Proteome of the encystation-specific vesicles in Giardia intestinalis.

The process of encystation is a vital and important part of life cycles of many organisms. One of them is Giardia intestinalis, a significant single cell parasite of vertebrates, including man. During the process of encystation there are unusual organelles, called encystation vesicles. These serve to accumulate material for the future cyst wall. The main proteins found in encystation vesicles that compose the cyst wall, are CWP1, CWP2 and CWP3. This work is focused on these proteins. The structure and properties of these proteins were used to study the main aim of this work, that is the identification of other proteins associated with the encystation vesicles. In the experimental part, a system was created, that allows targeted characterization of proteins and cell compartments, by utilization of biotin ligase of Escherichia coli. For this systém, we created specific constructs, that were successfully inserted into plasmids and into G. intestinalis cells. For the intention of cloning, a method of direct site mutagenesis was optimalized. The expression of chosen proteins was detected successfully during the in vitro encystation. The thesis investigates impal of external eviroment on the firm and it also analyses internal enviroment. The thesis defines strategic targets, contains proposal of competitive strategy of the company and time of its implementation.
The role of mitochondria in the pathogenesis

This thesis is focused on the existing data referring to role of mitochondria in the pathogenesis of several important diseases. It introduces mitochondrial proteins, their role and metabolism, dysfunction of which is behind pathogenetic processes. The thesis also summarizes possible mitochondrial damage, its progress and consequences, which can lead to diseases or aggravate their process. It also pays attention to the role of mitochondria during oncogenesis as well as important neurodegenerative disorders such as Alzheimer's or Parkinson's disease, Charcot-Marie-Tooth disease, amyotrophic lateral sclerosis or autosomal dominant optic atrophy. The thesis mentions the role of disrupted mitochondrial dynamics in type 2 Diabetes. In conclusion the thesis mentions the role of mitochondria and their damage in relation to infection by the parasites Toxoplasma gondii and Trypanosoma cruzi.

The aim of the diploma thesis was to process the dried drug of Zanthoxylum nitidum and to obtain the total extract. By means of preparative thin layer chromatogramy one alkaloid was isolated and based on structural analysis (NMR, MS) and comparison of data from the literature identified as (-)-edulinine. This alkaloid was tested for its inhibitory activity against human erytrocyte acetylcholinesterase activity (HuAChE) and human plasma butyrylcholiesterase (HuBuChE). The measured values for (-)-edulinine were IC50 HuAChE > 1000 µM a IC50 HuBuChE > 1000 µM. Galanthamin (IC50, HuAChE = 1,710 ± 0,065µM, IC50 HuBuChE = 42,30 ± 1,30 µM) and huperzin A (IC50, HuAChE = 0,033 ± 0,001µM, IC50 HuBuChE = >1000 µM) were used as positive standarts which are used in therapy of Alzheimer's disease. In comparison of standarts (-)-edulinine was inactive and it can't be considered as perspective substance in treatment of Alzheimer's disease. TITLE: Analysis of the evaluation criteria of kindergarten headmasters by founders in Pardubicky region AUTHOR: Bc. Lenka Marková DEPARTMENT: School management centre SUPERVISOR: Mgr. Jiří Trunda ABSTRACT: This thesis analyzes the current criteria for evaluating the work of kindergarten headmasters. The criteria were created by community representatives as founders of the kindergarten. The thesis also focuses on identifying indicators leading to the fulfillment of the evaluation criteria. Monitoring headmasters views on the current criteria, which they are valued, is the part of research. Through the analysis criteria, surveys and informal interviews with representatives of the founders and headmasters the deficiencies in the structure and list of criteria were found. The research has shown inconsistency and formality outlined evaluation criteria with absence of their indicators. Based on the findings was recommended evaluation criteria to include all areas of the kindergarten headmasters. Indicators, that show the way to meet the criteria, were also suggested. Development of Arnošt Lustig's Prose

The aim of the thesis was to discover the processes of Arnošt Lustig's interference into his previously published work and giving them the final form. The author of this paper was interested in the changes being only supplementary additions of the story or if the meaning is significantly influenced by writer's editing. At the same time, the question about the reason, function and meaning of the changes was raised. For the research three Lustig's books, were chosen: Noc a naděje, Modlitba pro Kateřinu Horovitzovou and Dita Saxová. They slightly vary in genre. The first chapter Poetics of Arnošt Lustig deals with Judaism as his inspiration, context of the time period in which he was active - the second wave of War prose, the topics of his prose being rather mono-thematically from Jewish background and with his characters, weak and humiliated people who remain moral winners although they often die. The second, third and fourth chapter are more about individual books. Their first publication is compared with the last one. The added texts are then sequenced into logical units, described and commented. All chapters contain short conclusion. The last closing chapter presents the results of the thesis. Performance traits of dairy cows stabled in tie stalls

The aim of this thesis was the evaluation of fertility and production traits stabled in tie stalls. 50 pure-bred Holstein cows were monitored and those traits were evaluated: interval of insemination, service period, calving interval, milk yield and content of milk protein and fat. All monitored cows finished standard lactation period (305 days) in year 2012. The evaluation was performed in relation to lactation order. Nearly all of fertility traits were evaluated as bad. The average interval of insemination was 83.7 days, service period was 171.6 days and calving interval was 444.9 days. Interval of insemination was rising up to 3rd lactation. However 4th and following lactation insemination interval was lower about 9.6 days in comparison with 3rd lactation. Service period values were getting worse according to higher lactation order, 2nd lactation service period was shorter about 19.2 days. Moderate statistical difference was found in calving interval between 2nd (505.4 days) and 3rd (395.1 days) lactation. Only 3rd lactation calving interval could be evaluated as satisfying. Calving interval values were rising up from 4th and higher lactation. The average milk yield of tie stabled cows was 9,869.5 kg per standard lactation period (content of milk fat 3.96 % and content of milk protein 3.44 %). Milk yield was rising up to 3rd lactation when it reached maximum values and slight decrease was recorded on 4th and following lactation. The average content of milk fat was increasing following higher lactation order. Statistically significant difference was found between milk protein content of 3rd lactation (3.37 %) and milk protein content of 4th and following lactation (3.53 %).
