Effect of mitochondrially targeted senolytic agents on the development of respiratory diseases
Rennerová, Michaela ; Štemberková-Hubáčková, Soňa (advisor) ; Neckář, Jan (referee)
Aging is a complex biological process characterized by gradual decline in the physiological functions of cells and tissues, leading to the activation of a process known as cellular senescence. This results in the development of structural and functional changes in the organism accompanied by an increased risk of various diseases. Respiratory diseases are among the most common chronic conditions associated with the accumulation of senescent cells in the body. Current treatment for chronic respiratory diseases primarily focuses on symptom relief rather than addressing the primary cause of these conditions. Given that senescent cells play a significant role in the pathogenesis of respiratory diseases, the future of therapies lies in their elimination using senolytic agents. Despite promising results from some studies, current research is limited by the heterogeneity of senescent cells, which is reflected in their sensitivity to senolytic agents and the variability of therapeutic responses. Mitochondria play a central role in the development and maintenance of the senescent cell phenotype by regulating key processes such as energy metabolism and cellular signaling, regardless of the type of senescent cell. Thus, targeting mitochondria in senescent cells may represent a promising therapeutic strategy...
The role of glutamine in leukemia cell metabolism
Sitdikova, Karina ; Hložková, Kateřina (advisor) ; Zelenka, Jaroslav (referee)
Leukemia is a heterogeneous group of hematological malignancies that result from the abnormal proliferation of immature blood cells. One of the hallmarks of tumor cells is their altered metabolism. Therefore, therapy targeting deregulated metabolic processes is an attractive strategy for the treatment of malignancies, including hematological ones. Amino acid metabolism is an important part of cellular metabolism, and targeting it appears to be a key attractive strategy in the treatment of leukemia. Glutamine, a conditionally essential amino acid, plays a crucial role in energy metabolism and maintaing the redox balance of leukemia cells, thereby contributing to their growth and proliferation. Strategies to treat leukemia by targeting glutamine metabolism include glutamine depletion, the use of glutamine transporter inhibitors and glutaminase enzyme inhibitors. To ensure the effectiveness of leukemia treatment, it is essential to recognize that glutamine is involved in numerous metabolic pathways, each of which is regulated by multiple factors. As a result, therapies targeting glutamine metabolism should be carefully designed to avoid affecting healthy cells and patient immunity. This thesis describes leukemia, including its types and treatments, and glutamine metabolism and its potential targeting...

Leszkow, David ; Pavlů, Dagmar (advisor) ; Reckziegelová, Petra (referee)
Title: Differences in Approaches to Prevention and Therapy for Female and Male Football Players with ACL Injuries - A Systematic Review Objective: The main aim of this thesis is to investigate whether there are different approaches to the prevention and treatment of ACL injuries in male and female football players. Methods: The thesis is prepared as a theoretical text in the form of a systematic literature search using PubMed, Web of Science and PEDro databases. The first part of the thesis contains the current status of the studied issue, theoretical background and treatment options for ACL injuries. The second part is descriptive-analytical in nature. It contains an analysis of the studies included in the thesis. Results: After the search, 4 studies met the criteria. Each of them focused on injury prevention, but unfortunately none of them investigated physiotherapy as such. However, the evaluation of the studies shows that both men and women use the same prevention programmes in football, which can reduce the risk of injury by up to 50 %. A limitation of the search is that most of the studies used did not examine the effect of prevention on both sexes simultaneously. Keywords: ACL, LCA, přední zkřížený vaz, ruptura, plastika, prevence, poranění, fyzioterapie, rehabilitace, postup, ženy, muži,...

Rohatec Special Regime Home
Čadová, Veronika ; Vostrejž, Dušan (referee) ; Boleslavská, Yvona (advisor)
The subject of the master’s thesis was to design an architectural study of a new building for a home with a special regime in Rohatec. This request was made in response to the tornado-destroyed building of the S-center in Hodonín. The request required the design of a multi-functional facility with a private garden for 90 patients and 100 staff working over two shifts. The selected land is located at the end of Rohatec, mostly surrounded by forest and a few family houses. At the same time, it‘s located by the main road, so it‘s easily accessible by everyone. The building is designed as a 3 story building with a flat roof and underground parking in the shape of an open U. The shape of the building itself is a compromise between the shape of the land, layout requirements for this type of facility, capacity requirements, and height restrictions according to the master plan. The shape allows for the creation of a private residential garden for the residents of the home, which is closed from the road, with a possible gate connection to the future forest park. The passage between the garden and the building will be enabled by a terrace around the entire perimeter of the inner side of the building. The individual home wings also enable layout separation of the individual departments needed for smooth building operations. Dispositionally, the home has a pharmacy open to the public, an underground parking lot, inpatient and administrative departments, a kitchen with a dining room, a cafe with a buffet, a library, a chapel, a part designated for therapy and space for mani-pedi with a hairdresser, rehabilitation with a gym, and more. The necessary technical background for the operation of the building, workshop spaces, a laundry room with a dryer, an ironing room, and storage rooms are also designed here.
