Evaluation of the Economy of the Town District Brno-střed

This thesis deals with an economy of city district Brno-střed. First part is theoretical, dedicated to functioning of self-government, description of this city district and financing state from budget of community. The second part is analytical and evaluates economy of last three years; period from 2012 to 2014. Third part describes suggestions for improvement of economy of city district Brno-střed with aspect to the next budgeting period.

Theory is described in the first part and serves as basis for following parts. Second part reveals analyses of an external and internal environment of the company. The last part states exact proposals and their implementation for the communication mix of the banking company.
Stress and strength analysis of the bicycle crank arm of a non-circular profile

This bachelor’s thesis deals with analytic solution of torque of a rectangular profile bar. Solution stands on Prandtl’s theory of torque of non-circular cross-section bar. Stress is thought about shear stress which is given by torsional moment and shear force, further about normal stress which is given by bending moment and tensile force. Application for this problem is stress and strength analysis of of the bicykle crank arm. Calculation is solved with MATLAB software.
Optimizing design of the slide CNC horizontal boring machines

This master's thesis deals with proposals for solutions to increase the stiffness of the slide for CNC horizontal boring standing in order to optimize the design of the slide. Selected problem was solved by sensitivity analysis using desegin of experiment in conditions of linear regression analysis. Data for the sensitivity analysis were obtained by using computational modeling using finite element method. The solution provides a view of the degree of influence on the deformation of the examined elements of the slide. Based on these results were selected elements with the greatest influence and created three ideological designs for improvement. Of these designs, was chose an optimal variant by PATTERN method acoording on the technical, economic and technical-economic aspect. This work offers a new look for the innovating the design to increase rigidity.
Mechanisms determining the identity of intestinal epithelial cells

(ENG) The intestinal epithelium has been heavily studied for decades. It is a traditional model to study the natural renewal of tissue during homeostasis and upon the damage, carcinoma establishment and progression and regulation of stem cells by Wnt signalling. Since the main interest in the tissue is focused on epithelial stem cells, it has been described quite recently that also all differentiated epithelial cells from the small intestine including enterocytes undergo drastic changes in properties during their short life. This phenomenon came to be known as villus zonation. In parallel with that observation, the heterogeneity of intestinal mesenchymal cells and their impact on intestinal epithelium have been gradually revealed. Distinct populations of mesenchymal cells secrete specific signalling molecules, which determine the destiny of epithelial cells. Those signalling molecules regulate stem cell maintenance as well as villus zonation. Our work combines those novel findings of intestinal enterocytes and mesenchymal cells. We studied the effect of distinct Bmp ligands produced by mesenchymal cells on the differentiation of enterocytes. Meta as a Digital Sovereign: Digital Aspects of non-state normative system in 21st century Abstract: This thesis examines the normative system appliccable at Meta platforms through the lens of the theory of legal pluralism and by analysing the decisions of the Oversight Board and attempts to answer the question of whether there is such a thing as Facebook law. In the second chapter, I describe the development of legal pluralism and its transition from a descriptive analysis of the plurality of legal orders to a normative theory of how to deal with this plurality. Within this chapter, I confront traditional views and understandings of the concept of law with alternative definitions and conceptions penned by the authors of legal pluralism, along with alternative views of the concept of authority. I conclude this chapter with three main strands of critique of legal pluralism that I have identified in the literature. The bachelor thesis focuses on the research of reverse logistics of waste in the banking company Raiffeisenbank a. s. The thesis consists of three parts. The first part covers theoretical background on the basis of which in the second part, the research is carried out using the analysis of the internal and external environment. The third part contains proposals for improving the waste recycling system at the Raiffeisenbank branch in Olympia and an evaluation of achieved results.

5 1 ÚVOD Buněčné signalizační kaskády představují klíčový molekulární mechanismus, který koordinuje veškeré životní procesy v buňkách, a tím i v celém organismu. Aktivita signálních kaskád je nezbytná pro regulaci buněčného růstu, diferenciaci buněk či jejich řízené odumíraní, a to jak v průběhu embryogeneze, tak v dospělém organismu. Signální dráhy zprostředkovávají odpověď buněk na okolní podněty, jsou nezbytné pro mezibuněčnou komunikaci a bez synchronizované aktivity složité signalizační mašinérie si nelze představit masivní buněčnou expanzi spojenou s tvorbou tkání během vývoje každého jedince. Jednou z těchto signalizačních kaskád je vysoce evolučně konzervovaná signalizační dráha Wnt. Předkládaná práce ve stručnosti uvádí do problematiky molekulárních mechanismů signalizace Wnt. Experimentální část byla zaměřena zejména na signalizaci Wnt v buněčném jádře a vztah mezi signalizací Wnt a jinými buněčnými regulačními drahami. 122 4 ZÁVĚR Cílem předkládané práce bylo přispět k poznání mechanismů, které ovlivňují aktivitu signalizace Wnt v buněčném jádře. Hlavní výsledky lze shrnout do následujících bodů: 1. Zjistili jsme, že transkripční korepresor CtBP1 má schopnost blokovat signalizaci Wnt, a to prostřednictvím inhibice transkripce cílových genů této signalizace. Represivní působení proteinu CtBP je podmíněno aktivitou s ním asociovaných histon deacetyláz. 2. Identifikovali jsme nádorový supresor HIC1 jako nový inhibitor signální dráhy Wnt. HIC1 přímo interaguje s klíčovým proteinem signalizace Wnt transkripčním faktorem TCF-4 a tato interakce je základem pro inhibičního působení proteinu HIC1. HIC1 relokuje transkripční komplexy TCF-4/β-katenin do specifických jaderných struktur a brání tak jejich vazbě na specifické cílové promotory, čímž dochází k zablokování transkripce. Pro efektivní relokaci komplexů TCF-4/β- katenin je důležitá strukturní funkce proteinu CtBP. 3. Nukleární recetor NHR-25 je shopen v buňkách prekurzorů gonády hlístice Caenorhabditis elegans interagovat s β-kateniny WRM-1 a SYS-1. Interakce s těmito β-kateniny moduluje transkripci cílových genů signalizace Wnt. Vzájemná interference mezi aktivitou signální dráhy Wnt a proteinem NHR-25 je rozhodujícím dějem, který determinuje osud somatických buněčných...
