MicroRNAs in Human Cancers Associated with Viral Infections
Dvořáková, Lucie ; Tachezy, Ruth (advisor) ; Drda Morávková, Alena (referee)
MicroRNA (miRNA) are short single-stranded RNAs that do not encode proteins. Their main function is the regulation of the gene expression on the level of translation. This regulation is mediated by the binding of miRNA to the partially complementary segments of mRNA, both cellular or viral. It is estimated that miRNAs affect expression of at least one third of human genes and thereby influence regulation of cellular growth, differentation and apoptosis of cells. Recently the miRNAs encoded mainly by DNA viruses were discovered. These miRNAs enhance the persistence of viral infection in the host and can contribute to malignant transformation. However, the oncogenesis is also significantly affected by the regulation of cellular miRNAs expression by viral proteins. The miRNA research is topical. MiRNAs are considered as potential biomarkers and their utilization as a cancer therapy is being intensely explored. In this thesis, I'm describing the biogenesis and regulatory functions of miRNAs. I'm also presenting an overview of viral miRNAs focusing on human oncogenic viruses which do not only code their own miRNAs but also influence the expression of the host miRNAs. Finally, I am focusing on current clinical applications of miRNA. Key words: viral miRNA, cellular miRNA, oncogenesis, viral infections,...
Data processing in accounting programs
DVOŘÁKOVÁ, Lucie
This thesis compares and assesses accounting data processing in two chosen programmes Pohoda and Ekonom. The first part of thesis is a literary summary of the theme historical progress of the accounting, particular phases of the progress included, and describes economical infor-mation systems and general standards for the right choice of accounting programme for a firm. The analysis goes in the module of billing, stocks and pay. The author assesses both of the programmes and compares their methods. In the next part of the thesis are given advantages and disad-vantages for users. The last part of thesis describes a concrete firm, her way of account-ing processing included. On the basis of the price calculation of the programmes the author will recommend a system for the accounting data processing.
Modifications of constitutional law in relation to the accession of the Czech Republic to the European Union
Dvořáková, Lucie ; Reschová, Jana (advisor) ; Syllová, Jindřiška (referee)
MODIFICATIONS OF CONSTITUTIONAL LAW IN RELATION TO THE ACCESSION OF THE CZECH REPUBLIC TO THE EUROPEAN UNION ABSTRACT The accession of the Czech Republic to the European Union was certainly one of the most important moments in the development of Czech legal order. The impact of this step may be seen in every branch of law - including constitutional law. The large number of publications on this issue indicates that the changes of constitutional law are an important issue in the Czech academic society. Yet, the literature is mostly fragmentary and mainly oriented at only some of the problems. This thesis tries to gather all the important changes in the branch of constitutional law, which occurred in relation to the Czech Republic's accession to EU. As the constitutional law is a very broad topic, the focus is laid on its institutional part and on the role of member states (and its institutions) within the EU. This thesis consists of an introduction, four chapters (which are further divided with respect to the single topics), and a final conclusion. In introduction, the focus of the thesis and basic questions are defined. Chapter One briefly follows the process of the Czech Republic's accession to the EU. Chapter Two deals with the position of Member States within the EU. It touches on issues such as the...
Molecular evaluation of novel BCR/ABL kinase domain variants in patients with chronic myeloid leukemia
Dvořáková, Lucie ; Peková, Soňa (advisor) ; Kozák, Tomáš (referee)
1 Abstract BCR/ABL is a constitutively active tyrosine kinase that has been shown to be at the heart of the development of chronic myeloid leukemia (CML) and about 30% of acute lymphoblastic leukemia (ALL). With the recent advent of tyrosine kinase inhibitors (TKIs), exemplified by Imatinib, Nilotinib, Dasatinib and Bosutinib, patients with Ph+ CML or ALL are candidates for the therapy with these agents. From the available TKIs, Imatinib is considered as front-line therapy for CML patients in chronic phase, while for Ph+ ALL patients, 2nd generation TKIs (nilotinib, dasatinib, bosutinib) might be considered as more effective therapeutic option. Since the treatment with TKIs is a long-term affair, a substantial proportion of patients acquire some sort of mutation in kinase domain of BCR- ABL, which could be a reason of treatment failure. To date, over ninety BCR/ABL kinase domain mutations have been identified, affecting over 50 amino acids. Recurrent BCR/ABL kinase domain mutations have already been in vitro tested to approximate for their in vivo behavior. Our goal is to invent in vitro technique that would allow testing TKI sensitivity of novel BCR/ABL kinase domain mutations, identified at very low MRD levels. The technique makes use of site-directed mutagenesis to create the novel BCR/ABL kinase domain...
