[cze][eng] 3 Abstrakt Antrální folikul poskytuje oocytu specifické prostředí - niku, která během oogeneze podporuje vznik kvalitních oocytů. Vývojová kompetence oocytů skotu je ovlivněna velikostí folikulu, ze kterého pochází. Oocyty izolované ze středních nebo větších antrálních folikulů (≥6 mm) mají vyšší vývojovou kompetenci (schopnost vyvinout se do stádia blastocysty). Změny cytoplazmatických faktorů, např. mRNA, odrážejí vývojový potenciál oocytu. S pomocí mikročipu jsme charakterizovali nematurované oocyty izolované ze středních folikulů (MF, 6-10 mm) a malých folikulů (SF, 2-5 mm) za účelem odlišit různě vývojově kompetentní oocyty na základě genové exprese. Celkem u 61 genů byl rozdíl v expresi ≥ 1,4×. Pro validaci výsledků získaných z mikročipu bylo pomocí kvantitativní RT-PCR vybráno 15 genů. Před maturací byly potvrzeny signifikantní rozdíly u nezralých oocytů izolovaných z SF a MF u genů ATP5C1, MAP3K13, MTRF1L, TAF1A a UBL5. U 12 genů byla stanovena hladina mRNA po maturaci u subpopulací oocytu rozdělených na základě atrézie kumulárních buněk a velikosti folikulu. Hladina transkriptu ATP5F1 se po maturaci neměnila u všech kategorií. V případě BRD7 zůstalo množství transkriptu na stejné úrovni u oocytů bez atretických kumulárních buněk nezávisle na velikosti folikulu. U TAF1A se snížila hladina mRNA u...4 Abstract The antral follicle provides a specialized microenvironment or niche, which is necessary for production of high quality oocyte. The developmental competence of bovine oocyte is influenced by the follicle size. Oocytes originated from medium or larger follicles (≥6 mm) have greater developmental competence (ability to develop to the blastocyst stage). The changes in cytoplasmic factors, for example mRNAs, could explain differences in oocyte developmental potential. Using Bovine Oligonucleotide microarrays the differences in gene expression profiles of oocytes at germinal vesicle and MII stages from medium (MF, 6-10 mm) or small (SF, 2-5 mm) follicles were characterized. The aim was to find differencies between oocytes diverse developmental competence. The expression fold change between the two experimental groups was in 61 genes. Subsets of 15 differentially expressed genes were validated by quantitative RT-PCR. Before maturation, significant differences were confirmed at the level of ATP5C1, MAP3K13, MTRF1L, TAF1A and UBL5. Subpopulations of oocytes were classified according to atresia of cumulus cells and follicle size. We determined the level of 12 individual transcripts after maturation.
