Application of mosses and lichnes for the evaluation of presence of the metals in the environment

This bachelor thesis deals with the use of mosses and lichens as plant bioindicators, to assess environmental pollution by metals. Defines the sources of metals in the environment, describes the morphology of species of mosses and lichens. The last part refers to methods of active biomonitoring which use mosses and lichens and proposes concrete species of mosses and lichens, which could be used to assess environmental platinum metal pollution.
Determination of palladium by ET-AAS after previous preconcentration by solid phase extraction

The theoretical part of this master’s thesis focuses on the characteristic of Palladium, his uses in the automotive convertors, the occurrence in the environment, bioaccumulation and human health risks. Next there are described the basics of biomonitoring and summarize suitable biomonitoring method for capturing airborne dust. There are also evaluated appropriate methods for preconcentration and determination of Palladium. The aim of experimental part of this thesis is description and evaluation optimalization preconcentration method SPE by using sorbents silicagel C18 and QuadraSil TA. Optimization of method conditions of electrothermal atomic absorption spectrometry (ET – AAS). The determination of Palladium using optimal conditions follows in real samples in the form of exposure lichen (Hypogymnia physodes) by automobile traffic in Brno.
Utilization of plant bioindicators in monitoring of environmental contamination

Environmental contamination may be observed by different analytical tools as well as by various types of bioindicators providing relevant information about the effects of toxic substances and their mixtures towards organisms in situ. The aim of this work is to provide a comprehensive overview of plant bioindicators and biomonitors suitable for biomonitoring of persistent organic pollutants (POPs) in natural ecosystems. For groups of organic pollutants such as PCBs, PCDDs / Fs, PBDEs, PAHs, PFCs, and OCPs, review of literature is provided including knowledge about different species of coniferous trees, angiospermous plants (including crops), mosses and lichens that can be used for monitoring. In this work, analysis examples of samples of pine needles, bark, moss and lichen tissue are quoted as biomonitor tools. An overview of plants for phytotoxicity and genotoxicity observation using pollen grains abortivity and online bioindication as an example on non-specific biomonitoring is also provided.
There is something in the air: Diversity of lichen symbionts in the aeroplankton

Lichens that reproduce primarily sexually rely on the presence of their symbionts in the environment. A new lichen cannot develop without direct contact between the filaments of the germinating spore and a compatible symbiotic alga. However, the results of earlier studies do not provide solid evidence for photobiont presence on lichen-colonized substrates. The objective of this thesis was to assess various environmental types (air, bark, soil, stone, and artificial substrates) as potential sources of symbiotic algae. To achieve in-depth understanding of the diversity of green algae at a long-studied Central Bohemian study-site, Illumina MiSeq metabarcoding of the ITS2 rDNA region was combined with traditional sequencing of lichen algal ITS rDNA and morphological observations. The methods used revealed 283 species-level lineages and 1313 OTUs, many of which belonged to unknown species or higher taxonomic categories. All photobiont species associating solely with sexual lichens at the site could also be found in the vicinity of lichen thalli. As a result, both the substrates and the air, where these symbiotic algae occurred as well, may act as photobiont sources. In contrast, many symbionts that associate only with asexual lichen species, were never found at the site outside of lichen thalli....
Bacteria in the lichen symbiosis

This thesis summarizes the most recent research on bacteria in lichen symbiosis. Lichen symbiosis and the partners participating in it are described in the first part of the thesis. The emphasis is mainly on the different functions that bacteria can have in lichen symbiosis. These functions include, for example, the fixation of atmospheric nitrogen, the production of cobalamin, essential for the growth of some algae, the secretion of antimicrobial substances and the recycling of nutrients contained in old parts of the litter. The main groups of bacteria that occur in the lichen symbiosis are also discussed, along with the factors that influence their occurrence. The most abundant group of bacteria in lichens are the Alphaproteobacteria. Their abundance in lichens is probably influenced by the frequent occurrence of these bacteria in the substrate on which the lichen grows. Other abundant groups of bacteria in lichens are representatives of Firmicutes, actinobacteria or acidobacteria. All important groups of bacteria are acquired by lichens through both vertical transfer and horizontal transfer. Complexity of lichen symbiosis

