Sequences forming G-quadruplexes in the amyloid beta precursor human gene and its homologues
Stránská, Anna ; Šedrlová, Zuzana (referee) ; Brázda, Václav (advisor)
The APP gene encodes the transmembrane protein amyloid beta precursor, which is expressed in many cell types, including neurons. Its functions have not yet been fully described, but it is clear that it is being cleaved before being exported to the extracellular space. It is degraded by various degradation pathways also undergoes homodimerization, which can produce particles with protective neuronal function as well as fragments that are toxic and cause nerve cell death. The formation of harmful amyloid beta plaques and their accumulation among neurons in the brain is closely linked to the onset and progression of Alzheimer's disease, a neurodegenerative brain disease manifested by death and loss of neurons, which leads to dementia, i.e. loss of cognitive functions. There is currently a lot of research that deals with the links between neurodegenerative diseases and the occurrence of G-quadruplexes in genes that are involved in disease manifestations. G-quadruplexes are non-canonical DNA and RNA nucleic acid secondary structures that arise in guanine-rich regions. They are important mainly in terms of their connection with biological processes such as the regulation of gene expression in genes and mainly in oncogenes because they occur in important regions of the gene such as the promoter. It is possible to stabilize them with small molecules, and it is this ability that is used in research into the therapeutic treatment of various diseases. A bioinformatics analysis of both the human gene and 346 other gene homologs was performed to determine the importance of G-quadruplexes localization and conservation in the human APP gene. For this purpose, the G4Hunter program was used, which provided information about the found sequences with the potential to form a G-quadruplex, such as their location or G4Hunter score. In vitro analysis was performed using thioflavin T reagent, which tested the ability of the found sequences to form G-quadruplexes under physiological conditions. The results confirmed the presence and evolutionary importance of G-quadruplexes found in the APP gene of Homo sapiens and their ability to assemble into quadruplex structures in the presence of salts such as sodium and potassium.
Reactions of avian predators to automimicry in milkweed bugs (Heteroptera: Lygaeidae):
Stránská, Anna ; Exnerová, Alice (advisor) ; Baňař, Petr (referee)
Automimicry, or intraspecific variation in defence mechanisms in aposematic prey, is very common in nature. Especially in chemically protected prey. This study contains two experiments. The first experiment investigated the reaction of naive predators, which were Great tits (Parus major), to automimetic prey. The prey was the black-and-red-bug (Lygaeus equestris). The tits were divided into three experimental groups and each group was presented with three bugs that were fed on an artificial diet with different amounts of cardenolides. The control group was presented with prey that had no cardenolides in the diet. The group tested with the low concentration received bugs that fed on a diet with a low concentration of cardenolides and the group tested with the high concentration received bugs that fed on a diet with a high concentration of cardenolides. In a generalization test, all groups were then offered a single firebug (Pyrrhocoris apterus). It was found that the group tested with the high concentration experienced a higher rate of aversive learning. This group also killed and consumed fewer firebugs than the other two groups. The group tested with high concentration generalized most to novel red-and-black prey because they were least likely to attack the firebug. The second experiment tested...
Generation Z and Digital Work
Stránská, Anna ; Miessler, Jan (advisor) ; Wolák, Radim (referee)
The Generation Z and digital work diploma thesis examines the currently ongoing entry of individuals born between 1995 and 2012 into the work environment. The specifics of Generation Z's approach to digital work are examined in terms of their use of digital communication technologies, their experience of stress and perception of collaboration with older generations. The conducted qualitative research aims to provide insight into the way digital work is performed and seen by Generation Z.

Automimicry-intraspecific variation in antipredatory defences
Stránská, Anna ; Exnerová, Alice (advisor) ; Šulc, Michal (referee)
Aposematic prey is characterized by the fact that individuals are warningly-coloured and unpalatable to predators. However, variability in antipredator defence may occur within one species. The presence of intraspecific variation in antipredator defences is called automimicry. Automimics can be palatable or have a different level of quantity or quality of chemical defence. One of the ways through which the variability arises is for instance consumption of specific plants which defence against predators by means of sequestration. Intraspecific variation in antipredator defence affects the behaviour of predators that can distinguish between palatable of unpalatable prey. Chemical defence is costly for unpalatable individuals. Palatable individuals on the other hand save much energy by simply resembling unpalatable individuals of the same species.
Implementation of support measures for pupils with specific behavioral disorders in primary schools
Stránská, Anna ; Němec, Zbyněk (advisor) ; Felcmanová, Lenka (referee)
The diploma thesis is focused on providing support to pupils with specific behavioral disorders at ordinary primary schools. Its aim is to find out how support is provided to pupils with specific behavioral disorders at ordinary elementary schools, what are the potentialities for providing optimal support to these pupils and suggest possible measures and procedures to improve them. The thesis consists of theoretical and research part. The theoretical part characterizes specific behavioral disorders, education and support for pupils with these disorders. The research part of the thesis focuses on the implementation of support measures for pupils with specific behavioral disorders at selected elementary schools. This problem is solved in two levels. The first plane is qualitative, it is a case study of the school (author's workplace). The basis for the preparation of case report are interviews with school special pedagogues and unstructured observations. The second part is aimed at supporting pupils with specific behavioral disorders on a wider scale. The basis for the data is the questionnaire survey, which was attended by teachers and teacher assistants from other schools. According to the results of the research, the pedagogical staff focuses mainly on the attentiveness and behavior of pupils with...
The novel Shengxia de dou shuyu ni by Xu Xing in context of author's oeuvre and Chinese literature of the period
Stránská, Anna ; Andrš, Dušan (advisor) ; Lomová, Olga (referee)
This bachelor thesis deals with the semi-autobiographical novel Shengxia de dou shuyu ni (Whatever's Left is Yours, 1989-1996) by the contemporary Chinese writer and documentarist Xu Xing (born 1956). A particular emphasis is placed on the novel analysis, and the thesis also deals with the author's life and his oeuvre, the circumstances of the origin and publishing of the novel. Last but not least the thesis offers a comparision of the Chinese (self-censored) version and the German translation of the full (not censored) version of the novel under scrutiny.
Young Author´s Reviews at the Beginning of 20th Years of 20th Century (Most, Orfeus)
STRÁNSKÁ, Anna
The magazines Bridge and Orfeus belonged to temporarily tribunes (locations) of young starting authors at the beginning of twenties in 20. century, between 1920 ? 1923. This diploma work deals with their research. Attempts to analyse separated available issues of the magazines and approach mainly contemporary literature creation in this way, while defining also other cultural events (art, music, theatre). Followingly the issues of the magazines are compared. Periodicals are introduced in the view of their historical and cultural background of twenties. Beside avant-garde field is also followed the line of authors, who although standing aside of avant-garde style, actively participated in cultural life of that time.
