Forensic Analysis of Biological Traces and its Use in High-school Education

This master thesis deals with forensic genetics. Primarily is forensic genetics put into the historical context. Then, there is described a biological material and the possibility of detecting biological traces. Further work is focused on the analysis of DNA, while there are elaborated in detail the techniques used as nucleic acid isolation, quantification of DNA, as well as types of polymerase chain reaction (PCR). Also the thesis deals with methods of obtaining DNA profiles - electrophoresis and sequencing and their variants. Finally, the thesis concern the issues of interpretation of forensic genetic data. Forensic biological exercises for high school students are processed in the didactic part. It has been tested in practice in biological seminars at Jiráskovo gymnázium in Náchod. Karyotype analysis of mygalomorph spiders of the families Hexathelidae and Dipluridae

Cytogenetics of spiders of the infraorder Mygalomorphae is largely unknown. My thesis is focused on the karyotype evolution of spiders of families Hexathelidae Dipluridae, which are basal groups of the superfamily Avicularioidea. In this thesis the evolution of diploid chromosome number, chromosome morfology, and also sex chromosomes are studied. The karyotypes of the mygalomorphs are quite interesting for their diversity. Many members of the family Hexathelidae exhibit extreme systems of a chromosomal sex determination which are composed by many gonosomes X. The meiotic division of males is analyzed. Some species exhibit modifications of this division such as achiazmatic meiosis. Selected markers have been detected by fluorescence in situ hybridization. Main aim of the thesis it to analyse and evaluate the visit rate at Vyšehrad and also its revenues from statistics, find out possible weaknesses and propose a solution.
