Production of extracellular polymeric substances by Aureobasidium pullulans
Horáček, Pavel ; Breierová, Emília (referee) ; Márová, Ivana (advisor)
The diploma thesis is focused on the study of the influence of cultivation conditions and arrangement for the production of extracellular polymeric substances by using yeast-like microorganism Aureobasidium pullulans. In the theoretical part a brief description of A. pullulans, its use in biotechnology and produced exobiopolymers, especially pullulan and poly-L-malic acid are presented. The first aim of the experimental part was to set the most appropriate cultivation conditions for A. pullulans CCM 8182. Growth and production properties in optimum conditions were compared with cultivation on waste substrates - oat bran, buckwheat husks, apple fiber and others. Waste substrates can be used as cheap nutrient sources which enable reducing cost of potential biotechnological production. As a further part of this work, optimization of HPLC/RI method for analysis of exobiopolymers has been done. Optimal mobile phase composition and chromatography conditions were proposed. Column Roa organic acid H+ was the most suitable for simultaneous separartion of glucose and malic acid. Before HPLC analysis hydrolysis of polymers was done. Sulphuric acid (5 mmol/L) was used as a mobile phase at flow rate 0.5 mL/min and temperature 60 °C. The highest production of pullulan occurred using oat bran as a substarate (13.03 g/L) at an initial pH 7.5. Maximum production of poly-L-malic acid was observed during the cultivation on apple peels (2.89 g/L) at pH 6. It was found that the higher production of poly-L-malic acid occurred at pH 6, while higher production of pullulan was at pH 7.5.
Antibacterial gels and their effectivity
Horáček, Pavel ; Vítová, Eva (referee) ; Vránová, Dana (advisor)
The Bachelor thesis focuses on the evaluation of the effectiveness of disinfectants and antibacterial hand gels, which are a good complement to personal hygiene and other factors to prevent various diseases, transmitted through the hands. A brief description of skin structure, classification of microflora on the hands, including a list of potential pathogenic microorganisms located on human skin and causing many diseases can be found in the theoretical part of this work. Content of the experimental part of this work was determination the effectiveness of tested antibacterial agents by indirect determination of the number of microorganisms on naturally contaminated hands by the cultivation method. Experimental part of this work evaluates the characteristics of the tested products by potential users in the form of sociological research.
Analyzing BitTorrent Behavior
Horáček, Pavel ; Grégr, Matěj (referee) ; Matoušek, Petr (advisor)
This thesis is focused on analysis of the BitTorrent network, from theoretical view point by analyzing protocols used by this network, as well as practical view by inspecting individual clients implementing these protocols. The aim of this thesis was to develop a monitoring method used to detect communication used by this network. Due to possible illegal data sharing as well as unwanted usage of the network bandwidth, it's important for network administrators to detect this kind of communication. Another aim of this thesis was to familiarize the reader with real communication used by clients that implement protocols used in the BitTorrent network. This thesis contains descriptions of individual phases used in three selected clients, as well as three experiments performed with one of these clients.

Ryan by Chris Landreth
Šafus, Martin ; PIXA, Libor (advisor) ; Horáček, Pavel (referee)
The thesis synthetizes traceable information about Chris Landreth’s partly biographical film Ryan, as well as summarizes events preceding and concerning its creation. In the second part of the thesis I analyze especially psychorealism that links to the personality of Chris Landreth, but lacks clearly formulated definition. The first part emphasizes information quality about the author Ryan Lrkin and understanding of circumstances of the film creation. In the second part I put the film through analysis and then search for analogies with other episodes. Using subsequent synthesis I search for new evidence. It results in an attempt to define psychorealism. In order to familiarize with artistic movements and their means it is essential to describe them theoretically and psychorealism waved spluttered in void. I intent to change it with this thesis.

Klíčové kompetence z pohledu žáků vybrané střední odborné školy
Horáček, Pavel
My bachelor thesis deals with key competencies in relation to pupils' opinions. The theoretical part is focused on the definition of key competencies, specifically in relation to employability and entrepreneurial activities. The main aim of the practical part of the thesis is to explore the views of pupils from the third grade of selected secondary vocational school, who attend The serviceman of motor vehi-cles branch and The body repairmen will receive vocational certificate, about acquiring competencies for the labor market and business activities. Scientific sources were collected and methods such as the creation of excerpts, analysis, comparison and synthesis were used for processing the theoretical part of the work. I conducted interviews as the exploratory method and the questionnaire was assembled as an instrument whose aim was to measure the above mentioned respondents' views about acquiring of key competencies for the labor market, employability and business activities as well as the perceiverance of the level of acquired competencies, whose adoption would lead to greater self-confidence when applying for a job or when starting own business activities.
