Foundations of Marie Margaret Countess of Waldstein

RESUMÉ MARIE MARKETA OF WALDSTEIN'S FOUNDATIONS This bachelor's work represents Marie Marketa, Countess of Waldstein, as the founder of sacral objects in her estates, and presents an overview of buildings with an explanation of their origin in the given locality. Bachelor's thesis is part of research on Waldstejn's patronage in northern Bohemia prepared for the exhibition of this important aristocratic family in the Museum of the Bohemian Paradise in Turnov. During my research, I did mapping of individual sites and I described them. From my point of view the work in the field was important, this means acquisition of photographs of individual churches and gathering additional information on the creation of buildings. Most reference period was the years 1724-1729, when the Countess of Waldstein was truly generous in promoting the arts, when the favorable financial situation help her. Thanks to this situation newly built or restored religious buildings on Waldstein's estates were built. Such as decanal church of St. James in Mnichovo Hradiště, St. Nicholas in Turnov, and St. Vojtěch in Kruh near Doksy, St. Wenceslas in Boseň and the chapel of St. John of Nepomuk at Waldstein, St. Stapin in Klokočka. 20th CENTURY AS AN EXPERIENCE OF ONE CONCRETE WOMAN

The aim of my diploma work is to find, how are concrete periods and events reflected in a life of concrete women, how were her acts influenced by gender norms and stereotypes of the society, and how much felt this woman limited by them. As a method was chosen the oral history, that enables working with the life stories of people of all kinds of social status and experiences. Its based on biografical life stories, that are analized and interpreted by the research. In the case of my narrator was chosen my grandma, born in the 1922 in agricultural society. This caused that her life experiences dont correspond with the picture of her period, that we know from the historiographical mainstream production, accenting the city middle classes. Therefore it is - by usány the oral history and the analytical category gender - possible to make an original wiev at the events and period of the twentieth century in the way lived by concrete women. This wiev, often marginalizing political events accented by mainstream history, and accenting everyday life influenced by the gender system more, is the vivid proof how much different can be the apprehention of the life according to the context and the concrete life experiences.
History of Skydiving - accuracy landing

Title: History of skydiving - precision landing Objectives of work: The aim of this work is to map complexly and integraly the history of skydiving with a focus on racing discipline of precision landing. Due to insufficient number of relevant compact resources and web sites, so the work becomes the first file that holds the issue in a comprehensive manner and develop a unified overview of the analysis problem. Method: This is a theoretical work, the kernel of the work will be at tracing specialized publications and websites, collecting information, filtering the relevant data in their comprehensive use. Results: Objective of thesis was largely achieved. It turned out that the topic is very broad to draw up a detailed summery of the entire history of the research question. The topic was therefore processed generally and is more like a preview than a perfect description. Intended overview was given. The Premises of Successful Cooperation between Coach and Athlete in athletics

Title: The Premises of Successful Cooperation between Coach and Athlete in athletics Objectives: The aim of this thesis is to determine the precondition for successful cooperation between coach and athlete. Methods: Literature research was chosen as a method of this thesis while printed, as well as electronic resources were used. Results: The given objectives and tasks of the thesis were successfully met. While working out the literature research, we came to the conclusion that the basic premise for successful cooperation between the coach and athletes is effective communication. We determined conditions that are necessary for successful communication in the specific environment of athletics. There are objectives set by athlete and coach, choice of suitable coach, development of communication skills, and athlete's perception of his coach's authority. Female choice based on colouration and song performance in passerines with sexual dichromatism

This thesis is about female mate choice and its influence on two most typical phenotype characteristics of sexually dimorphic passerines (Aves, Passeriformes): their colouration and song. This thesis reviews a range of scientific papers focused on this problematic. Results of these papers show that both colouration and song can serve as an honest signal and give information about health, condition and other characteristics of its bearer. Many papers also indicate that females put such male signals into account when choosing a mate. Special emphasis in this thesis is put on papers which dealt both with colouration and song. Although the number of such papers is very low, it is clear that the quite widely accepted opinion that brightly coloured birds do not sing quite melodic songs and vice versa is generally not quite correct. The biggest problem in the study of female mate choice is the lack of papers which dealt with more than one phenotype characteristics.
Josef Václav Scheybal: a documentalist and a collector

