Preparation and phytoextraction of 125-I labelled pharmaceuticals

Pharmaceuticals are group of organic substances with significant worldwide consumption in human and veterinary medicine. These compounds may be metabolized in the organism, but in some cases they remain unchanged and both are usually excreted via renal excretion in the native form or as metabolites. Large quantities of pharmaceuticals and their metabolites contaminate municipal wastewater. The wastewater treatment plants are unable to remove these substances completely, so they contaminate surface water, groundwater and soil as well. Due to the biological activity of pharmaceuticals, long - term effect may cause bacterial resistance, endocrine influence, DNA and renal damages in non-target organisms. The phytoextraction and the translocation of radiolabeled diclofenac with 125 I were experimentally studied by using of in vitro cultivated plants Helianthus annuus and Zea mays. Efficiency od phytoextraction was monitored as decrease of radioactivity of tested substance [125 I]diclofenac in Murashige-Skoog cultivation medium. Both species are able to extract tested substance during 8 to 10 days of cultivation, with efficiency approximately 85 % using Zea mays and 79 % using Helianthus annuus. The proteomic study of abiotic stress of plants.

Reason of this trend is ever-increasing number of diseases, lifestyle and still increasing demand for the drugs. With this trend growing interest in the analysis of the residues of pharmaceuticals in the environment which is result of incomplete wastewater treatment. This diploma thesis is studying effect of cytostatic drugs, specifically doxorubicin and one of the most widely used analgesics - ibuprofen, at the proteome level of the model plant Arabidopsis thaliana. Proteins isolated from plants exposed to the drugs were separated by two-dimensional electrophoresis. Comparing of protein maps by PDQest program (Bio-Rad, USA) was found several proteins whose expression was affected by the presence of drugs in the culture medium. Selected proteins were identified by LC - MS / MS.
Study of plant stress responces in presence of pharmaceuticals in cultivation medium

The aim of this study was to verify the possibility of ibuprofen degradation by selected plant cultures and determination of activities of antioxidant enzymes (peroxidase, catalase, ascorbate peroxidase and glutathione-S-transferase) as markers of oxidative stress caused by ibuprofen. Nicotiana tabaccum (cv. La Burley 21, cv. SR 1 and their GMOs) and Nicotiana glauca were used as experimental plants. The rate of removal of ibuprofen tested by tobacco was decreasing in the following order: N. tabaccum SR1 > N. tabaccum Zm-P60-1-T4 > N. tabaccum TRI 2T2 > N. glauca > N. tabaccum TRI 2T1 > N. tabaccum cv. La Burley > N. tabaccum Zm-P60-1-T5. As the most suitable tobacco for the removal of ibuprofen seemed untransformed N. tabaccum SR1. The long-term experiment showed that plant stress is being manifested even after longtime. N. tabaccum cv. La Burley 21 seemed to be the most tolerant to ibuprofen in compare with the total enzyme activities in cultures with the presence of ibuprofen and controls. N.glauca was the least tolerant cultivar. Use of Fenton Reaction for Drug Degradation in the Ecosystem

The elimination of unwanted substances from the ecosystem can be accomplished by advanced oxidation processes (AOP). AOPs are used preferably in treatment of wastewater. One of the AOP's methods is Fenton's reaction. The degradation of drug residues by this reaction was the aim the project. The experiments were focused on finding possible degradation products, which could be much more hazardous than parent compounds. The study was primarily concerned with non-steroidal anti-inflammatory drugs (acetaminophen, diclofenac, ibuprofen, naproxen and salicylic acid) and antiepileptic carbamazepine. The complete decomposition of tested compounds was not observed by using HPLC and MS analysis. The products of decomposition were found in reaction mixture, in case of diclofenac dechloration of aromatic ring occurred. In cases of most compounds with exception of acetoaminophen and salicylic acid the reaction led to the formation of more polar products than parent compounds. The results imply that even more advanced oxidation processes do not cause complete destruction of organic compounds. Ecotoxicological Evaluation of Polymers and Biologically Active Substances in Aquatic Environments

