Acanthamoeba: A Threat to the Human Eye
Veselá, Kateřina ; Říhová, Dagmar (advisor) ; Škodová, Jana (referee)
This thesis examines unicellular microorganism of the genus Acanthamoeba, as well as the disease it causes - acanthamoeba keratitis. It is an eye disease, which occurs mainly in people using contact lenses. Since acanthamoeba attacks visual apparatus, several parts of the thesis are dedicated to the anatomy of said apparatus, especially to the eye ball. The symptoms, diagnostics, treatment as well as prevention of the disease are included as well. The thesis furthemore mentions individual types of contact lenses, their proper hygiene and care and most of all, the danger, that comes with improper handling - acanthamoeba keratitis. A survey, that took place among elementary school and high school students, found out, that almost none of the elementary school students wear contact lenses, whereas contact lenses are worn by approximately 15% of students at high school. Among other issues, the survey concludes, that only a small part of elementary school students prefers contact lenses to glasses, though glasses are needed by a vast majority of them. At high school level, wearing contact lenses becomes more common, for aesthetics or other reasons. Ophtalmologist is regularly visited by 35 % of interviewed students. An analysis of six different natural science and biology textbooks for the 8th grade of...
Preimplantation genetic haplotyping in genetically risk families
Borgulová, Irena ; Macek, Milan (advisor) ; Trávník, Pavel (referee) ; Veselá, Kateřina (referee)
PREIMPLANTATION GENETIC HAPLOTYPING IN GENETICALLY RISK FAMILIES Abstract of Irena Borgulova's PhD study Page 1/1 ABSTRACT Preimplantation genetic diagnosis (PGD) is at the intersection of assisted reproduction and clinical genetics. PGD precedes prenatal diagnosis because consists in biopsy of a single embryonic cell and its examination excluding genetic risks before embryo transfer back to mother uterus. Methods within PGD can offer all spectrums of possible investigations of a single cell, whether focused on monogenic disorders, chromosomal aberration or abnormality of whole genome. Monogenic diseases in embryos can be detected by direct or indirect linkage analysis. Indirect linkage analysis has the advantage compared to direct analysis that it is able to indentify pertinent aberration of examined chromosome. Indirect linkage analysis is characterised by preimplantation genetic haplotyping (PGH) which is prime and important constituent of PGD cycle. PGH is based on family anamnesis for determination of pathologic/ high-risk (mutation-associated) haplotype and healthy/ low-risk (without mutation) haplotype by comparison with the haplotypes of other family members. PGD cycle requires in vitro fertilisation (IVF). IVF cycle includes hormonal stimulation, biopsy of oocytes and their fertilisation outside...
The Land Snail Velocity
Veselá, Kateřina ; Říhová, Dagmar (advisor) ; Loužecká, Ivana (referee)
This bachelor thesis deals with the usage of Garden Banded Snail (Cepaea hortensis) and Grove Snail (C. nemoralis) in teaching; especially in speed measurement. It is also addressed to laboratory breeding of this genus and basic requirements of land snails during keeping in order to provide specimens suitable for teaching as well as speed measurements. Despite of the compliance with the requirements for successful breeding, some specimens created epiphragm (temporary lid built during unfavourable conditions). Part of the thesis is addressed to this issue: why snails build epiphragm and how to activate them. The speed measurement itself is approached from the two different point of views: how to adapt the environment for measurement and how to perform the measurement in theoretical concept; and how to perform the measurements itself in a school class or in home breeding. The thesis uses responses of the students, who performed the experiment at school and shared the information about their feelings from working with the snails. Those feelings of disgust or even fear are discussed in a separate chapter. Apart from usage of land snails for the subject of Biology, this thesis also elaborates their inclusion into several other subjects.

Issues of elastic bandage in nursing practice
VESELÁ, Kateřina
The compression therapy is often being underestimated not only by medical staff and doctors, but also by patients. Therefore, it is often perceived as a simple task frequently handed over to hospital attendants. At the nursing schools the time devoted to the issue of compression therapy is minimal. That is probably why there are nurses with a very limited information at their disposal regarding this issue. The compression therapy is nevertheless an essential part of prevention and treatment of the venous and lymphatic diseases. When the compressive bandaging is carried out efficiently, it can improve the quality of life of a patient with a venous or lymphatic disease. Moreover, it can help in preventing thromboembolic diseases during surgical interventions. However, in order for the compressive bandage to be effective, several conditions must be met. First, it has to be carried out by a properly trained medical staff. Second, it is highly important that the patient undergoes a thorough education concerning compressive bandaging, so that he or she is able to keep the bandage effective when released home. Based on evidence, the aim of this thesis is to find out how effective is the practice of compressive bandaging within nursing care. The thesis consists of two parts. The first is theoretical and it contains the findings of various researches and publications related to the issue of venous diseases, aplication of the compressive bandage, differences between elastic stockings and elastic roller bandage and the findings concerning the education of a patient. The second part is practical. It consists of a quantitative research with a participation of 192 respondents who work as nurses in medical facilities and who are therefore closest to the patient undergoing compressive therapy. The thesis is interested in the amount of knowledge nurses have in relation to the compressive bandage, in the way they apply the bandage and in the process of the education of the patient. The research shows that the choice of a type of a compressive therapy is conditioned by the supply of the ward. Furthermore, the research reveals that there is no connection between the length of the professional experience of a nurse and her or his method of applying the compressive bandage. The recommendation for practice is an educational seminar for nurses aimed at completing their theoretical knowledge. This would also serve as an opportunity to practice not only the application of the bandage, but also the use of a measuring instrument Picopress.
Congenital disorders of glycosylation: methods of glycans analysis
Bittenglová, Kateřina ; Hansíková, Hana (advisor) ; Veselá, Kateřina (referee)
Congenital disorders of glycosylation represent (CDG) a group with more than 100 types of metabolic disorders, which are caused by defects in biosynthesis and modification of glycoconjugates. CDG manifest by broad spectrum of clinical symptoms, from disorders of nervous system to disorders of digestion and excretion. Identification of the specific type of CDG is not possible without broad spectrum of biochemical and molecular-genetic methods. The goal of this bachelor thesis was to describe the most often used methods for analysis of glycans, which are components of glycoproteins or glycolipids, in the theoretic part. Isoelectric focusing of selected blood serum glycoproteins, (e.g. transferrin and apolipoprotein C-III) serve as screening methods. Measurement of enzymatic activity, mass spectroscopy, PDO and LLO analysis (protein derived oligosaccharide,lipid-links oligosaccharide) HPLC and CZE (capillary zone electrophoresis and high performance liquid chromagraphy) are performed in second level. Molecular-genetic methods are used to confirm the final diagnosis by identification of causal mutations in specific gene. The aim of practical part of this bachelor thesis was to analyse the expression of genes OGA (N-acetylglukosaminase) and OGT (N-acetylglukosaminyltransferase). This enzymes take part...
