known species. Further, this work is focused on the chemical composition of elderberry, there are described in detail the content substances and briefly mentioned are medicinal effects of elderberry. Another theoretical part describes the natural processing of this significant plant and technological production of fruit juices. The second half of the theoretical part deals with the method of determining aroma volatile compounds by gas chromatography in combination with SPME technique, which is suitable for identification of aroma substances. In the experimental part of this diploma thesis SPME-GC method was applied to some samples of elderberry and aroma volatile compounds of individual samples of elderberry were determined.
Determination of antimicrobial and antioxidant activity of selected herbal extracts

Tetradenia spp. is herb naturally growing in Africa and is part of family called Lamiaceae. The herb is used in traditional medicine to cure malaria, angina or as treatment for tropical skin diseases. Maceration was chosen from number of extraction methods, to obtain bioactive compounds from this plant. Optimization was performed with aim to retrieve as much of bioactive compounds as possible. Optimal conditions chosen were: temperature 60 °C, extraction time 40 minutes, solvent 40% ethanol and solid-solvent ratio 1:5. In total 63 aromatic active compounds were found in optimal extract. Terpenes were the most abundant (88,60 %), from them oxygenated monoterpenes (55,65 %) and monoterpenes (29,52 %). Compounds with highest percentage quantity were camphor (51,76 %), cis--ocimene (8,17 %), 3-octyl acetate (7,71 %), trans--ocimene (6,76 %), -terpinene (4,93 %), o-cymene (3,81 %), terpinolene (1,33 %), limonene (1,33 %), camphene (1,19 %) and borneol (1,20 %). Further, total phenolic content was 0,31 ± 0,02 mgGAEml-1, which equals 1,52 ± 0,11 mgGAEg-1 of plant. Antioxidant activity was 417,56 ± 32,84 gTEACml-1 and percentage of radical scavenging activity ABTS•+ was 88,99 ± 7,00 %. Finally antimicrobial activity was determined against bacteria Bacillus cereus, Escherichia coli and yeast Candida glabrata. Against E. coli and C. glabrata there was no inhibition exhibited. Against B. cereus there was seen mild inhibition activity. The results confirm that Tetradenia spp. contain a number of interesting aroma active compounds, which shows antioxidant and antimicrobial activity. The herb has a good potential as natural source of nutritious and sensory active compounds to be used into various foods.
The chosen validation parameters of method for assessment of aroma compounds in elderberries

This bachelor thesis deals with the assessment of aroma active compounds in elderberries (Sambucus nigra L.). In the theoretical part the chemical composition and attributes of elderberries are described. Brief information on their healing properties is also given. The possible processing methods in food industry, as well as the traditional means of use at home, are discussed furthermore. Following chapters describe the gas chromatography method combined with the technique of solid-phase microextraction, utilized for the specification of aroma active compounds. The validation of this method is also included. The aim of the experimental part of the work was to verify a set of the selected validation parameters of the SPME-GC method, specifically linearity, the limits of detection (LOD), the limits of quantification (LOQ) and repeatability. Subsequently, the SPME-GC method was applied on the chosen elderberry juice samples. In total, 37 different volatile aroma active compounds were identified and quantified, 15 of which belonged to alcohols, 9 to aldehydes, 5 to ketones, 5 to esters and 2 other compounds. The chemical composition of individual samples was not uniform; no single sample contained all of the identified compounds.
Assessment of aroma active compounds in currant

This master thesis deals with the assessment of aroma active compounds in fruits of currant (Ribes spp.). The theoretical part was focused on basic characteristics and chemical composition of currants, as well as the basic principles of techniques used: solid phase micro extraction (SPME), gas chromatography (GC) and sensory analysis. The main aim of the experimental part was to determine aroma active compounds in currants using SPME-GC-FID method. The profile and scale tests were used for assessment of sensory quality and tastiness of fruits. In total 18 varieties, among them 6 white, 6 red and 6 black currants, were evaluated. Fifty aroma active compounds were identified including 21 alcohols, 10 aldehydes, 2 acids, 8 ketones and 9 esters. The results were statistically evaluated using principal component analysis (PCA) to investigate the influence of single compounds on sensory quality of fruits.
Characterization of the genus Plectrantus for use in the food industry

