1 ABSTRAKT Práce se zabývá vlivem kortikoliberinu a kortikosteronu na chování, paměť a učení a na histomorfologické změny hipokampu. Pokus byl proveden na potkanech - samcích kmene long-evans, linie AVČR, Krč - stáří cca 3 měsíce, váha kolem 350g. Zkoumané látky byly aplikovány kontinuálně; kortikoliberin osmotickou pumpou do pravé postranní mozkové komory po dobu 4 týdnů, kortikosteron subkutánní peletou po tři týdny. Zvířata byla rozdělena do tří experimentálních skupin a skupiny kontrolní. Skupina [↑CRH] (n=6) osmotickou pumpou podaný kortikoliberin (1,5 µg na zvíře a den), provedena adrenalektomie, substituce kortikosteronem podkožní peletou (1,4 mg na zvíře a den). Skupina [↑CRH↑CS] (n=11) osmotickou pumpou podaný kortikoliberin (1,5 µg na zvíře a den). Třetí skupina [↑CS] (n=10) měla kortikosteron podaný subkutánní peletou (9,52 mg na zvíře a den). Skupina kontrolní (n=10) bez zásahu. Všechny skupiny byly podrobeny čtyřem různým behaviorálním testům - test aktivního alotetického vyhýbání se místu (AAPA), step-through, test podmíněné chuťové averze (CTA), a testu v Morrisově vodním bludišti (MWM). U skupiny [↑CRH] bylo nalezeno prodloužení habituační doby způsobené anxietou a prodloužení doby učení a též porušení dlouhodobé paměti. U skupiny [↑CRH↑CS] byla narušena především kognitivní koordinace a... 1 ABSTRACT Dissertation deals with the influence of corticoliberin and corticosterone on behavior, memory and learning and the histomorphological changes in the hippocampus. The experiment was performed on rats - long- evans strain, line AV ČR, Krč (male, age ca three months, weight ca 350g). The hormones used were administrated continuously; corticoliberin by the osmotic pump into the right lateral cerebral ventricle for four weeks, corticosterone by the subcutaneous pellet for three weeks. The animals were divided into three experimental groups and one control group. The group [↑CRH] (n=6) osmotic pump-applied corticoliberin (1,5 µg per animal per day), it was made adrenalectomy with substitution by corticosterone subcutaneous pellet (1,4 mg per animal per day). The group [↑CRH↑CS] (n=11) osmotic pump-applied corticoliberin (1,5 µg per animal per day). The third experimental group [↑CS] (n=10) corticosterone was administered by subcutaneous pellet (9,52 mg per animal per day). The control group (n=10) was without any treatment. All groups were subjected to four various behavioral tests - test of active allothetic place avoidance (AAPA), step- through test, conditioned taste aversion (CTA), and test in Morris' water maze (MWM).
