Effect of cadmium chloride on P-glycoprotein in the blood-brain barrier
Zahradníková, Tereza ; Štaud, František (advisor) ; Hofman, Jakub (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Student: Tereza Zahradníková Supervisor: Prof. PharmDr. František Štaud, Ph.D. Consultant: Alexander Zaremba, Institute of Pharmacy and Molecular Biotechnology, Heidelberg University, Germany Title of diploma thesis: Effect of cadmium chloride on P-glycoprotein in the blood-brain barrier The blood-brain barrier (BBB) separates the central nervous system (CNS) and the peripheral blood circulation. It regulates the material transport between these compartments due to its specialised structure and cellular constitution. The endothelial cells forming the BBB are characterized by the expression of different multidrug resistance proteins which belong to the ATP-binding cassette (ABC) transporter family. These transmembranous ABC proteins actively transport molecules out of the BBB endothelia into the bloodstream and protect the brain against harmful xenobiotics, toxins and metabolites. On the other hand, ABC export proteins constitute obstacles to the delivery of many therapeutic drugs across the BBB into the CNS, thus the efficacy of CNS pharmacotherapy is limited. One of the most important efflux transporters is P-glycoprotein (P-gp). Cadmium is a heavy metal that is dangerous to human health....
Psychological aspects of pain management
Zahradníková, Tereza ; Niederlová, Markéta (advisor) ; Kebza, Vladimír (referee)
Bachelor's thesis deals with pain management from psychological perspective. It covers emotional, cognitive and behavioral factors, pain coping strategies and individual differences in pain management. This work focuses on the time dimension of pain. The aim is to describe psychological aspects of acute and chronic pain management - the differences between acute and chronic pain management and the role of psychological aspects in pain chronification. Psychological aspects of pain, especially pain coping strategies, are evaluated in terms of efficiency and adaptability. The research project is designed as a prospective study investigating if there are any pain coping strategies related to pain chronification and if the coping strategies change during pain chronifiaction. Keywords: pain management, coping strategies, acute and chronic pain, chronification of pain
Czech augmentative suffixes in translation from English
Zahradníková, Tereza ; Popelíková, Jiřina (advisor) ; Tichý, Ondřej (referee)
This BA thesis focuses on the analysis of English expressions that are translated into Czech by using words containing augmentative suffixes. The theoretical background presents the general concept of augmentatives and then it focuses on describing them in terms of the individual linguistic systems of English and Czech. In this section, they are discussed mainly from the point of view of their formation. While Czech makes use of suffixation to create augmentatives, English incorporates splinters and neoclassical compounds. Subsequently, the thesis draws a comparison between the English and Czech systems. The analysis examines English equivalents of Czech expressions containing 18 different augmentative suffixes. It focuses on the comparison of the individual translation counterparts and on analyzing augmentatives from the formal and semantic points of view. The search is conducted in the Czech- English parallel corpus InterCorp and the overall amount of data for the analysis is 68 lemmas and 593 tokens. The English expressions are divided into multiple formal categories and subsequently compared with their Czech translation counterparts. The results of the analysis showed that from the formal point of view, the most frequent equivalents of Czech augmentatives are nominal expressions, followed by adjectival...
Solubilisation, Purification and Reconstitution of Human 17beta-hydroxysteroid dehydrogenase type 3
Zahradníková, Tereza ; Navrátilová, Hana (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Mgr. Tereza Zahradníková Supervisor: Prof. Ing. Vladimír Wsól, Ph.D. Consultant: RNDr. Lucie Zemanová, Ph.D., PharmDr. Adam Skarka, Ph.D. Title of rigorous thesis: Solubilisation, Purification and Reconstitution of Human 17beta-hydroxysteroid dehydrogenase type 3 Short-chain dehydrogenase/reductase superfamily is a large group of NADP(H)/NAD(H)-dependent oxidoreductases. Human SDR enzymes are classified into 47 families, including cytosolic and membrane-bound ones. One of them is a human membrane-bound enzyme 17β-hydroxysteroid dehydrogenase type 3 (17β-HSD3), which participates in the biosynthesis of steroidal hormones and mainly catalyses the conversion of androstenedione to testosterone. The aims of this study were to find a suitable detergent for successful solubilisation, to purify the enzyme and to prepare a reconstitution system for further study of the 17β-HSD3 enzyme. Microsomes containing overexpressed 17β-HSD3 were isolated from Sf9 insect cells (Spodoptera frugiperda). The first step was the solubilisation process, which involved detergent screening. Six detergents at different concentrations were tested: ASB 14-4, C12E8, DDM, CHAPS, Igepal CA-630 and Triton X-100. The detergent ASB...
Sleep quality of older adults
Zahradníková, Tereza ; Kebza, Vladimír (advisor) ; Niederlová, Markéta (referee)
This master thesis focuses on subjectively percieved quality of sleep and its connections - fatigue, daytime sleepiness and insomnia in older age. Attention is brought to psychological methods of sleep measurement in comparison with objective methods (polysomnography and actigraphy). Theoretical part is based on current scientific knowledge of sleep quality and deals with psychological factors that contribute to improvement or reduction of sleep quality in older age. In context with quality of sleep focus is aimed also at presence of insomnia in older age compared to younger age and possibilities of enhancing sleep quality. Overview of subjective and objective methods of measuring sleep quality and other sleep characteristics is part of the theoretical background. Empirical part of the thesis is divided into two sections. First section compares sleep quality, fatigue, daytime sleepiness and insomnia of younger and older adults. This research is based on following questionnaire data: PSQI (Pittsburgh Sleep Quality Index), FSS (Fatigue Severity Scale), ESS (Epworth Sleepiness Scale) and ISI (Insomnia Severity Index). Second section of the research verifies the validity of psychological methods of measuring sleep quality - questionnaires PSQI, FSS, ESS, ISI and sleep diaries of older adults by...
