The tri-band fluorescent reader

This bachelor thesis deals with the issue of laboratory testing and design of an optical instrument called a tri-band fluorescent reader, which is designed for fluorophores Alexa Fluor 488, Alexa Fluor 555 and Alexa Fluor 647. It contains a theoretical part with key knowledge not only in the natural sciences, such as physics, chemistry, biology, but also the current technical capabilities that are necessary for the practical part. In the practical part is drafting the objectives of the thesis, which are the measurement and evaluation of the samples examined using a monochromator, acquisition and evaluation of images using the existing system, build spectral analysis throughput, create a simulation of custom solutions and create a system engineering design. The mechanical model of the reader assembly was made after measuring the samples and the device and creating the simulation.
Design of mechatronic system with electromagnetic damper for presentation of oscillation

Dynamic Characteristics of Exhaust System and Noise Transmition to the Car Cocpit

This thesis deals with the complex analysis of dynamic characteristics of the exhaust system and the transmission of noise from exhaust system to the vehicle. In the first part is performed modal analysis, which is compared to experimentally measured data. Modal analysis of the complete exhaust system is made with focusing on the influence of temperature of the exhaust system. There is also performed an analysis of noise transmission from exhaust system to the car cockpit.
Auxiliary winding supplying the voltage regulator of synchronous generator

The theme of this thesis is the design and verification of the auxiliary winding to supply the voltage regulator of the synchronous generator. The synchronous generator for which the auxiliary winding proposed is three-phase, low voltage with double layer random wound stator winding. The generator excitation is brushless with auxiliary rotating exciter. The auxiliary winding is designed for placement in the stator teeth, along with the main winding. The sizing of the auxiliary winding is derived from the power requirements the voltage regulator. Draft winding is validated analytical calculation and finite element method (FEM). The auxiliary winding has been implemented in the production of the generator. The Features of the auxiliary winding have been verified by measuring the by electrical testing room and the test results are evaluated.
Universal controller for car or motorcycle dynamo

The thesis deals with the issue of design of an electronic controller of charging for historical cars using a dynamo. It contains theoretical analysis of the machine itself (dynamo) and analysis of a shunt wound dynamo and a seperately excited dynamo. It~is also focused on possible usage of a dynamo in various vehicles. The work compares industrially produced controllers with amateur manufacture as well as confronts their positives and negatives. All requests for the implemented controller are deduced from the analysis of the industrial and amateur controllers. The thesis consists of process plan of controller connection including the final working version and also the project and realization of a PCB as well as following description of putting the controller into practice and installation in the car. The thesis includes the project of current and voltage controllers, following adjustment and operation tests with use of the real device.
Intracellular Calcium Measurement Methods

This work aims to focus on the issue of intracellular calcium. Not only the theory dealing with the history of fluorescent indicators but also the types of fluorescent indicators and methods of measurements are summarized. The individual steps of measurements on the fluorescent microscope are mentioned as well. There is also created a program for evaluting Ca2+ concentrations in cells made in matlab.
Tractor Internal Acoustics

The diploma thesis focuses on the analysis of the internal acoustics of a tractor using the finite element method. The thesis also contains the analysis of excitation effects from individual sources of vibrations and evaluating the response in the internal acoustic chamber with special focus on the driver’s head. The conclusion should suggest appropriate structural modifications to reduce interior noise.
T cells labelling by bimodal contrast agent for in vivo studies of stroke

Stroke is a serious brain injury, which causes sudden death or terminates in permanent neurological disability. Nowadays, tissue plasminogen activator (tPA) is used as the only effective treatment of stroke. One of the potential targets for novel therapy are T cells. Even though the explicit role of T cells in the pathogenesis of brain injury, amounts and timing of all T cell subtypes infiltrating into brain during the stroke still needs further investigation. The research in this field is complicated by the lack of efficient methods for in vivo cell tracking. Therefore the aim of this thesis was to develop a method of T cells labelling by MRI contrast agent in order to investigate T cells distribution in ischemic mice model using in vivo MR imaging. T cells were isolated from C57/BL6 mice in two step isolation protocol using gradient centrifugation and magnetic separation with the efficiency of 97 %. The isolated cells were labelled with 100 μg Fe/mL of Molday ION Rhodamine B contrast agent. The labelling efficiency after 17 hours of cells incubation was higher than 99 %. The labelled cells were cultured with CD3 and CD28 antibodies resulting into the 74 % viability of labelled T cells compared to 83 % viability of non labelled T cells.
