Recombinant expression of the mouse NK cell receptor NKR-P1C

NK cells are at the forefront of the body's defense against various threats, such as viruses or cancer cells. NK cells can recognize and destroy harmful cells in several ways that are very different from the defense mechanisms of, for example, T- or B-lymphocytes. NK cells carry a wide range of activating and inhibitory receptors on their surface that distinguish healthy cells from harmful or infected cells. NKR-P1 (natural killer receptor-protein 1) is the first discovered family of NK cell receptors similar to C-type lectins, which includes both activating and inhibitory receptors. NKR-P1C is an activating receptor capable of forming homodimers on the surface of NK cells. The major ligands of the NKR-P1 receptor family are the immunoenvasins of murine cytomegalovirus and other molecules structurally distinct from MHC class I glycoproteins. This work focuses on the recombinant expression of the NKR-P1C receptor protein. Using the pUT7 plasmid, we prepared a stably transfected HEK293T cell line carrying the gene for expression of the extracellular domain of NKR-P1C fused to the Fc fragment of IgG using the PiggiyBac system, and between them, they carry the TEV protease cleavage site. Drama as the Prevention of Bulying

The diploma thesis deals with the method of structured drama and its help in the prevention of bullying. Specifically, the aim is to understand the role of a silent witness as a person who can prevent bullying by stepping out of this role. The theoretical part defines the term bullying, describes the forms, causes and characteristics of bullying among pupils of the first grade of the primary school. This part of the thesis also gives a definition of the chosen method of structured drama, its procedures and techniques. The practical part deals with the created structured drama and its reflection. The questionnaire survey then examines the attitude shift of selected pupils in their decision to step out of the role of the silent witness. The period immediately preceding the realization of structured drama and the period after a few months are the initial and terminal phases of the examined attitude shift. TV Reporter and Publicist Zdeněk Velíšek

The master thesis Television Reporter and Publicist Zdeněk Velíšek describes the personal life and professional activities of the Czech journalist, publicist, interpreter and translator Zdeněk Velíšek. The text is arranged chronologically according to individual time periods from his birth in 1933 and childhood, through his university studies, translation and interpreting work, to his work at Czech Television until 2019. In each chapter you can find a foreword describing political, social and historical context, followed by an outline of the development of television in Czechoslovakia. Both contexts influenced Velíšek's life. Detailed personal interviews with Zdeněk Velíšek, work with the archived materials from Czech Television and Czech Radio, as well as Velíšek's books of commentaries and translations were important sources for this thesis. The main focus is Velíšek's work at Czechoslovak (later Czech) Television since the 1990s, which describes his work as a reporter, commentator, presenter, dramaturge and author of documentaries. The television work is followed by a section about cooperation with Czech Radio. Set of model activities of pre-vocational rehabilitation for patients with disability

OF THE BACHELOR THESIS Set of model activities of pre-vocational rehabilitation for patients with disability. Abstract: The thesis focuses on creating a set of model activities for patients with disability. This set was designed for practical use in the form of manual. The manual consists of seven model activities: manual work with tools, office work, activities with money, folding laundry, computer work, making a toast and washing up. I applied the set on six patients with disability and I evaluated their occupational potentional. The thesis is divided into two parts: theoretical and practical. The first part deals with the topic of the disabled, what occupation means to a person with disability and it probes into a problem with employment of the patients with disability. In the second part, I present the methodology and procedure I used in the set application on a sample of healthy population and on patients with disability. I state my observations and experience I have gained from the application of the set. It is followed by the discussion of achieved results. The final part of the thesis offers an answer to a question if it is possible to apply the set of model activities on patients with disability in order to help them find an appropriate job. Revitalization of the chateau park in Choltice

This work has the task to assess the current situation of the chateau park and at the same time the game preserve in Choltice, and suggest its revitalization in order to rejuvenate the vegetation and making the field available for the general public. Choltice is a market town in the Pardubice region, approximately 15 km west from Pardubice. At the end of 17. century a baroque castle was built here, which belonged to the count Simon Thun. His family belonged it until 1945. The chateau and the park is now owned by the borough Choltice. The chateau neighbours with the extensive English park, which passes the chateau game preserve, which is since 1992 a nature reserve and an important European locality. The diploma thesis contains a detailed theoretical basis and proposes solutions, which are directed both on the protection of the living conditions of protected and endangered species of a beetle Osmoderma eremita. This species occurs only rarely and his survival area is very limited, therefore it is necessary to create those conditions, and it is currently one of the main tasks of the revitalization. The next step is to help revitalize the introduction of spontaneous breeding fallow deer in the game preserve. This idea is not new, the fallow deer were bred there until 1945. The town of Choltice is very inclined towards this idea about farming and supports it. To increase the number of visitors this diploma thesis contains a proposal for the creation of a nature trail, this area would attract wider public than so far.