Lichens are currently viewed as complex symbiotic systems. In addition to the main mycobiont and photobiont, a variety of associated fungi, bacteria and algae/cyanobacteria (minor/accessory photobionts) have been recognized. Their diversity has been intensively studied, but is still far from being fully apprehended. Likewise, recognition of the significance of the associated organisms to the whole system is still at the beginning but various crucial roles, from constitutive morphogenetic through physiological to various means of increasing the lichen's fitness, have already been suggested. The present thesis attempts to approach lichens in their full complexity. Focusing on two model systems; the Cladonia-Asterochloris association and ecologically delimited communities of Verrucariaceae; it aims: i) to examine patterns in photobiont choice and their relationship to lichen ecology; ii) to set a framework for in-vitro mycobiont-photobiont compatibility testing; iii) to explore the diversity of selected associated fungi and their possible relationships with the lichen host. We have shown that Verrucariaceae in the intertidal zone associate with largely understudied Ulvophycean photobionts. Let's meet in the littoral: Diversity of symbiosis in the Verrucariaceae lichens

Lichens are organisms formed by several symbiotic partners, most importantly a fungus (mycobiont) and an alga or cyanobacteria (photobiont). Although most lichens are terrestrial, some can be found also in aquatic environments. Black crustose lichens from the family Verrucariaceae (Verrucariales, Eurotiomycetes, Ascomycota) were able to adapt to this environment very well and often dominate in the seashore near fluctuating water level. The ability of these lichens to inhabit various environments is associated with their unusually wide range of photobionts. However, this symbiont is not known to most of them. This work investigates the diversity of lichens of the family Verrucariaceae in the littoral zone of the Baltic and the North Sea coasts, the relationships between symbionts and the connection between lichen diversity and the salinity gradient. Using metabarcoding, the diversity of free-living algae in the vicinity of the studied lichens is determined and the symbiotic algae are compared with the photobionts found in the lichens. Laboratory and image spectroscopy for mapping of selected rocks in peak areas of the Krkonoše Mountains Abstract This thesis deals with geological mapping of selected rocks in peak areas of the Krkonoše Mountains. Four areas of interest were situated in two parts of Krkonše Mountains - on the west side it is the area of Vysoké kolo and Harrachovy kameny and on the east side there is the area of Sněžka and the area of Kozí hřbety. The main data were acquired by the hyperspectral sensor APEX. Ground spectral measurments of selected rocks and block fields were executed and the laboratory spectral measurments of geological samples and lichens were executed. Practical part aims at classification of rocks and lichens in selected areas using four classification methods: SAM, SID, MESMA and LSU. The spectral library is one of the outputs of this thesis. This spectral library contains the spectra of pure rocks and lichens and mixtured spectra of rocks and lichens. The output of this thesis is the comparation of used classification methods, the analysis of spatial and geological accuracy and evaluation of lichens influence on the classification results, spectral library and maps of classified rocks occurrence. Microclimate as a driver of species distribution and community composition of bryophytes compared with lichens

This bachelor thesis deals with the influence of microclimate on species distribution and composition of bryophyte and lichen communities. It defines the term microclimate, characterizes the specific properties of bryophytes responsible for their sensitivity to microclimatic conditions and includes a comparison of the role of individual microclimatic factors in controlling the distribution of bryophytes and lichens. The key microclimatic factors for bryophytes are humidity and air temperature, but the significance of each varies between functional groups of species and depending on the type of biome. With the available portable measuring devices with large battery capacity and memory storage, it is now possible to measure in-situ microclimate for the long-term and also with high spatial and temporal resolution. Despite that, there are currently only 12 bryological studies that provide continuously measured field microclimatic data on small spatial scales. The main part of the thesis summarizes these studies and compares the methodologies used. The paper can serve as a starting material for designing bryological microclimatic studies.
Lichens and eutrophication

Eutrophication is a process of organic nutrients enrichment in an ecosystem, especially nitrogen and phosphorus. This bachelor thesis is dealing with the influence of eutrophication on lichens, especially with the effect of nitrogen compounds in the air and the effect of the combination of nitrogen compounds with phosphorus on epiphytic lichens. Eutrophication is defined and information is provided on changes in abundance and diversity of lichen cover. The mechanisms of eutrophication tolerance in lichens are also described.
