New types of magnetic sorbents for phosphoprotein analysis

The method for the study of protein phoshorylation sites was elaborated. This method is based on the IMAC separation of phosphopeptides from protein proteolytic digests using new magnetic sorbents and on their subsequent identification by mass spectrometry (MS). Magnetic non-porous hydrophilic poly(2-hydroxyethyl methacrylate-co-glycidyl methacrylate) particles prepared by the dispersion polymerization and modified with iminodiacetic acid (IDA) with immobilized Fe(III) and Ga(III) ions were employed for the enrichment of phosphopeptides from the proteolytic digests of two model proteins, porcine pepsin A and bovine α-casein. The optimum conditions for phosphopeptide adsorption and desorption in both cases were investigated and compared. The phosphopeptides separated from both proteolytic digests were analyzed by matrix-assisted laser desorption/ionization time-of-flight MS. For the immunochemical separation of phosphoproteins, protein fraction containing antibodies was obtained from egg yolk of hens immunized with O-phosphoryl-L-serine conjugated to key limpet hemocyanin. The History of the Pharmacies of Prior Region Rychnov nad Kněžnou

Závěr Cílem předkládané diplomové práce bylo podat celkový obraz o historii i současné situaci lékáren na Rychnovsku tak, aby poskytla základní informace nejen dnešním zájemcům o dějiny lékárenství, ale aby též mohla být zdrojem poznatků do budoucna. Na začátku byly stručně zpracovány dějiny Rychnovska ukazující vývoj od dob zemědělství po zavedení průmyslu a rozmach regionu, jako významné a hojně navštěvované kulturní a turistické oblasti. Podrobněji pak byly rozepsány dějiny jednotlivých obcí a měst, kde lze konkrétně vidět postupný vývoj a zejména dnešní situaci jasně ukazující na potřebu lékárny v dané obci či městě. Zachytit historii starých historických lékáren se podařilo na základě studia historických materiálů (metodika zpracování je v předešlé kapitole). Roku 1980 bylo na území bývalého okresu Rychnov nad Kněžnou 9 lékáren. V současnosti existuje v okrese Rychnov nad Kněžnou 24 lékáren. Již tento údaj ukazuje oprávněnost zpracování diplomové práce, navazující na již dříve zpracované materiály. Bez diplomové práce by dějiny 24 lékáren nebyly zachyceny a v budoucnosti by nikdo neznal dějiny lékáren v letech 1989- 2004. Jedná se o historicky velice zajímavé období. The Nazi ideology in relation to persons with mental disabilities

Diploma thesis "Nazi ideology in relation to mentally ill people" focuses on starting points and ideas of this ideology, which in its consequences led to social exclusion and subsequent systematic extermination of people with a certain handicap. Although the thesis is limited in time from 1933 - 1945, it also follows development line of selected scientific disciplines, which were significantly reflected in the Nazi ideology. Primarily it concerns historical science, medical disciplines, genetics, racial hygiene, eugenic origins and other related disciplines. The thesis also takes into account the economic situation of that time with its many social problems which arose as a result of the new geopolitical arrangement after the World War I and the changes in the social world of that period and it also captures the process of changes in the social status of mentally ill people in the course of history. Last but not least, it describes the practical consequences of these changes, which were "forced sterilization" and "euthanasia" of both children and adults. The Nazi ideology is viewed in this regard from three perspectives: legal, theoretical with a broader historical context and on the practical level in the form of the consequences of the two previous perspectives. Concept of the individual in relation to God in Kierkegaard and Nietzsche

This work deals with philosophy of Søren Kierkegaard and Friedrich Nietzsche. It focuses on the problem of their relationship with God and religion. The aim is to show where these thinkers agree on and differ in relation to God, Christianity and change of the values of an individual. The first chapter is devoted to foundations of Nietzsche and Kierkegaard's work. The second chapter compares common and different values. The work compares masters and slaves' morality in the concept of Friedrich Nietzsche and morality built on authentic faith at Søren Kierkegaard.
Success factors of rural municipalities

