Economic Efficiency of Public Construction Project
Weber, Jiří ; Hromádka, Vít (referee) ; Korytárová, Jana (advisor)
This bachelor's thesis deal with economic efficiency of public construction project. Information about efficiency it takes from Cost-benefit analysis. Cost-benefit analysis compares economic cash-flow and it rates project´s usefullness versus total project´s cost. In the economic cash-flow are financial cash-flow, as investment, yield and cost, and socioeconomic cash-flow. In thit part is project´s effect on the community in the formof benefits and savings. This effect we can see in a project as a unit price. For this cash-flow result indexs of ekonomic efficiency, which evaluates efficiency, or demand of grant. The one of part economic efficiency is project life cycle´s financial sustainability.
The role of Hepatitis B virus capsid protein in the host ubiquitin proteasome pathway
Eliáš, Vratislav ; Weber, Jan (advisor) ; Šmahel, Michal (referee)
Hepatitis B virus (HBV) is a Hepadnaviridae virus infecting mammals. Its infection can result in an acute or chronic infection. Chronic infection can result in hepatocellular carcinoma and liver cirrhosis, potentially leading to death of the patient. HBV is a small 42 nm virus with a genome length of 3.2 kb encoding seven viral proteins. HBV Core protein (HBc) is a capsid forming protein which is pleiotropic in function. We have identified two ubiquitin ligases which could interact with this protein: F-box only protein 3 (FBXO3; E3 ubiquitin ligase) and Ubiquitin conjugating enzyme E2 O (UBE2O; E2/E3 ubiquitin ligase). By employing multiple methods we have confirmed these interactions. Co- immunoprecipitation and further western blot analysis unveiled multiple new insights into the ligases′ impact on HBc: FBXO3-mediated HBc polyubiquitination stimulation and UBE2O-mediated HBc monoubiquitination promotion. FBXO3's and UBE2O's role in HBV life cycle was investigated as well. By silencing the expression of FBXO3 and UBE2O respectively, we have observed changes in HBV replication levels: FBXO3 serves as an inhibitor of HBV replication, while UBE2O stimulates the course of HBV life cycle. Further investigation of these newly-discovered understandings may lead to a whole new HBV - host interplay...
Mapping history of music therapy in Bohemia using interview approach
Koukl, Jonáš ; Lipský, Matěj (advisor) ; Weber, Jana (referee)
This diploma work is focusing primary on capturing and approximation of historical development of the music therapy in the Czech Republic through the use of interview with several personalities taking active part in the music therapy opening or development. In the theoretical part, reader is introduced to definition of the music therapy. There are various definitions from worldwide institutions and figures. In the next chapter the work deals with various styles of the music therapy. The third chapter covers the important events and persons, which were closely connected with music therapy from prehistory till the present time. The practical part of this work forms transcriptions of mentioned interviews, concretely with these persons: Josef Krček. representative of anthroposophy music therapy, Jitka Vodňanská, Viktor Zappner and Jana Procházková as pioneers in clinical approach to music therapy, Zdeněk Šimanovský with his systemic conception, and Vlastimil Marek, perceiving by specialists as the most important person of esoteric music therapy. By the end of the work, based on these transcriptions, an actual summary comes with different kinds and views on the music therapy in the Czech Republic.

Hypertension and nutrition: Fat-soluble vitamins A, D and E Weber J1 , Vlcek J1 , Suarez-Varella MM2 1 Department of Social and Clinical Pharmacy, Faculty of Pharmacy in Hradec Králové, Charles University in Prague, Czech Republic 2 Department of Preventive Medicine and Public Health, Faculty of Pharmacy, University of Valencia, Spain Arterial hypertension (AH) is a disease affecting population globally, and thus considered as a problem of public health and socioeconomic. Studies are trying to identify the connection between diet and the prevalence of arterial hypertension. Objective of the study was to determine possible association between an occurrence of AH and fat-soluble vitamins A, D and E intake. The nested, case-control population study investigation was grounded on database from the Spanish Hortega study, and performed on a random sample of 1,514 people (50.3 % women, 49.7 % men). From this sample we selected those aged ≥ 40 years old and untreated for hypertension and divided them into two groups: non-hypertensive (n = 429; 63.6 %) (controls), and newly diagnosed AH (n = 246; 36.4 %) (cases). Biochemical and anthropometric measurements, data on dietary intakes, education, socioeconomic status, place of residence, health habits, comorbidities, consumption of alcohol and tobacco were used for our...
