Multi-year subsidy titles in the cultural sector at the municipal level

The aim of the thesis is to analyze and evaluate the variability of multi-year subsidy titles in the culture sector in a temporal and cross-sectional comparison, to determine the factors influencing the size and dynamics of subsidies, and then to compare the defined problematic areas with selected cities of the Central Bohemian Region.
Nursery Management in Pedagogical Process

The master's thesis deals with the management of nurseries and related requirements, focusing on the management and the quality of the pedagogical process with respect to managerial and administrative duties of head teachers. The goal of the study is to analyse the motivation for running for nursery head teacher, adaptation process and head teacher duties, including the impact of managerial and administrative responsibilities on the pedagogical process quality in nurseries. Special attention is paid to the comparison of head teachers of different types of nurseries. The empirical part of the master's thesis answers the following questions: What was the motivation of head teachers for applying for the job? How many hours a day do head teachers engage in pedagogical and managerial activities? Do head teachers believe that their managerial and administrative duties have a negative impact on the quality of the pedagogical process? What are the differences between head teachers' jobs at different types of nurseries? The thesis uses both qualitative and quantitative research. A survey was conducted among a larger number of respondents, providing a basis for a questionnaire for head teachers of eight selected nurseries. 1 Abstract of the thesis RŮŽIČKOVÁ M.; INVESTIGATION OF SUGAR MODIFIED PYRIMIDINE NUCLEOSIDES AS POTENTIAL INHIBITORS OF THYMIDINE (TP) AND URIDINE (UP) PHOSPHORYLASE Key words: uridine phosphorylase, thymidine phosphorylase, nucleoside, inhibitor, anticancer chemotherapy, substrate specifity The work deals with the investigation of novel potential inhibitors of recombinant thymidine and uridine phosphorylases from E. coli and Salmonella typhimurium. The main goals of this study are the determination of kinetic parameters for natural substrates (thymidine and uridine) for recombinant TP and UP, assessment of the role of phosphate anion in the enzymatic transformations and search for new inhibitors of TP and UP. Study of UP and TP substrate specifity and activity under the various reaction conditions is desired to obtain better view to their functions and to design novel structures of potencial inhibitors. The determination of the Michaelis constant KM and reaction rate Vmax was performed at the spectrophotometer Varian Cary-300 Bio. The representation of obtained kinetic data we used the Lineweawer-Burk plot. Reference gene selection for mRNA quantification in Haemonchus contortus

Genom and transcriptom have been published in 2013, allowing gene expression studies to be conducted. Reference genes for this studies have not been validated yet. Use of suitable reference genes is essential for accurate normalization of gene expression levels. Aim of this work was to identify and validate potential reference genes for gene expression studies in H. contortus adults. Eleven genes were chosen, stability of their expression was assessed in males and females of two H. contortus strains, one drug-susceptible (ISE) and one multi-drug-resistant (WR). Total RNA was extracted and reverse trancribed to cDNA. cDNA was diluted and analyzed using quantitative real-time PCR with SYBR Green I detection. Effectors of RAB GTPases and their role in plant secretion

Rab GTPases are small signaling molecules that play an important role in vesicle trafficking in eukaryotic cells. Correct signaling through small GTPases allows orchestration of vesicle transport among cellular organelles and also to the cell wall providing cell wall material for cell growth and elongation. Engagement of Rab GTPases in the regulation of endomembrane trafficking is one of the evolutionary conserved aspects of secretion regulation. The network of Rab GTPases interaction includes also various downstream effectors. One of them is the exocyst complex involved in vesicle docking at the plasma membrane. It is a complex composed of eight different subunits (Sec3, Sec5, Sec6, Sec8, Sec10, Sec15, Exo70 and Exo84). Exocyst was discovered as Sec4p Rab GTPase effector in yeast and also data from animal models describe the Sec15 exocyst subunit as the Rab-interacting partner, but data from plants are missing. On the other hand, numerous studies identified exocyst role in tip growth of pollen tube and root hairs, seed coat formation, cell plate and cell wall formation, hypocotyl elongation, and importantly also PIN auxin efflux carriers recycling and polar auxin transport. Influence of the satellite spatial resolution and scan time on convective storms'overshooting tops detection

This paper deals with the possibilities of convective storms' overshooting tops detection as seen from satellites. These tops are characterized by a rapid development, their size is only a few kilometres in diameter and they are connected with the presence of hazardous weather. They can be identified as small clusters of very low temperatures in satellite imagery, but this nature depends on the scan time and the scan frequency of the specific satellite and on the other features presence at tops of convective storm. The temporal variability of minimum IR brightness temperatures at the cloud tops of selected convective storms is studied in this paper, namely on the basis of the Meteosat Second Generation (MSG) satellite datasets. But the main emphasis of this work is to analyse the influence of the operative meteorological satellites' spatial resolution on the minimal IR brightness temperature, which is related to height of the cloud top. The datasets acquired by the sensors SEVIRI, AVHRR and MODIS have been chosen for these purposes. Mostly the convective storms that occurred over the Europe in the year 2008 have been diagnosed. Religion in the Czech Republic: Basic analysis and the relationship to selected socio-demographic topics Abstract The aim of this thesis is to analyse the inhabitants of the Czech Republic according to religious belief using data from surveys and also individual 2011 census data. Contemporary position of religion in society is described in the first part, focusing on characteristics of believers and non-believers. Results of surveys concerning religious belief in our region are stated in detail in the next part, oriented mainly on the period after 1989. The part with the analysis of differences in attitudes to problematic demographic behaviour according to religious belief follows, using the data from European Values Study and International Social Survey Programme. Individual data from 2011 census are analysed more in detail in the last part, focusing on newly spread religious groups and furthermore there is an analysis of believers using logistic regression.