To determine the ecotoxicity of analgetics, first the individual ecotoxicity values of individual analgetics are determined and then a mixture of two analgetics is tested. To determine the toxicity, both standard and alternative toxicity tests are used (daphnia magna, sinapsis alba, scenedesmus subspicatum, vibrio fischeri, thamnotoxkit FTM a daphnotoxkit FTM magna). The analgetics being whish tested are the commonly used medicines ibuprofen, ASA, diclofenac and paracetamol, which are the most frequently used medicines in the Czech Republic and whole Europe. To determine the ecotoxicity of the polymers, I‘m using an indirect method of examination, in which I determine the antagonistic or synergistic effects of the mixture of monomers from which the polymer is prepared and into which it slowly decomposes in nature. For the determination both standard and alternative toxicity tests are used. The polymers the toxicity of which is being determined are the habitually used polymer PET and the formaldehyde resine known as bakelite in Eastern Europe.
Chiral analysis of drug residuals in waste waters

The theoretical part shortly describes chirality with focus on chiral pharmaceuticals. The processes of their absorption, distribution, metabolism and elimination in human body are discussed. These points are very important to understand possible fate of chiral drugs in the environment as there is only little data concerning their environmental behaviour. The occurrence and enantioselective toxicity of chiral drugs is also discussed here. One of the chapters describes non-steroidal anti-inflammatory drugs, as they are analyzed in the wastewater in the experimental part, and their occurrence in the environment. The experimental part describes optimization of the enantioselective HPLC method using Chiralpak AD as column for ibuprofen and ketoprofen. Reproducible separation of enantiomers wasn’t achieved for naproxen. Optimized methods were then applied for analysis of samples from municipal wastewater treatment plant in Brno-Modřice.
|Preparation and characterization of micro- and nanoparticles with selected drugs

The presented bachelor thesis is focused on the way to prepare combined micro- and nanoparticles containing some natural extracts with antioxidant effects, ibuprofen as the anti-inflammatory and analgetic agent and clotrimazole as the agent with an antifungal effect. The theoretical part contains an introduction of the methods of liposome preparation and a review of encapsulation techniques, methods of particles characterization, and analysis of active substances content. Furthermore, the yeasts of the genus Candida and the methods of the antimicrobial activity determination are described. The mentioned components were encapsulated into liposomes, alginate and chitosan particles. The antioxidant activity of the extracts was determined spectrophotometrically. The encapsulation effectivity of active substances into particles and the long-term stability of encapsulated components were determined and compared too. The HPLC method was used for measuring of the concentration of drugs and spectrophotometric method was used to quantify the content of polyphenols. Prepared particles were characterized with DLS method (size) and zeta- potential (stability). Finally, antibacterial properties of prepared particles and extracts using the test system Candida glabrata were studied. The antimycotic effect of particles, disturbed liposomes and extracts was compared. Finally, potential utilization of prepared liposomes in a cosmetic or pharmaceutical product is proposed.
Evaluation of ecotoxicity of selected chemicals with using algal test.

This bachelor thesis deals with the ecotoxicity of 2 - [(2,6-dichlorophenyl)aniline] benzene acetic acid (diclofenac), ?-methyl-4-(2-methylpropyl) benzene acetic acid (ibuprofen) and mixture of cadmium and diclofenac and studies the above using algal toxicity test in accordance with ISO 8692. The theoretical part describes the issue of pharmaceuticals in the environment, and characteristics of standards algae tests and alternative tests. Freshwater green algae Desmodesmus subspicatus were used as testing organisms in all the implemented algal toxicity tests. By the means of these tests, the IrC50 values for the standard substance potassium dichromate, the drugs diclofenac and ibuprofen and cadmium were determined. The toxicity of cadmium in mixture with diclofenac was also evaluated.
