Utilization of advanced techniques of electron microscopy in altrastructural analysis of microbial cells and microbial biotechnology products

In the theoretical part, this thesis deals with direct and indirect mechanisms of plant growth-promoting rhizobacteria, which is followed by a section on the selected model organism Azotobacter vinelandii. The second part of the theoretical section deals with electron microscopy starting with the possible interactions of the electron beam with the sample, through types of electron microscopes to various methods of preparing biological samples. The experimental part discusses the cultivation of selected bacterial cultures, the gelation of the cell cultures using chosen cross-linking agents (calcium chloride, malic acid, glucono--lactone with calcium carbonate) and, above all, various methods of preparation and analysis of pure and encapsulated cultures by the means of electron microscopy. In particular, cryogenic scanning electron microscopy and scanning transmission electron microscopy imaging were selected. Using both techniques, poly-3-hydroxybutyrate granules, which were confirmed to be elastic even at cryogenic temperatures, were observed inside the bacteria. Furthermore, it was found that cross-linking of extracellular alginate occurs even in samples without the addition of cross-linking agents. However, the hydrogel network present in these samples was significantly thinner than in samples with the addition of all cross-linking agents used. In this work, the density of the hydrogel network cross-linked with glucono--lactone was also compared. When using this agent with the addition of calcium carbonate, a significantly denser hydrogel network was observed in comparison with using the agent alone. Last but not least, when comparing the obtained results with the literature, it was found that the A. vinelandii bacteria form capsules, during the formation of the capsules the shape of the cells changes from an oval to a spherical bacterium and also the size of the given cell is reduced. From the resulting images, it was found that these capsules are made up of two layers, to which literature refers as electron-transparent intine and electron-dense exine.
The Project of Social Services of the Business Entity

This thesis deals with a project whose mission is to optimize the social services of the entity, so as to increase the motivation and productivity of employees. The project is focused on building the corporate childcare facilities for pre-school children of employees. The work also includes an analysis of the risks associated with the construction of facilities and fund the project from European funds.
Possibilities for Financing of the Public Building Order from the Aspect of Investor

The aim of Bachelor´s thesis is to point out the possibilities of financing public procurements on construction works, focusing on the public sector. The theoretical part explains the main concepts related to the issue based on the Novel "LPP" with explaining basic forms of Public Administration. The practical part deals with identifying an closer analysing of appropriate financing funds for a certain public construction procurement.
Preparation and characterization of quatsomes

This bachelor thesis deals with preparing and characterizing quatsomes and similar vesicular systems. The preparation consisted of weighing an equimolar amount of sterol and surfactant, hydration with deionized water, and subsequent sonication with an ultrasonic probe with an energy of 5, 10 and 15 kJ. In this work, two types of quatsomes were prepared, the first contained equimolar amounts of cholesterol and cetyltrimethylammonium bromide (CTAB), and the second consisted of cholesterol carbethopendecinium bromide (Septonex). The optimal dispersion energy of 10 kJ was selected for both types based on turbidity measurements, visual observation and dynamic and electrophoretic light scattering analysis. Furthermore, the observed properties were found to be constant from day 7 after sonication. In addition to quatsomes, similar vesicles were prepared, which contained a surfactant in the form of a sulfate salt. Once again, two types were prepared, the first containing sodium dodecyl sulfate (SDS) and cholesterol, the second sodium tetradecyl sulfate (STS) and cholesterol. Based on the same measurements as for quatsomes, optimal dispersion energy of 10 kJ was selected. Furthermore, mixtures with different ratios of cholesterol and a given surfactant were prepared, in which the effect of cholesterol addition was monitored. The analysis of these mixtures was performed by the same methods as the analysis of quatsomes.

This Diploma thesis describes the marketing strategy of Infinit Ltd. The author focuses on the Vysočany Pool and Sauna world branch, which is located at the Wellness Hotel Step. In the analytic part the author uses the following methods: situation analysis, bench-marking and SWOT analysis. Within the marketing strategy the author describes the characteristics of the marketing mix. The result of this thesis is to give a suggestion that would improve the current situation of this branch.
Destination Management of the microregion Telčsko

This Master theses deals with the most sophisticated form of tourism management -- Destination Management -- specificly in the area of microregion Telčsko. The aim is to analyze a potential of the selected destination, determine its current tourism management and propose possible development on the basis of findings and carried out research. The first chapter introduces basic concepts related to the main issue and concretizes the evolution of Destination Management in the Czech Republic. In the next chapter there is an inroduction of the microregion Telčsko. The heart of the work is an analysis of tourism potential of the selected area, survey, SWOT analysis and suggestions of development opportunities.
