Postural stability was measured using computerized dynamic posturography SMART EquiTest System from Neurocom. Measured data were processed using the program NeuroCom Balance Manager software. In the Framework of study were conducted two intrumental measurements in the range of two months. After the first measurement all probands were instructed how to...
The Presence of Silence in Human Lives and Relationships
Skýpalová, Monika ; Milfait, René (advisor) ; Žáková, Lenka (referee)
This bachelor thesis deals with the presence of silence in human lives. The thesis concretely identifies the silence as a very important part of human lives because the silence helps people to find their spirituality. On the other hand this thesis says that people are afraid of silence because they don't know what could be found in the silence. In the first part of this thesis the conception of silence isspecified. In this part there is also the quality of silence important for human spirituality discussed. This part is also engaged in the development of silence in monasteries where silence is often used. The experience of st. Benedict are especially mentioned. The other part of this thesis is aimed to the fear of silence. It also describes the possibilities how to reduce this fear and how to work with silence in our lives. The applied part of this thesis introduces the own experience of the author with silence in The Silence cafeteria and in the Trappist monastery. It also presents several dialogues about human relation to silence.
Synthesis of peptides as potential ligands of GCPII protein as identified by phage-display method
Trajhan, Filip ; Konvalinka, Jan (advisor) ; Žáková, Lenka (referee)
Glutamate carboxypeptidase II (GCPII, PSMA-prostate-specific membrane antigen) is a human transmembrane metalloprotease highly expressed in prostate cancer, which makes it a suitable target for diagnostics or targeted cancer therapy. Therefore, specific and tight-binding ligands of GCPII are needed. One way of finding potential low molecular weight peptide ligands is the screening of combinatorial libraries. One of the methods of high-throughput screening is phage display, which uses genetically modified bacteriophages for the study of interaction between peptides and target protein. A library of potential peptide ligands was prepared based on the results of phage display targeted against various forms of GCPII. Parallel peptide synthesiser SPENSER was used for the synthesis and subsequent cyclization of linear peptides. The prepared bicyclic model peptide was characterised and his binding affinity for studied enzyme was confirmed. The possibilities of enhancing the cyclisation efficiency and the use of optimized cyclisation step for preparation of other peptide libraries are discussed. Key words Glutamate carboxypeptidase II, phage display, peptide synthesis, bicyclic peptides, combinatorial chemistry

Title: Comparison of Special Breathing Exercises versus Specialized Physiotherapeutic Interventions for Static Apnea Objectives: The aim of this diploma thesis is to look deeper into the issue of improving performance in static apnea, one of the competitive disciplines of freediving and compare its training with 1) special breathing exercises for divers (Mana, 2018) and 2) physiotherapeutic intevrentions based on working with the diaphragm and deep stabilization system of the spine (Kolář, 2009). The aim was to find out which group achieved a more effective result and what is the overall diference between the training result of both groups in terms of posture quality and performance in static apnea. Methods: The experiment involved 14 healthy people, men and women, aged 19-23 years old. The experiment consisted of three parts: 1) Initial measurement of static apnea and examination of four tests to determine the quality of one's postural muscles, 2) four-week lasting training according to the division into groups (breathing exercises for divers or training based on physiotherapy intervention), 3) final measurement of static apnea and re-examination of posture tests. The obtained data were recorded and processed. Results: It appeared that due to the small amount of statistically signifiant data and...
IGF2 and preptin, biologically active peptides derived from pro-IGF2
Mrázková, Lucie ; Žáková, Lenka (advisor) ; Janovská, Petra (referee)
Pro-IGF2 is a 156 amino acid long protein and it is a prohormone from which two bio- logically active peptides are formed, IGF2 and preptin. IGF2 is an insulin-like growth factor 2 that, together with IGF1 and insulin, shares a similar 3-D structure and similar biological effects. It is a peptide whose sequence corre- sponds to amino acids 1-67 of pro-IGF2. It is produced mainly by the liver, but it is formed in most cells of our body. It is involved in the regulation of growth, differentiation, embryogenesis and in recent years the interest in IGF2 has been increasing mainly due to its function in the central nervous system. Preptin is a newly isolated 34 amino acid long peptide whose sequence corresponds to amino acids 69-102 (E-domain) of pro-IGF2. Preptin affects glucose metabolism as well as bone homeostasis. Disorders in glucose metabolism usually lead to diabetes mellitus, and disorders in bone homeostasis most often lead to osteoporosis. Preptin has been shown to increase the second phase of insulin secretion and to have a positive effect on the proliferation and differentiation of osteoblasts, bone cells promoting bone growth. Keywords: pro-IGF2, preptin, IGF2, E-domain, gene expression, imprinting IGF2
Study of interactions between low-molecular mass compounds and the DNA-binding domain of forkhead transcription factor FOXO3
Kohoutová, Klára ; Obšil, Tomáš (advisor) ; Žáková, Lenka (referee)
This bachelor thesis is part of a project focused on studying low-molecular mass compounds able to inhibit the interaction between DNA-binding domain of human forkhead transcription factor FOXO3 and the target DNA. FOXO3 is one of four members of FOXO class transcription factors which belong to forkhead family of transcription factors. Forkhead transcription factors are evolutionary conserved proteins playing important roles in numerous cellular processes. These include cell-cycle regulation, oxidative stress response, control of cellular metabolism and apoptosis. FOXO3 plays an important role in cancer cells where it acts not only as a tumor suppressor but also can enhance their resistance to chemotherapy. Considering its biological functions, the study of small-molecule inhibitors of FOXO3 transcription factor is of particular importance. This bachelor thesis was focused on compound S9 oxalate as a potential inhibitor of FOXO3-DNA interaction. Main goals of this thesis were: (I) preparation of both unlabeled and 15 N labeled DNA- binding domain of FOXO3 transcription factor, (II) characterization of interactions between FOXO3 DBD and compound S9 oxalate using NMR and electrophoretic mobility shift analysis (EMSA), and (III) prediction of binding conformation and interactions between FOXO3 DBD and...
