Microscopic Structure of Intestinal Anastomoses and the Use of Animal Models in Experimental Intestinal Surgery
Kural, Tomáš ; Tonar, Zbyněk (advisor) ; Kachlík, David (referee) ; Kocián, Petr (referee)
Despite extensive research and the implementation of modern surgical methods, the incidence of anastomotic leaks remains high. The exact pathophysiological mechanisms responsible for these leaks are still unknown, making prevention a challenge. The use of animal models is extensive in the research of intestinal anastomotic leakage. The literature review of this thesis, therefore, presents a brief history of the usage of experimental animals as well as major differences in terms of anatomy and histology between humans and the experimental mammals utilized in intestinal healing studies. The thesis is based on four studies describing the possibilities of using quantitative histological methods in different applications, animal models, and experimental surgical techniques in the research of intestinal anastomotic healing. The conclusions of these four studies can be summarized as follows: Conclusion 1: TeIGen software is readily accessible to scientists researching fibrous and porous materials using micro-CT analysis. This software aids in producing virtual image sets with defined morphometric attributes, and it can also help with fine-tuning of quantification tools required to examine micro-CT scans. The software also enables the identification of material morphological properties and image...
The role of the immune system in colorectal and ovarian cancer
Kocián, Petr ; Hoch, Jiří (advisor) ; Prausová, Jana (referee) ; Žaloudík, Jan (referee)
Prediction of clinical outcome in cancer is usually achieved by histopathological evaluation of tissue samples obtained during surgical resection of the primary tumor. Traditional tumor staging (AJCC/UICC-TNM classification) summarizes data on tumor burden (T), presence of cancer cells in draining and regional lymph nodes (N) and evidence for metastases (M). However, it is now recognized that clinical outcome cansignificantly vary among patients within the same stage. Data collected from largecohorts of human cancers has demonstrated the impact of immune-classification, which has a prognostic value that may add largely to the significance of the AJCC/UICC TNM-classification. In our study we examined the immune cells that infiltrated the tumor tissues of colorectal and ovarian cancer patients. In a cohort of newly diagnosed colorectal cancer patients we examined the correlations between the KRAS mutational status, patterns of tumor-infiltrating immune cells and the presence of tumor recurrence. Our data suggest that colorectal cancer patients with low levels of tumor-infiltrating lymphocytes, a high CD1a/DC-LAMP tumor-infiltrating dendritic cells ratio, and a KRAS mutation in codon 13 are at a high risk of disease recurrence. In ovarian cancer patients we focused on the dynamics of the tumor-infiltrating...
Comparative analysis of the current system of funding universities in the Czech Republic and in selected OECD countries
Kocián, Petr ; Vostrovská, Zdenka (advisor) ; Barák, Vladimír (referee)
This Barchelor thesis deals with the current system of funding universities in the Czech Republic. The main goal of thesis is find out or create some improvement proposals of universities funding in Czech Republic. These proposals are based on comparsion of education systems (US, Great Britain and Slovak Republic). The theoretical part describes fundamental theoretical aspects university funding, different systems of university funding, indicating their essential characteristics and identification of differences of systems. The main attention is paid to the system of university funding in Czech Republic. The practical part is a presentation of the results of a comparative analysis of relevant OECD indicators in these funding systems - with reference to strengths and weaknesses. Results of the study demonstrate that for the main weaknesses of of the current system university funding can be considered: 1) the high proportion of public funds and on the other side - very low proportion of of funds from private sources; 2) very low support for students from public funds and on the other side strengths of czech systém is the share of expenditures on R & D in tertiary education. Disadvantages of the system with tuition fees at public universities are reflected in Great Britain. Students in GB are faced to the higher risk because of fees and their debts means burden on public finances due to pay off student loans. (In conclusion, recommendations are shown)