Josef Václav Scheybal : A documentalist and a collector The diploma thesis deals with Josef Václav Scheybal (1928 - 2001) and his activity mainly in northern Bohemia. It attempts to capture all aspects Scheybal's personality, because he was a painter, graphic artist, illustrator, art historian and ethnographer. He was an expert of folk architecture and sculpture, folk furniture, technical equipment, the popular clothing, prints, folk songs, was the creator of multiple exposures. He also cooperated with the Institute of ethnography and folklore, ethnographic research, participated throughout the country, with results published regularly. His focus on folk architecture resulted in a lifetime's research and documentary work. He gathered the valuable work of drawing documents, publications of folk architecture and free of the open air. The thesis also includes his family, the father Josef Scheybal and his wife Jana Scheybalová. Cold adaptation in stationary phase in Bacillus subtilis

Cold adaptation in stationary phase in Bacillus subtilis One of the most important abiotic factor which influences life of bacterial cells is the ambient temperature. A decrease of this temperature is usually accompanied usually with the loss of the fluidity of bacterial cytoplasmatic membrane. While the mechanisms of the responses to the cold shock during the exponential phase of growth are well known for Bacillus subtilis, the responses of stationary phase cells had not been studied yet (despite the stationary phase is the most common state of microorganism in the nature). There are two independent mechanisms which restores much needed fluidity in Bacillus subtilis - short-term adaptation and long-term adaptation. Short-term adaptation is based on the function of fatty acid desaturase coded by des gene. Long-term adaptation relies on the change in ratio of iso- and anteiso- branched fatty acids. In this work we examinated membrane adaptation during stationary phase under two different conditions, namely under cultivation at stable low temperature and after cold shock. The highest activity of Pdes was observed for cultivation at 25 řC and for the cold shock applied from cultivation in 37 řC to 25 řC. Abstract The thesis deals with the reaction of body to postural loading during stance and walking. The theoretical part of this thesis is focused on postural control and postural reaction during stance and walking. Particular attention is paid to gait cycle, dynamic plantography and Zebris FDM-T Rehawalk system, which has been used in the practical part of this thesis. Two chapter deal with postural loading, the first is focused on symmetrical loading (e.g. backpack) and second on asymmetrical loading (laptop back - ipsilateral, contralateral and held in hand). The practical part presents the examination of 32 volunteers who were either standing or walking on a dynamic plantograph. The volunteers were exposed to either symmetrical or asymemtrical 5 or 10 kg load. Study of phosphorylation of inorganic pyrophosphatase from Streptococcus pneumoniae

The human patogen Streptococcus pneumoniae encodes a single copy of eukaryotic-like Ser/Thr protein kinase StkP. StkP regulates virulence, competence, stress resistence, gene expression and plays a role in the regulation of cell division cycle. Analysis of phosphoproteome maps of the wild type and stkP mutant strain of S. pneumoniae showed that in vivo StkP phosphorylates several putative substrates including Mn-dependent inorganic pyrophosphatase PpaC. Mass spectrometry analysis identified two phosphorylation sites in an active site of the protein. Pyrophosphatases are essential enzymes that catalyze hydrolysis of inorganic pyrophosphate produced during various biosynthetic reactions that utilize ATP. Changes in pyrophosphatase activity have been described to have global effects on cell metabolism, growth and division of bacteria. The aim of this thesis was to investigate the phosphorylation of inorganic pyrophosphatase PpaC in S. pneumoniae. Gene ppaC was cloned, expressed in E. coli and protein was purified via affinity chromatography. Phosphorylation of PpaC by StkP was examined in a kinase assay but we did not confirm that PpaC is a direct substrate of StkP in vitro. Further we prepared a set of mutants in ppaC gene.
