Charles University in Prague University Roma Tre Faculty of Pharmacy in Hradec Králové Department of Biology Department of Biochemical Sciences Candidate: Jan Hájek Supervisor: PharmDr. Iva Boušová, Ph.D. Prof. Sandra Incerpi, Prof. Jens Z. Pedersen Title of diploma thesis: Biological activity of antioxidant compounds in monocytes THP-1 Elaboration of this work is based on the experiments carried out in the Laboratory of Physiology at University Roma Tre in Rome, under the supervision of Prof. Sandra Incerpi. The team of this laboratory studies the antioxidants of different structures and evaluates their ability to prevent the production of reactive oxygen species (ROS). These reactive species are essential for many physiological processes, but they can also have deleterious effects and participate in the development of various diseases. In this work, I dealt with a study of antioxidant properties of different polyphenolic compounds (mosloflavone, negletein, 5,6-dihydroxyflavone, baicalein, gallic acid, and compound 1625) in the cell culture of human monocytes THP-1 and also in the in vitro test. Three different methods were used: intracellular ROS determination (dichlorofluorescein assay), cytotoxicity assay, and electron paramagnetic resonance spectroscopy (EPR). Mosloflavone showed scavenging...
Ekonomická sladěnost České republiky s eurozónou ve světle finanční krize
Hájek, Jan ; Mertlík, Pavel (advisor) ; Cingl, Lubomír (referee)
This paper deals with the issue of synchronization of the Czech economy and the economy of the eurozone. The OCA theory which is key theoretical basis for the analysis contained in the paper is briefly described in the first part. In the second part cyclical and structural synchronization is evaluated by the means of correlation coefficients of the development of GDP, industrial production, inflation and interest rates. The obtained results show sufficient level of synchronization, impacts of the financial crisis also rather contributed to the current level of synchronization. Arguments for and against synchronization are discussed in the third part as well as mutual dependency of the both economies. Two quarters time lag of the Czech GDP growth rate behind the eurozone is discovered in the pre-crisis period. At the end OCA- index for the Czech Republic is recalculated, the value of the OCA-index for the Czech Republic is the lowest among tested countries.
Synthesis of ellipticine and its pharmacologically more efficient derivative 13-hydroxyellipticine
Hájek, Jan ; Stiborová, Marie (advisor) ; Černá, Věra (referee)
Ellipticine (5,11-dimethyl-6H-pyrido[4,3-b]carbazole) is plant alcaloid isolated from Ochrosia elliptica1) (Apocyanaceae). Ellipticine's anticancer effect is mainly caused by its intercalation into DNA, formation of covalent adduct with DNA and inhibition of topoisomerase II. Known syntheses of ellipticine are based on chemicals containing two or three aromatic cycles that are used in cyklisation and/or addition reaction to get ellipticine or ellipticine derivates. 13- hydroxyellipticine can be prepared de novo as a product of casual ellipticine synthesis, or as a modification of already synthetised ellipticine. Keywords: ellipticine; synthesis; derivates of ellipticine

Estimating the quadratic almost ideal demand system and the effects of population ageing in the Czech Republic
Bílý, Michael ; Janský, Petr (advisor) ; Hájek, Jan (referee)
The aim of this thesis is to estimate the Quadratic Almost Ideal Demand System of non-durable goods on the Czech Republic Household Budget Survey data in two distinct models. Respective income, own and cross price elasticities are calculated and we interpret the resulting income, price and demographic characteristics of the household demand. The first model is used to estimate the effects of the 2015 Value Added Tax reform in the Czech Republic, which introduced 10% reduced rate on medication and books. We estimate how much the reform changes the consumption behaviour of households, the biggest changes are in the commodities of clothing, books, medications and household goods. The households are estimated to spend 1,049 billion CZK more on non-durable goods and the VAT revenue is estimated to decrease by 818 million CZK. The second model is used to predict the effects of the population ageing on the household demand. We do this in four scenarios to separate effects of the household composition change and to analyse the composed effects of increasing wealth and redistribution among the working and retired households. The population ageing has the highest impact on the commodities of food, fuel and light, transport and leisure services. The shift in composition affects the consumption mostly in the same...
The Prague and it's logistic support. Transformation of the city and it's technical networks in the years 1913-1952.

