Patrik Zmij Supervisor: RNDr. Ondřej Souček, Ph.D. Title of diploma thesis: Lymphocyte reconstitution after allogeneic hematopoietic stem cell transplantation Allogeneic hematopoietic stem cell transplantation is the most effective treatment option for patients with hematologic malignancies, including acute myeloid leukemia, myelodysplastic syndrome, or the rarer myelosarcoma. After a successful transplantation, the immune system cells are regenerated, and this master's thesis focuses on one aspect of that process: lymphocyte reconstitution. The experimental part of the study involved patients who underwent allogeneic transplantation of blood- forming cells based on their unfavorable diagnosis. These patients were then monitored for one year at the Institute of Clinical Immunology and Allergology at the Faculty Hospital in Hradec Králové. Observations were conducted at three months and continued until the end of the first year, using flow cytometry as the method of assessment. Immunophenotype is determined by flow cytometry using monoclonal antibodies against antigens expressed by these cells. Antigen CD44 is one of many CD markers used in immunophenotyping of leukemias. Protein crystallography of a complex between CD44 antigen and its ligand, hyaluronate, bring more detailed information about the structure of the binding site, which may help develop strategies for influencing the binding and thus for potential therapeutic intervention.
Eight color flow cytometry test development for primary imunodeficiency patients

Primary immunodeficiencies represent a heterogeneous group of hereditary immune system malfunctions with very variable causes and symptoms. Multiparametric flow cytometry has become an important tool in primary immunodeficiency diagnostics and research because it provides detailed information on the phenotype of individual immune cells and their proportions in circulation. We have developed a complex monoclonal antibody panel composed of five eight-color tubes which is designed for immunophenotyping of basic lymphocyte subsets and further analysis of B and T cell subpopulations. We have optimized and standardized the panels so they will identify any changes originating from primary immunodeficiencies and provide comparable data on the level of cooperation between more laboratories. This was achieved by cooperation of six European research facilities which are all parts of the Euroflow consortium. The panels have been validated both on peripheral blood samples from healthy donors and patients with either gentically defined primary immunodeficiency or common variable immunodeficiency. Novel antibody array validation and application to determine new prognostic markers in childhood acute leukemia

v anglickém jazyce Leukemia is the most common malignant childhood disease, which occurs in Czech Republic every year in about 100 cases. That's why has been devoted over the last 5 decades much attention and funding to this research and the children with leukemia have a high chance for achieving complete remission. Nevertheless, for about 10% of patients leukemia still remains lethal and these cases are subjects of current studies. Leukemia is a complex disease in its pathology involving lots of aberrations, which pathologically manifest at the DNA, mRNA and protein level. Nevertheless some symptoms of this disease are only at the protein grade (and not DNA or mRNA). Since a broad part of the protein profile in leukemic cell changes, it is necessary to develop newer, more sensitive and especially more efficient methods, which are able to follow up changes in large sets of proteins and detect prognostic and diagnostic markers. Within diploma study a new method - multiplex antibody array - has been tested. Our laboratory tests this method in collaboration with a Norwegian laboratory (Rikshospitalet University Hospital, University of Oslo) under leading by Dr. Lund-Johansen, who is the author of this method.
