Cardiotoxicity of anthracycline antineoplastic drugs and possibilities of pharmacological cardioprotection
Jirkovský, Eduard ; Štěrba, Martin (advisor) ; Kolář, František (referee) ; Hrdina, Radomír (referee)
- CARDIOTOXICITY OF ANTHRACYCLINE ANTINEOPLASTIC DRUGS AND POSSIBILITIES OF PHARMACOLOGICAL CARDIOPROTECTION Chronic anthracycline (ANT) cardiotoxicity is an important clinical problem which can have a substantial impact on morbidity and mortality of cancer survivors. Dexrazoxane (DEX) is the only agent with clearly evidenced cardioprotective effects in both experimental models and clinical trials. Despite intensive research, precise pathogenesis of chronic ANT cardiotoxicity and molecular mechanisms of cardioprotective effects of DEX remain unknown. Current clinical guidelines recommend not using DEX from the beginning of ANT therapy, but instead only from the cumulative ANT dose of 300 mg /m2. The aim of this work was to study functional, morphological and molecular changes associated with induction of chronic ANT cardiotoxicity and their further development in the post-exposure (follow up) period. Special attention was paid on the role of oxidative stress and possible response of protective antioxidant pathway regulated by Nrf2 as well as on mitochondrial impairment and response of mitochondrial biogenesis pathway. Chronic ANT cardiotoxicity was induced in rabbits by repeated intravenous injections of daunorubicin (DAU, 3 mg/kg, once weekly for 10 weeks). At the end of the treatment, the animals...
Analysis of the bidding process of Prague for Summer Olympic Games 2016
Bošková, Linda ; Dovalil, Josef (advisor) ; Kolář, František (referee)
Title: Analysis of the bidding process of Prague for Summer Olympic Games 2016 Objectives: The aim of this thesis is the analysis of Prague's bid for Summer Olympic Games 2016, the following determination of main causes of his failure and than a suggestion of the future steps leading towards the future organisation of Olympic Games. Methods: We have used three managerial methods in this thesis. It was at first the system approach that identifies all stakeholders and their relations, which are all connected. This method permits to understand our subject as a complete system. After that SWOT analysis of the bid and the Fishbone diagram was used to identify main causes of the bid's failure. Results: Main six causes were identified that have caused the failure of the Prague's bid. These causes are more a kind of internal serious problems that show that Prague is not ready yet for organising Olympic Games. Following suggestions for future steps are reacting on these results. The aim of that is to make a long-term strategy to improve the environment and relations between the sport and state and prepare good conditions in the country for the next Olympic bid. Developmental mechanisms of arrhythmias - role of connexins in arrhythmogenesis
Beneš, Jiří ; Sedmera, David (advisor) ; Kolář, František (referee) ; Král, Jiří (referee)
Developmental mechanisms of arrhythmias - role of connexins in arrhythmogenesis MUDr.Jiří Beneš Abstract: Objective: The aim of this study is an improvement of our knowledge concerning the role of connexins in arrhythmogenesis. The main focus is on the role of connexin40 (Cx40) in heart development in mice and changes in connexin43 expression in volume overload heart failure rat model. Methods: The infuence of Cx40 on heart develoment was studied on transgenic mouse Cx40:GFP model using the method of optical mapping. Volume overload heart failure was examined in the rats with aortocaval shunt. Morphological changes in hearts were examined using imunofluorescence microscopical techniques. Results: In the atria, Cx40 is important especially during the early stages. Cx43 can partially substitute its function from 12.5 embryonic day on. Cx40 deficiency leads to decreased conduction velocity and ectopic sites of activation. Absence of Cx40 in ventricular conduction system leads to the development of right bunde branch block. Volume overload in rats leads to excentric hypertrofy and later to heart failure. We described morphological as well as microscopical changes in failing hearts. Without the presence of fibrosis, the main arrhythmogenic substrate we found was reduced amount of Cx43 and its dephosphorylation....