Kryštof Drnek The City of Prague and it's logistic support. Transformation of the city and it's technical networks in the years 1913-1952 Abstract The Prague's evolution into capitol of the new republic is divided into three stages, split by two wars. The progress of the city begun deep in the era of the Austria-Hungary imperium - in the middle of the 19th century, when the new town council was elected from czech townsman upper classes. It led into the process started with series of construction and administration projects. The city was upgraded with new bridges, railwaystations. On the place of the fortified walls, which were cancelled in the year 1866, shortly after Prussian- Austrian war, was quickly built piles of houses that bound inner Prague with it's suburbs. The administration were trying to connect the Prague's suburbs with the inner city but only few of them this procesess accepted. The foursome of the inner satellites (Žižkov, Karlín, Královské Vinohrady and Smíchov1) this projects declined and until the World War I remained on their own. Building of the logistic sites city underwent from the middle of the 19th century. First two logistic sites, the water supply system and the sewer system, were ancient systems supplying the city from the Middle Age. As the city was growing, the systems weren't...
The agrarian share sugar refinery Němčice nad Hanou (1910-1948)
Lecián, Petr ; Jakubec, Ivan (advisor) ; Hájek, Jan (referee)
The purpose of my paper was to outline the time of appearance and development of the sugar refinery industry in both central and northern Moravia and Silesia on the basis of the historical and current literature and utilisation of the archive funds of MZA in Bmo. The thesis covers a time period from the first half of 19th century till the year 1948 when the sugar refineries were nationalised. I have divided my paper into several chapters following the establishment of single sugar refineries in Moravia and Silesia. The first half of the paper is focused on the attempts of sugar production from the maple juice or the sugar beat. The sugar production expansion started by the foundation ofthe first sugar refinery in Kostelni Vydfi in the year 1829 and by the continuous sugar production development starting with the aristocratic establishments, the enterprises owned by the wealthy entrepreneurs and following up with the small fanner's stock sugar refineries, whose stockholders were the Czech fanners. The second part of this paper is dedicated to one of the most significant and most modem small farmer's sugar factories in Moravia, which was located in Nemcice nad Hanou. It was the first small fanner's sugar factory with its own refinery for the white sugar production. There is also one chapter clarifying the...
Change of the Landscape and Life in the Time of Industrial Revolution on Šumava
Blažková, Tereza ; Matoušek, Václav (advisor) ; Hájek, Jan (referee)
5 ABSTRACT This thesis was written to contribute to study of the post-medieval cultural landscape, especially the industrial landscape. It observes changes of landscape and settlement, which were affected by industrial activities in the middle of Šumava Mountain region. The reason why the landscape pattern has changed were industrial activities and also fabrication of Vchincko-tetovský floating channel at the turn of the 18th and 19th century. Forests in this region were previously used mainly by glass and iron works, but the industrial activities and settlement culminated in the exploitation of the forests just because of oportunity of the wood floating. Formating and function of the channel was business intention of prince Schwarzenberg, who could due to floating of Vydra river and then other two rivers called Otava and Vltava fill the gap in the market with firewood in Prague. The landscape was already in the first decennium of the 19th century reformated negatively, namely in forest crops. The number of colonies of woodworkers increased. The problems came in 1870's with windbreake and bark beetle calamities. Crisis and termination of the wood floating in the region came on the first half of 20th century in connection with world war and political changes. But the landscape and settlement experienced...

Electricity market: Analysis and prediction of volatility Abstract Vladimír Kunc July 30, 2015 The last two decades can be characterized by restructuring of energy industry and the creation of new, competitive energy markets, where accurate forecasts of elec- tricity prices and price volatility are valuable both to consumers and producers. The aim of this work is to analyse several models for prediction of the price volatility of electricity on the Czech Electricity Day-ahead market on price data provided by OTE, a.s. for years 2009-2014. This work compares 144 different models' configura- tions for three distinct classes of models - autoregressive models, GARCH models, and artificial neural network models. This work provides comparison based on five different criteria, each describing the model in different way. Keywords: price prediction, volatility prediction, GARCH, neural networks, LSTM 1
Development of German Alpine Clubs in Czech Lands until 1938
Chaloupská, Pavlína ; Waic, Marek (advisor) ; Štumbauer, Jan (referee) ; Hájek, Jan (referee)
Mgr. Pavlína Chaloupská, abstrakt disertační práce Abstract: The thesis analyzes activities of the most important German Alpine clubs operating in the Czech Lands and interwar Czechoslovakia since their establishment until 1938. The history of alpine clubs is explained, and the organization of DuÖAV (Deutscher und Österreichischer Alpenverein, German and Austrian Alpine Association) is analysed, on the basis of general and specific introductory information related to the alpinism in the late 19th century. The thesis is divided according to geographical location of the constituent sections. For each section of DuÖAV, the circumstances of its establishment are monitored, the sphere of its activity is defined, the internal organization is described, and the economic functioning is analyzed. The activities of Alpine clubs in Bohemia and Alps are charted in detail, and based on the research it was discovered that the most important activity of the Alpine clubs was the construction of the Alpine huts, owned by German clubs from Bohemia. Among other activities there were organization of guiding and rescue operations, giving lectures, balls and excursions in their locality. Two chapters are devoted to the most important personalities of the German alpinism in the Czech lands: Johann Stüdl and Rudolf Kauschka.
