The immune response of mice against avian schistosomes: from local to systemic perspective
Majer, Martin ; Macháček, Tomáš (advisor) ; Kverka, Miloslav (referee) ; Sombetzki, Martina (referee)
In the host bodies, helminths often undergo complicated migrations that afflict various organs. Defining the immune response is then usually focused only on isolated spatio-temporal fragments but lacks the whole-body context. To broaden the knowledge about the tissue-specific and systemic immune response, this thesis focused on mice infected with two model species of avian schistosomes of the genus Trichobilharzia. T. regenti penetrates the skin and enters the spinal cord within a few days. Most of the parasites die in the skin which is accompanied by an influx of leukocytes. We confirmed that it was partially mirrored also in skin draining lymph nodes by increasing the T helper 2 cells proportion. Interestingly, there were reduced numbers of T cells in the spleen with an increased proportion of the regulatory T cells. Therefore, we conclude that adaptive systemic response differed and the regulatory phenotype dominated. However, the skin phase-induced eosinophilia in blood influenced the milieu of the central nervous system (CNS). Schistosomula which entered the spinal cord triggered inflammation and enhanced expression of eotaxins, which attracted the eosinophils from the blood into the CNS. The antigen-presenting cells contribute to the communication of CNS with the periphery. We employed and...
The effect of amyloid beta on viability of selected neuropathogens: challenging the infectious hypothesis of Alzheimer disease
Vosála, Ondřej ; Macháček, Tomáš (advisor) ; Černý, Jan (referee)
Alzheimer's disease is the leading cause of dementia worldwide, affecting millions of individuals. Researchers have identified amyloid beta (A ) as the primary agent responsible for neuropathogenicity, but the factors triggering its overproduction remain uncertain. An relatively new theory, known as the infectious hypothesis, sheds light on this phenomenon by associating the excess production of A with its potential role as an antimicrobial peptide-an element of the innate immune response against neuroinfections. This hypothesis posits a transition from acute physiological overproduction during neuroinfection to a chronically dysregulated and pathological process targeting both pathogens and neural tissue. So far, experimental evidence supporting this theory has been primarily obtained through studies involving viruses or bacteria. Aim of this study was to investigate the role of multicellular pathogens in stimulating A overproduction and, on the other hand, A role in elimination of such infections. Toxocara canis was chosen as a main model of neuropathogenic parasite. Although successful infections were achieved in BALB/c mice, there was no observable increase in A production even after 16 weeks of infection. To further challenge the survivability of Toxocara larvae in increased A concentration in...
The skin immune response of mice infected with avian schistosomes
Revalová, Alena ; Macháček, Tomáš (advisor) ; Kolářová, Libuše (referee)
Invasion of the mammalian skin by cercariae of avian schistosomes of the genus Trichobilharzia is associated with the skin pathology called cercarial dermatitis (CD). Although the manifestation of CD, characterized by the presence of maculo-papular rash and pruritus, is believed to be linked with the previous sensitization, the role of the infection dose and the specific causative agent is rarely considered. Therefore the skin immune response in mouse pinnae infected by 100, 1000 or 4x100 (repeated infections) cercariae of two Trichobilharzia species - T. regenti and T. szidati - was examined in this work. To compare the effect of these factors (the infection dose and species), a complex approach was ensured by employing both in vivo and in vitro methods. Firstly, histopathological changes in the pinnae were examined by classical histology and infiltrated leukocytes were characterized by flow cytometry. Sera of mice were used to detect the systemic production of cytokines and parasite-specific antibodies. In vitro cultivation of mouse pinnae was used to detect the local production of cytokines. The most remarkable observation was that the infection dose largely impacted the course of the immune response, both locally and systematically. In primary infected mouse pinnae, 100 cercariae of both...
Immune response against skin-penetrating helminths with a focus on schistosomes
Revalová, Alena ; Macháček, Tomáš (advisor) ; Leštinová, Tereza (referee)
Breaching the vertebrate skin and overcoming the local immunity represents a critical step in the life cycles of many helminths. This bachelor thesis summarized the current knowledge of the skin immune response against schistosomes. Both innate and adaptive immune mechanisms are activated soon after the infection. Despite certain differences between mice and humans, complement, granulocytes and especially CD4+ T-lymphocytes are considered as key players in anti-schistosomal immunity of both species. However, several aspects of the host immune response, such as the initial source of cytokine IL-4, IL-10 or expression pattern of certain co- receptors remain unclear and warrant further research. A comprehensive understanding of the host immune response in the skin as well as the respective parasite immune evasion strategies is needed to boost vaccine development. Keywords: immune response, skin, lymph nodes, helminths, schistosomes
