Glykoproteiny se běžně vyskytují v organismech a hrají významnou roli jak za fyziologického, tak patologického stavu. U glykoproteinů se na jednom glykosylačním místě mohou vyskytovat odlišné glykany a změna glykosylace proteinů může být projevem onemocnění. Díky tomu mohou být využity glykoproteiny v diagnostice jako markery. Pro analýzu glykoproteinů se často využívá jejich naštěpení na glykopeptidy, které jsou poté identifikovány. Hojně používanou metodou pro studium glykopeptidů je kapalinová chromatofrafie propojená s hmotnostní spektrometrií a pro separaci glykopeptidů je možné použít hydrofilní interakční chromatografii (HILIC). Tento typ chromatografie umožňuje separovat glykopeptidy na základě vlastností jak peptidu, tak navázaného glykanu. Prvním cílem práce bylo určení míry vlivu náboje peptidové páteře glykopeptidů při jejich separaci na HILIC. Pro tento účel byly použity kolony: Penta-HILIC a HILIC-B. Penta-HILIC obsahuje polyhydroxylovou stacionární fázi, zatímco stacionární fáze HILIC-B je tvořena silikagelem modifikovaným kladně nabitými aminopropylovými skupinami. Na obou kolonách byly separovány glykopeptidy IgG1 a IgG2. Pro určení míry vlivu náboje a hydrofobicty při separaci na HILIC-B byla provedena propionylace a esterifikace, při kterých došlo k eliminaci náboje na nabitých... Glycoproteins are commonly found in organisms and play an important role in both physiological and pathological conditions. In glycoproteins, different glycans may be present at the same glycosylation site and altered glycosylation of proteins may be a manifestation of disease. As a result, they can be used as markers in diagnostics. For the analysis of glycoproteins, their cleavage into glycopeptides, which are then identified, is often used. Liquid chromatography coupled to mass spectrometry is a widely used method for the study of glycopeptides and hydrophilic interaction chromatography (HILIC) can be used for the separation of glycopeptides. This type of chromatography allows separation of glycopeptides based on the properties of both the peptide and the bound glycan. The first aim of this work was to determine the extent to which the charge of the peptide backbone of glycopeptides affects their separation by HILIC. Penta-HILIC and HILIC-B columns were used for this purpose. Penta-HILIC contains a polyhydroxyl stationary phase, while the stationary phase of HILIC-B consists of silica gel modified with positively charged aminopropyl groups.
