Methods and consequences of some neurosurgical interventions in animal model and in human clinic. Pinealectomy and intracerebroventricular application.

Clinical surgical techniques of pinealectomy and intracerebroventricular (i.c.v) cannulation are still used in the clinic in indicated cases. In research, there is an effort to replace these classical surgical techniques by other means. However, these various new models often do not reflect the complexity of the functions taking place in the living organism as a whole. The thesis shows that these surgical techniques should be also a part of biomedical research in the future, as they still bring new important information. In the experiment, the pinealectomy can serve as a model of resection or depletion (removal of natural hormone secretion), while i.c.v. application of the active substance to the ventricles on the contrary as a model of addition or substitution. The practical part of the thesis is divided into 4 areas: A) pinealectomy - an animal models, B) pinealectomy - in the experiment, C) pinealectomy - in the clinical practice and D) intracerebroventricular application - in the experiment. The work describes in detail the surgical techniques and discuss possible consequences of pinealectomy in 6 animal species (and their comparison) and in humans. The influence of corticosterone and corticoliberin on damage of the hippocampus and their relation to cognition

1 ABSTRACT Dissertation deals with the influence of corticoliberin and corticosterone on behavior, memory and learning and the histomorphological changes in the hippocampus. The experiment was performed on rats - long- evans strain, line AV ČR, Krč (male, age ca three months, weight ca 350g). The hormones used were administrated continuously; corticoliberin by the osmotic pump into the right lateral cerebral ventricle for four weeks, corticosterone by the subcutaneous pellet for three weeks. The animals were divided into three experimental groups and one control group. The group [↑CRH] (n=6) osmotic pump-applied corticoliberin (1,5 µg per animal per day), it was made adrenalectomy with substitution by corticosterone subcutaneous pellet (1,4 mg per animal per day). The group [↑CRH↑CS] (n=11) osmotic pump-applied corticoliberin (1,5 µg per animal per day). The third experimental group [↑CS] (n=10) corticosterone was administered by subcutaneous pellet (9,52 mg per animal per day). The control group (n=10) was without any treatment. All groups were subjected to four various behavioral tests - test of active allothetic place avoidance (AAPA), step- through test, conditioned taste aversion (CTA), and test in Morris' water maze (MWM). The influence of corticosterone and corticoliberin on damage of the hippocampus and their relation to cognition

Dissertation "The influence of corticosterone and corticoliberin on damage of the hippocampus and their relation to cognition" deals with the cognitive, behavioral and histological changes in experimental rat strain long-evans that closer describe the consequences of long-term continuous application of corticoliberin and/or corticosterone. Testing of the behavioral changes was divided into two phases. The first one - within three or fourweeks respectively administration of these hormones, therefore until their early effects - and the second phase - after four weeks of completion of the first phase at the time of the possible late effects. In the twelfth week the experimental animals were killed and in the group which had exogenously elevated corticosterone, the morphological changes in the hippocampus were monitored and measured. In all experimental groups alteration of behavior was observed. Histological and morphological changes in the brain we have found. Layout of experiments in two testing phases allowed differentiation of the early changes and the late and persistent changes. The diploma thesis deals with sociology of province through village Vedrovice placed in South Moravia. The area of sociology of province is quite extensive so the thesis is focused on issues of selected areas of villagers's life. Available sources of technical literature and literary overview of current knowledge in rural areas were valorized in the first part. The next part deals with the village itself. This part is processed through the study of documents and own empirical research. The issues between old-time villagers and new immigrant residents were examined by interviews and questionnaires. The results and the evaluation of hypothese are processed by graphs and charts. The last part includes suggestions and recommendations.
