Use of specific inhibitors of carbonic anhydrase IX for fluorecent imaging of cancer cells

Human carbonic anhydrases are metalloenzymes that are involved in many physiological processes in the body, but also play an important role in the pathogenesis of numerous diseases. Under regular conditions, expression of carbonic anhydrase IX (CAIX) is very limited, unlike that of other 14 human carbonic anhydrase isozymes. But in hypoxic tumors this enzyme is highly overexpressed on the cell surface. For this reason, this enzyme represents a good target for therapy and diagnosis of tumors and thus various anti-CAIX monoclonal antibodies and specific inhibitors are being developed. In this work we investigated the possibility to use fluorescent polymer conjugate carrying a CAIX specific inhibitor for florescent labeling of tumor cells. Specific binding of polymer conjugate to different cell lines was investigated by flow cytometry and confocal microscopy. Binding properties of the polymer conjugate was compared to CAIX specific monoclonal antibody M75 and its single-chain fragment scFv M75. Ability of the polymer conjugate to inhibit CAIX enzyme activity was also investigated. For these experiments, recombinant protein CAII was prepared and purified, which was also used for protein crystallization. Historical Changes in the Discourse of Sexuality in the Czech Criminal Law

This thesis is focused on historical changes in the social constructions of sexuality, particularly in the czech criminal law. Primary sources for the analysis are criminal codes issued in 1950, 1961 and 2009, including relevant amendments. The aim of the research is (by use the in-depth textual analysis and critical gender position) to highlight the historical changeability of the discourse of sexuality and to uncover related meanings in all texts used for the analysis. I also focus on the specific way, how it comes to the social construction of gender and thus which representation of masculinity and femininity is possible to see in the criminal law related to the defined criminal acts related to the sexuality. Criminal codes are understood as the official, universally applicable documents. How the czech criminal law contributes to the social control of sexuality? The most imporant keywords are: gender, discourse, language, criminal law, sexuality.

Řezáčová, MD., Ph.D., Institute of Organic Chemistry and Biochemistry, Academy of Sciences of the Czech Republic Thesis Title: Study of some markers of human leukemia Abstract: Immunophenotyping of leukemia cells is an important part of leukemia diagnosis. Immunophenotype is determined by flow cytometry using monoclonal antibodies against antigens expressed by these cells. Antigen CD44 is one of many CD markers used in immunophenotyping of leukemias. Protein crystallography of a complex between CD44 antigen and its ligand, hyaluronate, bring more detailed information about the structure of the binding site, which may help develop strategies for influencing the binding and thus for potential therapeutic intervention.
Thermodynamic Stability of Thaumasite

This work deals with the resolution of the identification characteristics of the thaumasit, which falls into the same group of AFt phases as the mineral ettringit. Both of these stages have the same structural arrangement and the morphology. This work’s specific goal is the synthetic preparation of thaumasit in a variety of conditions and its evaluation.
Motivation and Stabilization of Key Employees in Ceska zbrojovka a.s.

This master thesis focuses on the problem of motivation, stimulation and stabilization of the key employees in the company Česká zbrojovka a.s. The main aim of this thesis is to identify insufficiencies in the current system of employee motivation and to suggest potential changes leading to its improvement and assessment from the economic impact of the proposed improvements to the company. The theoretical part of the thesis contains the fundamental knowledge and base of the human resources management and also discusses various theories of work motivation. Reviewed problems of demotivation and unwanted employee turnover follow. The practical part of the thesis shows a qualitative analysis of the current system of the staff motivation in Česká zbrojovka a.s. There is also linked and combined theoretical knowledge with the corporate practice. The final part of the thesis is based on facts found from the survey which identifies imperfections in the existing system and formulates potential recommendations for improving employee motivation and stabilization in the analysed company in order to improve work performance, enhance job satisfaction and loyalty of employees to Česká zbrojovka a.s.
