Dissecting the nuclear function of the interleukin-1alpha

Interleukin-1alpha (IL-1alpha) is a well-known proinflammatory mediator acting as a secreted molecule. However, in addition to its ability to activate its membrane-bound receptor, there is growing evidence on its noncanonical nuclear function, which classifies IL-1alpha as a "dual function cytokine". This nuclear action depends on the evolutionary conserved N-terminal domain of IL-1alpha. After proteolytic processing, the N-terminal domain of IL-1alpha translocates into nucleus. Histone acetyltransferase (HAT) complexes were previously identified as nuclear targets of IL-1alpha precursor. However, the specific protein which is responsible for the interaction between IL-1alpha and HAT complexes has not been identified yet. To dissect this interaction, the N-terminal domain of IL-1alpha was produced in yeast. Suitability of this experimental setup for testing the interaction between IL-1alpha and eukaryotic HAT complexes was evaluated in this study. Study of the mammalian oncogenic transcription factors in the yeast model

Yeast serves as a useful tool for studying cellular processes and therefore a large amount of techniques and protocols has been developed. There are special methods for studying the transcriptional factors in yeast, such as modified yeast two-hybrid screens, yeast one-hybrid screens and systems studying ability of transcription factors to transactivate a reporter gene. Oncogenes from AP-1 complex, Myc and Myb protein families are described in this work. Using a yeast model the structural-functional properties of proteins can be easily studied and in some cases even their ability of oncogenic transformation can be predicted (FASAY or ability of c-Myc to transactivate a reporter gene). However, results from yeast models must be confirmed in mammalian cells. 1
Perception of ecosystem services in an urban environment

This thesis deals with city ecosystem services and how are these services perceived by the visitors of the city nature areas. Specifically, the thesis deals with ecosystem services provided in the city Carlsbad. As the city Carlsbad is the spa city, great deal of visitors of the city nature areas are tourists and that is why are included in the research both city residents and also temporary visitors of the city. From the point of view of the usual division of the ecosystem services, as it was defined by Millennium ecosystem assessment (provisioning services, regulating services, cultural and supporting services), this thesis focuses mainly on the cultural and supporting ecosystem services, because this spectrum of services is presented in the greatest measure in cities in general and these services are overall the most appreciated in the urban areas. As a methodology was chosen a quantitative research, which was conducted by standardized questionnaire consisting of 19 questions. Methods of Reliability Analyses of Operation and Testing Data of Airplanes

The doctoral thesis deals with reliability (dependability) analyses of operation and testing data of the Airplanes. Requirements of airworthiness regulations on aircraft hydraulic systems (with a focus on US FAR-23 and European CS-23 regulations) are taken into account. Mentioned regulations include requirements for the structural design, design of systems, etc. They cover wide range of airplanes from small sport airplanes to 19-seats transport aircraft. Also options for predictive reliability analyses (resources) and reliability tests are discussed in the doctoral thesis. Practical application is done on small transport airplane (currently in the development). The failure report is designed. Expected major contribution of the work is selection and practical application of the most suitable procedures for safety assessment on the field of aircraft hydraulic systems, with a focus on the small transport aircraft. Also the comparison to different data source is shown.
Design of VUT200 TwinCobra landing gear

The goal of following master thesis is to design variety of configuration and retraction options of VUT200 TwinCobra landing gear. For each option are a wheel base and a gauge set up by possibility of main landing gear retraction. Next, CS 23 demands and stress analysis are followed. There is a view of twin engine aircraft landing gear disclosed as well.

Objective of this diploma thesis is to find suitable approach for Supplier Quality Management in the company operating small batch production. History and use of the helicopters in the Czech Republic

Final work concerned with rotocrafts which have ever been used on Czechoslovak region, further especially on region of Czech republic. The work focused on the oldest czech rotocraft aviation pioneers together with accentuating the importance of original czech helicopters. Whole review of helicopters are completed with Attachment 1, which shortly describes all types of used helicopters. Then the work gives in detail using rotocraft by institutions when was looked for historic affairs.
Evaluation of Robert Bosch spol. s.r.o.

Subject of the thesis is evaluation one of the plants of the biggest subcontractor of automotive components. The main goal of the thesis is to determine the enterprise value of Robert Bosch spol. s.r.o for the company management who wants to manage this value in terms of maximum fulfillment of the basic objectives of company owners. The value of the company refers to January 1. 2013. Evaluation is based on publicly available sources, mainly on informations from the annual reports. This data was consulted with employees of the company. In the theoretical part of the thesis, the process of evaluation is explained and some of the steps are examined in detail. In the practical part, introducing with the company and the business environment is made. Within the evaluation, financial and strategical analysis is prepared. The eavulation is based on yielding methods, method of discounted free cash flow and discounted economic value added.
