Pre-clinical model of acute promyelocytic leukemia: Study of the anti-leukemic effect induced by ATRA and DNA vaccination

DOCTORAL THESIS 2012 POKORNA Abstract We have used a well characterized transplantable transgenic mouse model which mimics human acute promyelocytic leukemia (APL), both in its biological characteristics and its response to conventional therapeutic drugs. The aim of our study was to better characterize the efficacy of the combined treatment and to determine molecular markers of clinical outcome. We established a minimal residual disease monitoring based on the high sensitivity of detection of PML-RAR transcripts by polymerase chain reaction (PCR) technology in APL mice. We showed that oncogene-specific PCR-based assays allow, like in patients, the diagnosis, follow-up and prediction of disease evolution. Furthermore, PCR assay was used to assess various tissues and organs for the presence of PML-RAR-positive cells in minimal residual disease free long-term survivors. As expected, majority of mice had no measurable tissue level of PML-RAR demonstrating the efficacy of immunotherapy. However, tracking the oncogene-positive cells reveals for the first time that extramedullary PML-RAR-positive cell reservoirs such as the brain may persist and be involved in the leukemia relapse. Diagnosis of the nutritional status focused on overweight and obese population

Title Diagnosis of the nutritional status focused on overweight and obese population. The aim of the work The aim of thesis is to summarize and describe the methods that are used for determining the nutritional status of overweight and obese individuals. Methodology The thesis is a theoretical work with a search character. The main method in the creation of this type of work is a critical content analysis of available resources for the work, and the subsequent comparison of selected findings about diagnosis of nutritional status of overweight and obese population. Results Review article shows that in the clinical practices of identifying a person's nutritional status of overweight or obese at the first contact with the examined using BMI. The orientation index is also used index WHR. Due to the approximate nature of these indexes, however, provides for additional precise examinations body composition, which are for example caliper measurements or bioimpedance. Essential components of a diagnosis of nutritional status are also constitutional typology, family and personal history, immunological examination, biochemical tests and monitoring dietary habits. Media Representation of Farmers'Markets

The aim of the thesis "The media image of farmers' markets" is to describe the development of the media image of Prague farmers' markets, as presented by MF DNES in 2010 and the first half of 2011. The research was carried out using a combination of quantitative and qualitative analysis and of a survey among the market shoppers. The result is a comparison of the three analyzed periods and focuses on an evaluation of the phenomenon of farmers' markets and presentation of the problems that accompanied them. The analysis showed that the media image in 2010 was uncritical. Articles about farmers' markets were distinctly favorable, showing only positive feedback. Markets were also associated with policies of the respective Prague districts, which were given credit for establishing them. Although the daily began to report on problems of the markets in the first half of 2011, survey among the shoppers showed that negative messages did not affected their shopping behavior and confidence in markets.
Transformations of tramping movement after 1989

The bachelor thesis "Transformations of tramping movement after 1989" investigates changes, which the tramping movement underwent after the fall of the Communist regime. Tramps were persecuted before 1989 and could freely express themselves only in very limited ways. The work attempts to answer the question, of how tramps have coped with the new freedom and how it changed the movement itself. The research was carried out by qualitative means, because current tramping is almost totally neglected in expert literature. The method of autobiographical narrative interview was followed throughout the data collection process. It allows one to analyze the changes mentioned by the informants in the context of their life story. The empirical part of the thesis presents the changes of tramping, their origins, and their evaluations from the narrators' perspectives. The part also investigates the impact of tramping itself on their lives and the changes of their perception of the movement throughout life. Despite tramping not being officially banned nowadays, tramping suffers significant hardship from state and national park rangers. The Personnel Dilemmas about Home Hospic Care in the Area of Havlíčkův Brod

The thesis discusses dilemmas situations of The hospice staff in Havlíčkův Brod. The occurrence of these situations concerning a social worker, medical and caring staff was determined. The aim of the thesis was to find out how these dilemma situations are solved. The theoretical part of this work describes issues related to the hospice care. It defines the history hospice care and the current hospice care in the Czech republic. The work is also dedicated to the palliative care, mainly to the hospice home care. It considers closely issues of the death, dying, dying person and also his relatives, who take an irreplaceable role in this process. There is also the important description of the multidisciplinary team which creates base of providing palliative care for dying people and their relatives. At the end of the theoretical part, I concentrate on dilemmas and problematic situations. The practical part came from the quality research. The tool of this research was the questioning and the method of the instruction interview. There were three participants in the research. The data processing from the interviews was evaluated by the method Open coding. It was proved by the research that every dilemma situations was always solved on an individual basis of the hospice home care staff with the respect to the bio-psycho-socio-spiritual side of the client and his relatives. The caring staff try to support the idea of hospice movement which is raising public awareness of their work among professional and non-professional people. They acknowledge the teamwork, they solve the problematic situation with the intervision, they use their own experience and they improve their specialization by education. It is clear from the research that the staff need to be supported by their families to cope with difficult situations and also the faith in the sense of their work gives them strenght.

Business Development in Social Services in the Region

The bachelor thesis focuses on financing of social services in the region. The thesis is specifically aimed at chosen retirement homes in the South Bohemian region. The aim of the study is to analyze and compare the financing of these retirement homes. These homes have different founders - the state, the municipality, or a private entity The theoretical part of the work contains description and explanation of basic terms and definitions, which serve for better understanding of the topic. For the practical part, chosen retirement homes were contacted. Based on personal visits, the data necessary for financial analysis was provided. This data consists of profit and loss statements, annual reports and additional information. In conclusion, the chosen retirement homes were compared in the areas of cost efficiency and profits, which represent the financial sources to cover the costs, using the method of costs per bed in a given time interval. The comparison is aimed at financial resources in the present according to the law n. 108/2006 Coll. about social services.
Gifted pupil in contemporary school

Key words: gift, talent, education of gifted children, teaching methods and forms of working oganization This graduation theses is focused on gifted children and their signs. The theoretical part contains legislature relevant to educating of gifted children in the Czech Republic and various options of the optimalisation of educating process for gifted pupils. It also focus on different methods of recognising gifted children, educating them in school, options of educating and its problems, it deals with the formes of organisation of the teaching methods and basic approaches to educating of gifted pupils. The experimental part of this work is divided into two parts. The first one shows the pupils' oppinion about education, who visit the school for gifted children, the second one contents interview with teachers and examples of the good work with educating children.
