Restoration of limestone in Grygov

The topic of the bachelor thesis is the restoration of former limestone in the village of Grygov. The document is based on an architectural study prepared within the subject AG034 Studio of Architectural Creation – Restoration of Monuments. The limestone building is located southeast of the built-up part of the village Grygov in connection with a significant landscape phenomenon of limestone quarries. Restoration is solved as a conversion, when the building is changed its original function. At the same time, it is a modernization of the building with an emphasis on preserving its original character. The intention was to convert the industrial building and industrial spaces into a multi-purpose cultural centre. The concept of restoration is to preserve the unique areas of the limestone factory, searching for a connection between the industrial character of the limestone factory from the 19th century and new, modern constructions. The object was approached by the principle of preserving what is valuable and replaceable or cancelling non-renewable parts. The proposed changes do not disturb the overall architectural character and appearance of the existing building. Dispositional changes are proposed by resolving the disposition of the limestone factory to the multifunctional centre. Unique spaces and structures will be preserved, namely an oval furnace, a walkway, a wooden ceiling, a perimeter wall made of quarry stone, a chimney and the eastern part of the roof. The building on the northwest side is in a demolished condition due to a collapsed roof structure. The spaces on the 1st floor are functionally divided into three parts. Exhibition, multifunctional spaces, sanitary and technical equipment, small restaurant with bar. The spaces on the 2nd floor are divided into two parts. A large hall, which is connected by a staircase to the ground floor. The attic space of the 2nd floor is divided by the chimney.
Identification and characterization of ciliary tip proteins

Distální konec řasinky/bičíku, také známý jako doména ciliární špičky (CTD), je nepostradatelná pro strukturu a funkci eukaryotických řasinek. Lepšímu porozumění této domény brání nedostatečné znalosti jejích proteinových komponentů. Cílem dizertační práce bylo ověřit lokalizaci vybraných známých proteinů CTD savčích řasinek, a testovat lokalizaci kandidátních CTD proteinů, jejichž ortology se nacházejí na špičce bičíku evolučne vzdáleného prvoka Trypanosoma brucei. Pomocí našeho lokalizačního postupu jsme identifikovali dva proteiny, které výrazně lokalizují do CTD primární řasinky. Jeden z těchto proteinů, ZC2HC1C, navíc také lokalizuje do stacionárních bodů podél axonemy, jejichž pozice se shodují s místy zastavení a otáčení intraflagelárních vlaků. Předpokládáme, že se může jednat o konce sub-distálně končících axonemalních mikrotubulů. Dále prokazujeme, že protein ULK4 lokalizuje do CTD pohyblivých ependymálních řasinek, ale ne do CTD primárnich řasinek, což je v souladu s dříve publikovanými fenotypy u ULK4 knock- outových myší a dokládá rozdíly ve složení CTD těchto dvou typů řasinek. Nakonec jsme ukázali, že Expanzní mikroskopie je rychlá a robustní super-rezoluční metoda, vhodná pro ultrastrukturální a lokalizační studie CTD savčích řasinek a bičíků T. brucei.

Experimental and numerical investigation of quantum turbulence in He II

Supratekuté 4 He (He II) je kvantová tekutina ktorej prúdenie je silne ovplyv- nené kvantovo-mechanickými javmi. Táto dizertačná práca predstavuje experi- mentálne a numerické štúdium turbulentného prúdenia v He II - tzv. kvan- tová turbulencia. Experimentálne je kvantová turbulencia študovaná v tepelnom protiprúde, v čistom supratoku a koprúde pomocou tlmenia druhého zvuku, precíznej lokálnej termometrie a pomocou vizualizácie héliových excimérových molekúl. Skúmaný je ustálený stav a rozpad hustoty vírových čiar v čistom sup- ratoku a v protiprúde a univerzálny kváziklasicý rozpad je charakterizovaný me- raniami efektívnej viskozity. Všeobecné dynamické správanie je študované de- tailne pre nestacionárny tepelný protiprúd, kde sú testované viaceré teoretické modely. Je navrhnutý nový model, v ktorom stredná krivost' vírového klbka závisí na hustote vírových čiar. Teplotná závislost' a zosilnenie intermitencie v kváziklasickom prúdení v brázde pohybujúcej sa mriežky boli pozorované pomo- cou vizualizácie. Numericky bola študovaná interakcia vírového klbka s pevnými trasovacími časticami, kde bola zistená spätná reakcia klbka na častice a dis- kutovaná je relevancia tohto javu pre vizualizačné experimenty....

Cieľom tejto bakalárskej práce je porovnať marketingovú stratégiu firmy Semo s. r. o. s trendmi svetového marketingu a popísať špecifiká podnikania na trhu s osivami. Vysvetľuje využitie alebo nevyužietie nástrojov marketingového mixu a stanovuje pozíciu firmy v porovnaním s konkurenciou.
Ekonomické efekty reformy Nového Zélandu

New Zealand has gone through remarkable reforms which have encompassed of wide range of government policies which have changed the incentives in private and public sector. This article tries to show the development of New Zealand economy before reform period, shows development of the reforms and tries to assess the economic effects of these reforms. First it analyses the effects in the primary sector, where it defines primary sector as itself and then analyses the incentives that have changed the sector output and conditions. Next the firm dynamics is being analysed as per the effects of the reforms. It is shown that first in some sectors the output proportion in GDP is lowering because of reforms were being made, later it shows this proportion to be rising again. Last, the view is made in effects analysis in labour market, where it has been found, that labour productivity is not growing enough, so the reforms were made too agresively. Next part of this article analyses macroeconomic effects such as public debt, which in brutto has been lowered through reforms to less than 30% of New Zealand GDP. First, inflation which was few years after reforms still in very high rates, but in 1990s it has been reduced well. And finaly the GDP growth itself, where it has been found out, that New Zealand has sacrifised a part of its GDP amount for agressive and fast reforms. Overlooking the period and its effects it has to be said that New Zealand has now a great market economy with transparent public sector, and the sacrifises were probably worth it.
