Comparison of different tandem mass detection scans in the analysis of cannabidiol

The aim of this bachelor thesis was to compare different tandem mass spectrometer scans in order to develop an LC-MS/MS method suitable for the determination of cannabidiol in rat serum. In this bachelor thesis the mass spectrometer and high-performance liquid chromatography method were optimized. The optimum parameters were as follows: chromatographic column Acquity UPLC BEH C18 50 × 2,1 mm, 1,7 μm from Waters (Wexford, Ireland). The mobile phase consisted of methanol and water of LC-MS purity. The flow rate of the mobile phase was 0,4 ml/min, the column temperature 40 řC, the temperature in an autosampler 15 řC, the sample injection volume was 1 μl and the analysis time was 7 minutes under gradient elution conditions. The MRM transitions of highest intensity were monitored. For CBD, the MRM transition in positive mode was 315,2 → 193,1 (Q1 = -10 V, CE = -24 V, Q3 = -20 V), for isotopically labeled cannabidiol D3 was chosen 318,4 → 196,0 (Q1 = -10 V, CE = -23 V, Q3 = -20 V). The calibrations in solvent and rat serum were performed in MRM and SIM mode. No calibration was constructed for the SCAN mode due to its low selectivity, which made it able to detect cannabidiol only from concentration of 312,5 ng/ml. Determination of bitrex using ultra-high performance liquid chromatography

In this work, a method for the determination of bitrex (denatonium benzoate) using ultra- high performance liquid chromatography was developed. A Kinetex C18 column (Phenomenex) with dimensions of 100×2.1 mm and a particle size of 1.7 μm was used for the determination. A mixture of acetonitrile with 0.1% formic acid (component A) and deionized water with 0.1% formic acid (component B) was used as the mobile phase. The best separation was achieved with the gradient program set: 0-2 min: 20 % → 70 % A (vol. %); 2- 3 min: 70 % A (vol. %); 3-3,5 min: 70 % → 20 % A (vol. %); 3,5-5 min: 20 % A (vol. %). The temperature of the column during the separation was 40 řC and flow rate was 0,3 ml/min. The developed method was partially validated. Repeatability was measured, the linearity of the method was verified for sample injection 1 μl and 5μl. For the same sample injection limit of detection and limit of quantification was determined. Using the developed method, seven real samples containing bitrex were measured. These samples differed in matrix. Determination of antiepileptics in breast milk with chromatographic methods

The present thesis is devoted to the development of a new method for the determination of a selected group of antiepileptic drugs (brivaracetam, eslicarbazepine, felbamate, gabapentin, lacosamide, perampanel, pregabalin, rufinamide) in human breast milk. The rationale for identifying antiepileptic drugs in this rather complex matrix is to assess and optimize the use of these drugs in breastfeeding mothers to avoid unnecessary and risky exposure of infants. The aforementioned drugs have various undesirable side effects that can have a negative impact not only on the mother but also on the breastfed infant. The protein precipitation method using acetonitrile/methanol mixture was used for sample preparation. A method based on liquid chromatography with tandem mass detection was developed and tested for the determination of the concentrations of these drugs. In the course of testing, different chromatographic columns with different stationary phases were tested to achieve optimal separation conditions and experimental set-ups for the full range of antiepileptic drugs tested.The whole methodology will be subsequently validated according to the general guidelines for clinical and toxicological method validation according to international standards and its use in routine laboratory practice will be intended.

This bachelor thesis is focused on performing in a band The Tap Tap and its value in the life of a person with a physical disability. The thesis is divided into theoretical and practical parts. The first chapter of the theoretical part is devoted to the definition of physical disability. It describes basic terminology, and division of movement disorders and introduces social-psychological aspects of the physical disability. The second chapter of the theoretical part is devoted to the social inclusion of people with a physical disability. It describes models of approaches to people with disability, and the impact of physical disability on family, partnership, sexuality, and employment. Further, the chapter describes social care and services and organizations and associations for people with a physical disability. The third chapter is devoted to the introduction of the band The Tap Tap, its history, and the present. The practical part is realised as qualitative research. For data collection was chosen the method of semi-structured interviews. The interviews were conducted with members of the band The Tap Tap with a physical disability. Electrokinetic chromatography in nonaqueous and mixed solvents: Determination of critical micellar concentration

This thesis deals with the investigation of micelle formation following the addition of a surfactant to a background electrolyte in micellar electrokinetic chromatography and the transfer of these systems to nonaqueous and mixed solvents. The addition of organic solvents to the background electrolyte has a significant effect onmicelle formationandhence separation in electrokinetic chromatographyusinga micellar pseudo-stationaryphase. This effect has not yet been studied in depth. Information on the presence and properties of micelles in a given separation system is important for the understanding of separation processes but also for the development and optimization of analytical methods. In this work, the critical micellar concentration (CMC) of two surfactants, sodium dodecyl sulfate (SDS) and cetyltrimethylammoniumbromide (CTAB), was determinedby introducingthem into a mixed environment with non-aqueous solvents, acetonitrile and methanol. Study of retention and separation of peptides in HPLC

This diploma thesis deals with the study of retention and separation of structurally different peptides and digest products of cytochrome c using reverse phase and mixed modes of liquid chromatography. As the model peptides, five unmodified dipeptides, nine dipeptides containing a protective benzoyl group on the N-terminal amino acid and eight therapeutic peptides containing five to ten amino acids were selected. The chromatographic behaviour of the model peptides was tested on three different columns: XBridge C18, XSelect CSH C18 and Atlantis Premier BEH C18 AX under different conditions. The influence of the pH of the aqueous part of the mobile phase, i.e. 2,1; 3,0; 4,7 and 6,8, and the amount of acetonitrile in the mobile phase on retention and separation of model peptides was tested. The effect of the concentration of ammonium acetate buffer, pH = 6,8 in the mobile phase on the retention and peak symmetry in the concentration range of 10-50 mmol L-1 was assessed. The chromatographic conditions for the baseline separation of 20 different model peptides were optimized. Comparison of the Frequency of Bullying in Upper-primary and Lower-secondary Levels of Schools.

The aim of this Bachelor thesis is to assess the frequency of bullying within the second level of primary schools and the lower grades of comprehensive schools. The first part of the work will primarily focus on theoretical explanation of bullying itself. It will introduce this behaviour along with its forms of occurrence in more detail. It will describe its development, step by step, and not only from the aggressor and the victim's points of view, but also from the audience's point of view (the audience are made up mostly of the schoolmates from the same class). Last but not least, it will focus on the preventive measures, which are supposed to prevent bullying in schools, and also how to detect it, solve it and eventually eliminate it. The practical part will be pay attention to the responses obtained from the questionnaire which will be given to pupils from upper primary schools and students from lower levels of comprehensive schools. The questionnaire will be focused on bullying. Based on the respondents' answers, graphs will be generated which should tell us two things: firstly, to what extent bullying behaviour occurs within the specified levels at these two types of schools and, secondly, how much the pupils and the students are aware of bullying. In other words, how much theory they know about bullying.