The role of capsid protein in hepatitis B virus life cycle
Eliáš, Vratislav ; Weber, Jan (advisor) ; Drda Morávková, Alena (referee)
Hepatitis B virus (HBV) can cause either an acute or a chronic infection of hepatocytes, often leading to a hepatocellular carcinoma or a liver cirrhosis. HBV encodes seven proteins in its 3.2kb genome. Two of these proteins are transcribed from the same ORF (HBcAg and HBeAg). HBeAg, as a soluble variant of the core protein, plays a crucial role in virus immunogenicity. HBcAg, apart from building viral capsid, represents a pleiotropic protein engaging in multiple viral stages. Its primary structure is divided into two domains: an assembly domain and a C-terminal domain (CTD). The latter, containing four arginine clusters, is an important life cycle regulation element. By interaction with multiple viral and cellular factors, it directs the virus through a successful replication. In this thesis, we have collected the available data about the various roles of HBcAg in viral life cycle. We believe that further investigation of this field could lead to therapeutic advances resulting in the decrease of HBV infections worldwide, subsequently to a lower lethality connected with the diseases caused by HBV. Contribution of gag region to overall HIV replicative fitness in patients with different disease progression
Suchý, Tomáš ; Weber, Jan (advisor) ; Šmahel, Michal (referee)
Human immunodeficiency virus (HIV) is globally spread virus without available cure. Since its life-long presence, virus is carefully monitored as well as patient's immunological status. Replicative fitness of the virus is one of important aspects which can be taken into account, when monitoring HIV. Here, we are measuring HIV replicative fitness of gag recombinant viruses and comparing the results with replicative fitness of primary isolates. Further, we are comparing our findings of replicative fitness change over time with disease progression in the patient. We found that gag can be major contributor to overall fitness, although not in all cases. Additionally, we observed a correlation of replicative fitness development and slope of patient's CD4+ T cells. Moreover, this relation was even more noticeable in patients with slow disease progression or in carriers of protective alleles. In summary, our results extend the understanding of replicative fitness and its role in disease progression; and pave the way to use the recombinant HIV for replicative fitness measurement in clinical practice. Keywords: HIV, replicative fitness, recombinant virus, HIV disease progression, gag
Music Communication, Projection and Analogy of Handicapped Children in Music Therapy
Lipský, Matěj ; Květoňová, Lea (advisor) ; Mastnak, Wolfgang (referee) ; Weber, Jana (referee)
/Abstract Music Communication, Projection and Analogy of Handicapped Children in Music Therapy Presented work takes an interest in music contents produced by handicapped children attending music therapy sessions. The contents of music were gained from the children by the method of improvisation, particularly by "concert technique". In the theoretical part we present philosophical background for the music therapy in a field of special education and research. This background thought we have found in a gnoseology of a critical realism and a radical constructivism. Afterwards the definition and description of used music therapy terminology follows. We present three music therapy models that put great emphasis on the contents of music produced by clients. These models are: the German Morphological music therapy, the Dutch Analogical Music Therapy and the Geneape music therapy model from the Czech Republic. We also discuss general possibilities of using music therapy while working with mentally handicapped clients, autistic clients and clients suffering from sight defects, because these kinds of handicaps are included in our research group. At the end of the theoretical part we explain the terms of music communication, projection and music analogy. They are always connected to the contents of music, as...