The aim of this thesis is to evaluate the development and success of rural municipalities within the administrative district of the Hradec Králové municipality. The thesis will take into account the influence of external development factors on the success of the municipalities, i.e. the influence of size in the form of population and the influence of location in the form of distance from the city Hradec Králové. In the spirit of the currently prevailing institutional directions of development, the emphasis on internal development potentials and actors and the so-called bottom-up approach in the rural development paradigm, municipalities will be evaluated on the basis of indicators based on these concepts, which can be used to assess the level of development and therefore the success of a rural municipality. The output of the evaluation will be an overview of each village and how its location in relation to the centre and its population affect its development. The final determination of successful and threatened rural municipalities will be compared for the entire Hradec Králové ORP. Peptide inhibitors immobilized on magnetic particles and Sepharose used for separation of stomach aspartate proteinases

IN ENGLISH Human gastric juice contains mainly aspartate proteinases: pepsin A and pepsin C. Both pepsins are produced by gastric mucosa as inactive pepsinogens and they are activated to the corresponding pepsins in the acidic environment of the gastric lumen. The levels of pepsinogens in serum reflect the morphological and functional status of gastric mucosa. A subject of this thesis is a part of a long-term investigation that focuses on the elaboration of methods for separation gastric aspartate proteainases that would be suitable for diagnostic purposes. The preparation of new type ligands was a concrete subject of PhD. thesis that after their immobilization they can enable the separation of aspartate proteinases. Four heptapeptides containing D-leucinyl residue were synthetized (Val-D-Leu-Pro-Phe-Phe-Val- D-Leu, Val-D-Leu-Pro-Tyr-Phe-Val-D-Leu, Val-D-Leu-Pro-Tyr-Tyr-Val-D-Leu and Val-D- Leu-Pro-Phe-Tyr-Val-D-Leu. The prepared heptapeptides immobilized on agarose magnetic particles were used for the study of their interaction with porcine pepsin A and rat pepsin C. While porcine pepsin A was adsorbed to all heptapeptides immobilized to magnetic particles, rat pepsin C was not retarded. Similar results were obtained using heptapeptides immobilized to Sepharose. Analysis of Presentation of Motif of Slum and Life in it in Czech Media in 2010-2017

The thesis deals with the media presentation of world slums in the Czech environment throughout 2010-2017. According to the latest estimates, more than one billion people live in slums today. It is safe to assume that the population in these slums will grow in the next decades. Most slums are located in the urban areas of developing states, which have higher urbanisation rates compared to the more developed countries. Slums are crowded places in cities where the residents live in substandard conditions that includes lack of drinking water, poor sanitation, lack of sewerage and health care and so on. The thesis focuses on the media coverage of five slums (Kibera, Dharavi, Khayelitsha, Ciudad Neza and Orangi Town), which rank among the biggest in the world. The work briefly summarizes the social construction of reality theory according to T. Berger and P. L. Luckmann, the media construct of reality and news value. It explains the concepts of slum, developing country, urbanisation, and the refugee crisis. It examines the texts by combining quantitative and qualitative content analysis. Selective Attention in Exchange Rate Forecasting

We analyze the exchange rate forecasting performance under the assumption of selective attention. Although currency markets react to a variety of different information, we hypothesize that market participants process only a limited amount of information. Our analysis includes more than 100,000 news articles relevant to the six most-traded foreign exchange currency pairs for the period of 1979–2016. We employ a dynamic model averaging approach to reduce model selection uncertainty and to identify time-varying probability to include regressors in our models. Our results show that smaller sizes models accounting for the presence of selective attention offer improved fitting and forecasting results. Specifically, we document a growing impact of foreign trade and monetary policy news on the euro/dollar exchange rate following the global financial crisis. Overall, our results point to the existence of selective attention in the case of most currency pairs.

Child Soldier in International Law

Zuzana Kučerová: Child Soldier in International Law Abstract This thesis deals with child soldiers from the perspective of the international law. It is predominantly concerned with two questions: protection of children from recruiting and their possible criminal responsibility for international crimes. After a short introductory chapter, which covers a brief history of child soldiers, the second part gives an overview of international legal instruments which aim at preventing the underaged from being recruited into armed groups, as well as from taking part in hostilities. Those instruments belong to three different branches of international law: humanitarian law, human rights law and international criminal law. The author concludes that international criminal law in particular is the best instrument to protect children from becoming child soldiers. The reason is that international criminal law applies directly to individuals, including non-state actors such as commanders of paramilitary forces. It is also in force at the times when there is no armed conflict as defined by international humanitarian law.
