Changes of the content of some active substances in aplle and pear fruits during storage

In presented bachelor thesis low molecular substances antioxidants found in plant food were described. Antioxidants from apples and pears were studied in detail, particularly their changes during storage in different conditions. Fruit species were described more closely in this study. The theoretical part is focused on ripening and storage of the fruits. The most common diseases of apples and pears are introduced too. In the practical part changes of some antioxidant levels in apples Idared (red-yellow) and Golden Delicious (green-yellow) were analyzed. Apples were stored for 2 months in three various storage conditions – at standard ambient temperature in the lab, in cellar and in refrigerator. In regular intervals samples were taken and levels of low molecular antioxidants (vitamin C, total phenolics, total flavonoids) and total antioxidant activity in apple fruits were analysed and compared. A surface microflora of the apples was analysed during the whole storage time. The most suitable storage conditions exhibited cellar (9 °C, 38,4 %). Phenolic changes in apples were substantially lower than flavonoid changes, while ascorbate content decreased dramatically with the storage time. Antioxidant activity was related mainly to ascorbate levels, influence of flavonoid changes was observed too. Antioxidant parameters were analyzed also in pear fruits, whose were stored only for very short time.

This diploma thesis was focused on study of low-molecular antioxidants present in various kinds of fruits, namely berries. In theoretical part detailed characterization of antioxidants studied in presented work and their biological effects are introduced. Further, principals of processing and storage of specified species of fruits are discussed. Changes observed during storage of specific fruits are described too. In practical part changes in content of selected antioxidants in berries were followed. As studied material strawberries, raspberries, blueberries, red and black currants were used. All kind of these fruits was stored in fridge for six months using several different kind of processing. Berries were stored as whole fruits, whole osmotically processed fruits by sucrose, raw pulp and sweetened pulp. At first, starting values of selected antioxidant parameters were measured in fresh fruits (ascorbate, total antioxidant activity, total phenolics, catechins and individual flavonoids). After freezing the same parameters were measured regularly in two month periods. As the most suitable method of fruit processing, with regard to antioxidant content, storage of the whole fruits or pulp can be recommended. Content of ascorbate decreased in all forms and species of the processed fruits during all the time of storage. Oppositely, the content of other studied antioxidants exhibited fluctuations or increasing trend. Sensory analysis was proved for the whole fruits and also for whole sweetened fruits.
Use of coaching as an educational method in business organizations

Considering the frequent and fast changes, technology development, new processes and globalization, education becomes more and more important. It impacts not only individuals but also organizations that need to have high quality employees who will be able to react to the changes and adapt to new conditions. To ensure process effectiveness, it is necessary to select appropriate educational methods when educating employees. This thesis deals with coaching, and its aim is to evaluate the possibility of using coaching as an educational method in business organization. In the theoretical part, the comparative and content-analysis methods were used and in the empirical part the data were collected through questionnaire and semi-structured and unstructured interviews. Research was executed among employees of business organizations, human resources personnel and managers who had experience with coaching. It was supplemented by an interview with a professional coach who provides coaching in organizations. The goal of the survey was to identify benefits of coaching for employee education and to detect its potential constraints. Milne method and phase-amplitude method

Bound and quasi-bound states of one-dimensional systems play an important role in quantum physics. In this thesis we solve the Schrödinger equation numerically via the Milne method. We show the relation between the Schrödinger and the Milne equation. The quantum action and the quantum number function are defined. We find bound-state energies of chosen symmetrical and unsymmetrical potentials. Dependence of bound- state energies on the number of points and the step size is studied. We also analyze the influence of the choice of trial energies and the number of iterations. Then we modify the Milne equation and using the phase-amplitude method we illustrate the search of resonances on the example of a diatomic molecule. 1
Application of the method RFLP PCR , PCR ARMS and reverse hybridization for detecting the most common thrombophilic mutations in the Czech population

Recently the term thrombophilia is becoming a current topic. It is an increased precondition for the creation of tromboses and the following complications as the result of increased blood coagulation which could be very serious or even fatal. A timely diagnosis of thrombophilia mutations helps patients to avoid many critical situations (e.g. in case of operations, pregnancy etc.). In these days there are at least 30 genetic laboratories in the Czech Republic which research thrombophilia mutations. Unfortunately this medical examination is covered by insurance companies just in case of indicated symptoms which are under strict criteria. If someone is interested in a medical examination and a doctor does not indicate him with the symptoms, they are forced to pay the examination on their own. There are a few insurance companies which contribute with some amount for these examinations as a part of their preventive programmes (e.g. Česká průmyslová zdravotní pojišťovna, Zdravotní pojišťovna ministerstva vnitra ČR or Zdravotní pojišťovnu Revírní bratrská pokladna). The aim of my bachelor thesis was a practical mastery RFLP-PCR method, PCR ARMS method and a reverse hybridization on strips to detect the most common mutations of thrombophilia in the population of the Czech Republic and deviloving on the issue. In the theoretical part, I focus on a description of the most important thrombophilic mutations in the Czech population and their risks. I also describe particular genetic methods by which each of thrombophilia mutations is being examined in genetic laboratories. My own results of the examination of four major thrombophilia mutations and polymorphs are shown in the practical part. These are the Leiden mutation, prothrombin mutation, MTHFR 1298 and MTHFR 677 polymorphs. For my examinations I used these genetic methods: RFLP-PCR method, PCR ARMS method, reverse hybridization on strips. I made all these methods in the genetic laboratory Genlabs in České Budějovice. For the examinations I used DNA samples of my family.