In my work I deal with the selection of suitable methods for reducing the alcohol content in the vineyard or later in winemaking. With properly chosen method the wine should show a lower alcohol content, without any impact on wine sensory quality.
Nutrition of patients on surgical departments

The bachelor work called the Nutrition of Patients at Surgical Wards occupies with nutrition of surgical patiens, their energetic nutrition intake and the intake of the basic nutrients during their stay in hospital, ie proteins, fats and carbohydrates. The aim of the work is to find out how many calories on average the patiens at the surgical ward consumed per day. The theoretical section is divided into two parts. In the first part there are described the basic components of food and their recommended daily intake. The second part deals with the problem of various illnesses typical at surgical wards. This part also deals with diets that are often indicated at the area of surgery. For realization of the practical part there was used the method of mixed research. The facts were gained with the help of observation and semi structured interviews. The research team was created by men and women of various age categories who were healed at the surgical ward in Pribram hospital in August 2014. For three days it was recorded how much food the patiens consumed. Then, they were interviewed. The effort was to find out the reason why the patiens had not eaten the meal or if they had not consumed their own food. On the basis of the collected facts there was made a graph (frequency of the consumed hospital food), there was made each patient´s menu using the programme Nutriservis, comparison of the food energy that the patients were given during these free days and that they really ate. Energetic values of hospital food were compared just roughly with energetic needs of an average woman and an average man. To sum up this research, it was realized that only one patient during this period was rating nutritionally rich food. Other 17 patients were not eating enough. On average, the patiens were eating 60% of recommended energy during this research. One of the reasons was the type of the respondents. The patients at the surgical ward had serious healthy problems, they did not feel well and thatsway they ate less than usually. Some patiens did not have a good appetite or they did not like the hospital meals. The next reason were the planned medical examinations and nutritionally poor food given the patiens by the hospital.

The thesis "Attribute in Spanish language" is aimed at the analysis of the word patterns that are used to build an attribute. The target of the thesis is to define attributes as a part of a sentence and describe every particular kind of them. This analysis is based on the study of linguistic materials coming from both, Czech and Spanish sources. First, the theoretical part will be focused on the unification of the terminology. It will also be attempted to connect these two very different language systems in one. Further, the attribute will be described in detail from various formal points of view. In the practical part the findins from the theoretical part will be applied to analyze some text taken from Spanish periodicals. The results of analysis will be stated in the third part of the thesis, conclusion. The graphical illustration of the chosen results is would also be presented in the conclusion. Résumé in Spanish is also part of this thesis.
Therapy with corset in patients with scoliosis

For my bachelor thesis I selected the topic "Brace Treatment for Scoliosis Patients". Scoliosis is defined as a three-dimensional deformity of the spine with side curvatures in the frontal, sagittal and transverse planes at the same time. A scoliotic curvature is accompanied by deformations of vertebras, changes in ribs and spatulas and changes in the position of crista iliaca. One of the conservative scoliosis treatment options is brace treatment. Brace treatment aims at generating a positive effect on the curvature and, most of all, at preventing its further development in the period of an individual's growth. Brace treatment is a controversial method to a certain extent, considering a large number of diverse opinions of doctors, orthopaedists and physiotherapists concerning the treatment with torso orthoses. The theoretical part summarizes the findings relating to scoliosis, including anatomical structures influenced in scoliosis. It contains information on basic terminology, scoliosis classification, diagnostics and treatment options. The thesis is focused particularly on conservative treatments but also provides information on possible surgical treatments. Introduction into the issue of brace treatment is addressed in more detail. The history, kinds of braces, manufacture, and the principles of the effect of torso orthoses are outlined there. The practical part of the thesis includes objectives, description of the method, characteristics of the set and particularly the processed results presented in the form of case histories. The data were obtained in a qualitative research. The research involved two patients suffering from idiopathic scoliosis with indicated brace treatment in the Rehabilitation Department of the Hospital of České Budějovice. The girls were selected on the basis of their diagnosis and brace indication. The medical history of the probands was gained and the initial and final kinesiological analyses were performed. The analyses consisted of a visual static and dynamical examination, palpation examination, evaluation with a plumb line, and examination of hypermobility, gait and breathing mechanism. Both patients had the length of their lower limbs measured. The three-month research was aimed at creating a physiotherapeutic procedure for individual patients and outlining the effect of brace treatment on the overall effectiveness of the treatment. The research has confirmed the necessity of an individual approach to each patient suffering from scoliosis. Seemingly similar curvatures may show a completely different development and require a different therapeutic approach. It follows from the research that the best results may be achieved in patients if a combination of several various methods is used. Particularly auto-correction in front of a mirror, Vojta's reflex locomotion and de-rotational breathing proved successful in my research. Auto-correction in front of a mirror before and after the treatment was found particularly beneficial because the patients themselves could observe the changes of their posture during the treatment. In my experience, the use of Vojta's reflex locomotion always served as an excellent preparation for the following exercises because the abdominal wall, autochthonous muscles and ribs become activated and the breath wave is rearranged. Since both patients suffer from scoliosis with a considerable rotation element, the application of de-rotational breathing showed to be suitable because it brought visible results in each treatment for each patient. The brace treatment influences the effectiveness of the entire treatment particularly by not allowing the patient to keep a pathological posture all day long. This fact makes the work with a patient easier and has an effect on the patient's auto-correction. The bachelor thesis may be used in clinical practice of physiotherapists. The thesis may also be used as an educational material for patients.