The aim of this diploma thesis was to determine the optimal extraction conditions for obtaining an extract from the plant Plectranthus amboinicus. Plectranthus amboinicus is an aromatic herb of the Lamiaceae family containing a large number of bioactive compounds. Because of this, it has a number of biological effects – antimicrobial, anti-inflammatory, antioxidant, analgesic, etc. To maintain the maximum of sensory and nutritionally valuable components of P. amboinicus, a simple maceration of fresh chopped herb was chosen. Maceration was performed under the following conditions: 40 minutes at 40 ° C, solvent 40% ethanol, weight 20 g of herb per 100 ml of solvent. The extract prepared under these optimal conditions was comprehensively characterized in terms of the content of total polyphenols, their concentration was determined to 0,08 ± 0,02 mgGEA·ml-1, which corresponds to 0,40 ± 0,07 mgGEA·g-1 of the plant. The antioxidant activity of the extract was 241,24 ± 29,24 µgTEAC·ml-1 and the radical scavenging activity of ABTS•+ was determined to be 49,29 ± 5,97 %. The antimicrobial activity was verified by the agar well diffusion method. Two types of bacteria were selected for the determination – gram-positive Bacillus cereus, gram-negative Escherichia coli and yeast Candida glabrata. The results showed that the extracts have the highest antimicrobial activity against B. cereus. There were 64 volatile aromatic compounds identified in the extract, with monoterpenes and sesquiterpenes dominating. The obtained results confirm that P. amboinicus, or its prepared extract, has a good potential for use as a new and non-traditional flavor in various foods and delicacies, which can also increase their nutritional value.
Study of the aroma profile of selected varieties of Moravian wines

The subject of this thesis was the determination of the profile of volatile aromatic compounds of varietal wines and the subsequent statistical analysis, aiming to find varietal markers of selected varieties – individual substances, their typical contents, mutual correlations. The HS-SPME-GC-MS technique was used to determine the aromatic profile. The main focus of this work was the analysis of the varieties Pálava, Hibernal, Johanniter and Solaris. The data obtained were statistically processed using descriptive and multivariate statistical methods. On the basis of these analyses, models were developed which, from the contents of 14 selected compounds of the aromatic profile, allow the Piwi varieties Hibernal, Johanniter and Solaris to be distinguished from each other and from the Pálava variety and, for the Pálava variety wines, to determine their origin in the Znojmo or Mikulov wine sub-region.
Influence of coffee making process on chemical composition of brewed coffee

This bachelor thesis deals with the influence of baristic coffee preparations on the chemical composition of the obtained beverage. Teoretical part of this thesis focuses on coffee growing, processing technology and the chemical composition of coffee beans. In the experimental part were measured samples of espresso coffee, samples prepared by moca pot, flip-drip method, filtrated coffee, samples prepared by french-press technique, vacuum pot samples and boiled coffee. The samples were prepared according to the baristic techniques at first and later with adjusted water and amount of coffee so these methods could be compared with each other. This work focuses on the determination of caffeine and acrylamide content by liquid chromatography, validation of HPLC method, determination of total phenolic content by Folin-Ciocalteu reagent, determination of organic acids by ion chromatography, determination of selected macro and micro elements by inductively coupled plasma in optical emission spectrometry and analysis of aromatic substances using gas chromatography. Based on the processed results and compared methods, it was shown that the moca pot had the greatest influence on the content of caffeine, acrylamide, organic acids and total phenolic substances, and the flip-drip with vacuum pot method had the greatest influence on the content of aromatic substances. From a baristic point of view the highest concentration of caffeine was 3689 mg/l in coffee based on flip-drip method, and concentration of acrylamide was 25,4 mg/l in the coffee based on the same method. The highest value of total phenolic content was determined in moka pot to c = 5,23 g/l. Regarding the mineral representation, the mineral decomposition of coffee beans and espresso were compared, and the highest values were reached by potassium, namely the concentration was 577 mg/l per 1 g of mineralized sample, in the cup of espresso the potassium concentration was 2461 mg/l. The highest content of organic acids was determined in the beverage prepared by moca pot, in terms of selected acids, the largest representation had the succinic acid with the concentration 599,8 mg/l. The highest content of aromatic substances was found in coffee from a moca pot and in coffee by flip-drip method. The biggest part was~made by furfuryl alcohol.
