Usage and environmental stability of MSWI bottom ash in road construction
Pospíšilová, Eliška ; Ettler, Vojtěch (advisor) ; Šebek, Ondřej (referee)
This bachelor thesis is a compilation of scientific and technical literature focused on treatments of municipal solid waste incinerator (MSWI) bottom ash such as carbonation or electro-kinetic remediation for the purpose of the decrease of the contaminant lechability. The carbonation the cheapest solution and is generally used as a first stage of MSWI bottom ash treatment. The second part of the thesis deals with possible applications of MSWI bottom ash for the construction purposes. Addition of bottom ash to the concrete is used as the alternative of natural purposes. Addition of bottom ash to the concrete is used as the alternative of natural gravel. Due to similar physical properties with crushed aggregate, bottom ash is also commonly used as a subbase of roads or pavement in numerous countries. Mixed bottom ash with the Portland cement can also find some applications as a light concrete with similar physical and mechanical properties.
Recombinant expression and studies on DCL-1, receptor of dendritic cells
Pospíšilová, Eliška ; Bezouška, Karel (advisor) ; Novák, Petr (referee)
Recombinant expression and studies on DCL-1, receptor of dendritic cells Eliška Pospíšilová Charles University in Prague, Faculty of Science, Department of Biochemistry ABSTRACT Dendritic cells, which could be found in large numbers in many tissues, exprime C-type lectine receptor DCL-1 (in CD nomenclature CD302) on their surface. This molecule belongs to the class one transmembrane proteins. N-terminal sequence a C-type lectin-like domain (CTLD) it has on the extracellular side. Whereas the C-terminus, which contains potential phosphorylation site and signals for intracellular transport, is in the cytosol. As the sequence of this protein is highly conserved through all mammalian species, it is likely to play an important role in the immune system. Despite this fact, the DCL-1 molecule is still poorly investigated. This work deals with the extracellular part - especcially with the CTLD containing part - of the DCL-1 receptor. To learn more about this molecule, we attempted its recombinant expression in bacteria. We used pET-30a(+) based vector pDCL1E, and bacterial cells E. coli BL21 (DE3) Gold, which produced the target protein in the form of inclusion bodies into the cytoplasm. Therefore conditions for in vitro renaturation were optimalized, and it was prooved that the protein had native conformation...
Rare earth elements in electronic waste
Pospíšilová, Eliška ; Šebek, Ondřej (advisor) ; Strnad, Ladislav (referee)
Rare earth elements consist of 15 lanthanides, Y and Sc. They are currently very important materials in many industrial branches (metalurgy, pollishing). One of these options is the usage as the compound of phosphors in electronic devices (television and computer displays. Because of their manufacturing cost, options of recovery from waste electrical and electronic equipment (WEEE) are investigated. This thesis deals with compoud elements of phosphors from displays of type CRT (cathode ray tube) and plasma TV. The 37 samples of phosphors from different displays were taken and processed for study of the compoud elements. Phosphors were separated to 4 subgroups in accordance with the corresponding type of displays: black-and-white CRT television displays (CRT_CB), color CRT television displays (TV_B), color PC displays (PC_B) and monochromatic PC displays (PC_M). The study was carry out by multiple features analysis that combined the inductively coupled plasma mass spectrometry (ICP-MS), the inductively coupled plasma optical emission spectrometry (ICP-OES), the resistance furnace with infrared detector (RF-IF) and the atomic absorption spectrometry (AAS). Based on the results of this analysis, main elements and potentionally hazardous elements of phosphors subgroup were defined. Concentration of rare...
Studies on DCL-1, receptor of dendritic cells, using NMR techniques
Pospíšilová, Eliška ; Kavan, Daniel (advisor) ; Schneider, Bohdan (referee)
(EN) The DCL-1 receptor (CD302) is predominantly expressed on the surface of dendritic cells and according to its sequence similarity DCL-1 is classified as a C-type lectin. Since its extracellular domain lacks single motives for carbohydrate binding in coordination with calcium ions, it is probable that the process of carbohydrate binding does not occur through the classical pathway as described in the case of mannose receptor or the DEC-205 receptor. Due to its colocalization with F-actin there is a presumption, that DCL-1 plays a role in cell adhesion and migration. Another role of DCL-1 could be the participation in endocytosis and subsequent targeting to lysosomes. DCL-1 was also put in connections with various pathologies in last few years Experiments described in this work can be divided into three sections. In the first part I dealt with production of a protein construct based on the extracellular domain of DCL-1. The protein was produced in M9 minimal medium with the only source of nitrogen 15 NH4Cl and the only source of carbon 13 C glucose. The result was 15 N,13 C labelled protein, used for NMR measurements. The second part is dedicated to the analysis of NMR spectra, which enabled us to assign frequencies of protein backbone and aliphatic side chains atoms. On the base of the backbone...

TITLE Use of non-traditional aids in physical education classes at elementary school AUTHOR Eliška Pospíšilová DEPARTMENT Department of Physical Education SUPERVISOR PaedDr. Ivan Přibyl ABSTRACT The diploma thesis deals with the use of non-traditional aids in physical education classes at primary school, both from the point of view of physical education teachers and primary school pupils. The theoretical part is focused on the definition of basic concepts on the given issue, the Framework Education Program for basic education and movement games using non-traditional aids. The practical part is based on a questionnaire survey with physical education teachers and interviews with pupils. The task of the research is to find out whether physical education teachers at the first level of primary schools in Prague 4 use non-traditional aids in their classes and if not, what are the reasons for doing so. The aim of the diploma thesis is to evaluate the results of the research and to summarize the contribution and the negatives of using non-traditional aids in the classes of physical education for pupils. Part of the diploma thesis is "list of games with the use of non-traditional aids" which are verified in practice and supplemented by a final reflection. KEYWORDS: Non-traditional aids, traditional aids, physical...
