Lyofilizace lipozomů pro perorální aplikaci léčiv
Vacková, Michaela ; Šklubalová, Zdeňka (advisor) ; Dittrich, Milan (referee)
The protein or peptide substances are not appropriate for oral application. One of the many obstacles, is an aggressive environment of the gastrointestinal tract. One way to protect these substances is the preparation of liposomes. The aim of this work was preparation of liposomes of the different composition, choice of the optimum aqueous media, and stabilization using freeze-drying. All samples consisted of egg phosphatidylcholine(EPC) and cholesterol (Chol), with the addition of one out of the substances: cholylsarcosine (CS), stearoylamine (SA) and vitamin E (TPGS), respectively, in various ratio. Liposomes were prepared using a film method and manual extrusion through membrane with pore size 200 nm. The liposome size and polydispersity was measured by photon corellation spectroscopy. After the extrusion, the liposomes underwent freeze-drying in the presence of selected appropriate cryoprotectants: sucrose and trehalose, respectively. Stability has been established by evaluating the visual characteristics of the product (the appearance), solubility in the water and measuring of liposome size and polydispersity.

The main goal will be to explore the real opinions of farmers about the project of Farm in the named area and how will be possible to improve the occasions for the target groups - the children, youth, adults (especially unemployed), parents with children (on the maternity or paternity leave) and seniors. The thesis oabout the Farm follows from the reaction on the technical progress these days. The...

Monetary policy in Czechoslovakia 1919 - 1929
Prokeš, Jan ; Szobi, Pavel (advisor) ; Jakubec, Ivan (referee)
This study focuses on describing and analyzing monetary policy in Czechoslovakia between 1919 and 1929. Initially devoted to theoretical definition of monetary policy and approaches to the nature of money. The analytical part describes the most important events of the decade in terms of monetary policy. Particular emphasis is placed primarily on monetary reform, which made Alois Rašín in 1919. The work also describes a change in direction from the deflationary policy to monetary stabilization, on which has the largest merit Engliš, second significant figure in the Czechoslovak monetary policy in those years. As Rašín and Engliš contributed significantly to successful monetary developments in postwar Czechoslovakia. Rašin deal with strong inflationary pressures and gave rise to solid and credible crown. Engliš is behind the most significant economic boom in the history of republic.

Aspects of financing the LBO acquisitions
Durdil, Lukáš ; Jílek, Josef (advisor) ; Rybáček, Václav (referee)
This diploma thesis deals with the acquisition of companies by means of the Leveraged Buyout (LBO) method. This method differs in many respects from the standards M&A. This work examines the specific features of the given business transactions from the viewpoint of the target companies, procedures and entities involved in these transactions. As the key element of the LBO method is the debt financing of the acquisition, the work also deals with this aspect and evaluates the positions of the creditors from the viewpoint of the influence on the stability of the financial system. The thesis further analyses the question whether the LBO method has an effect leading to an increased effectiveness of processes in a company, or whether, conversely, the proceeds of acquisition investors are a consequence of a transfer of values from other entities. The text is based on available theoretical and empirical sources which are supplemented by the author's analyses.

Symmetrical aminophosphinates
Procházková, Soňa ; Kubíček, Vojtěch (advisor) ; Vojtíšek, Pavel (referee)
Title: Symmetrical aminophosphinates Author: Bc. Soňa Procházková Department of Inorganic Chemistry, Faculty of Science, Charles University Supervisor: RNDr. Vojtěch Kubíček, Ph.D. Supervisor's email address: kubicek@natur.cuni.cz ABSTRACT Two types of α-aminophosphinates were prepared and studied: phosphinate analogues of iminodiacetic acid − aminobis(methyl(hydroxymethyl)phosphinic acid) (IDPhm ) and aminobis(methyl(2-carboxyethyl)phosphinic acid) (IDPce ), and bis(aminomethyl- phosphinates) − (hydroxy(fenyl)methylen)bis(aminomethylphosphinic acid) (PheOHABPin) and (methylen)bis(aminomethylphosphinic acid) (ABPin). All prepared compounds were fully characterized (by NMR, MS, elemental analysis). One crystal structure was determined by X-ray singlecrystal analysis. The diphosphinates were synthesized by Mannich type reaction, whereas the bis(aminomethylphosphinates) were prepared by multistep synthesis including reactive silylesters. Compounds were prepared in moderate or good yields Acid-base and coordination properties of ligands were investigated and compared by potentiometric and NMR titrations. The values of protonation constants of aminogroups, which determine stability of complexes, showed surprising results. An extremely low basicity of nitrogen atom was found in diphosphinates. So, these compounds...

Improvement of retroviral vectors for efficient gene transfer
Šenigl, Filip ; Hejnar, Jiří (advisor) ; Šmahel, Michal (referee) ; Ruml, Tomáš (referee)
44 CONCLUSIONS Retroviral vectors are transcriptionally unstable in mammalian cells. The ASLV- derived vectors are the most affected by silencing. We modified the vector by insertion of the CpG island inner element (IE) into the vector LTR in three different positions in either sense or antisense orientation. Each vector variant exhibited a certain extent of stabilization. The vector with insertion of a tandem of two IEs between the enhancer and the promoter was the most stable and exhibited almost no silencing after four months of cultivation in rodent and human cells. The IE comprises two high-affinity Sp1 binding sites. The presence of Sp1 binding sites is important for the protective effect of IE, but at least a part of the entire anti-silencing capacity is maintained in IE with mutated Sp1 sites. We identified the Tvc receptor of ASLV. The tvc gene encodes a 488-amino-acid protein most closely related to mammalian butyrophilins, members of the immunoglobulin protein family. To confirm the identification of the Tvc receptor, we disrupted both tvc alleles in normally susceptible DT40 cells. The DT40 tvc-/- clone was resistant to the ASLV(C) infection. We identified the mutation that results in decreased susceptibility to infection by ASLV subgroups B and D and resistance to ASLV subgroup E of line M...

Slope stability calculations for small dams
Hajda, Jindřich ; Hodák,, Jiří (referee) ; Říha, Jaromír (advisor)
There are approximately 20 000 small dams in the Czech Republic. This number entails many ponds or small flood attenuation reservoirs, which provide protection against floods. We do not know much about the structure and material properties of these earth dikes. Concerning the small dam stability the current technical standard ČSN 75 2410 focuses foremost on the angle of both the upstream and downstream slopes in case of different materials, and the method of the stability assessment in case of a dam height being 6m or more. Most of these earth dikes are still standing in spite not being built according to any technical standards. They do not follow any standards mostly because they were built before the modern technical standards were published. This diploma thesis focuses on assigning the degree of a reserve in the safety factor for studies of 5 homogeneous earth dikes and 3 inhomogeneous earth dikes. The calculation was done using the Plaxis 2D software using to the shear strength reduction method. The Calculation of the safety degree is made for 4 typical load cases.

Comparison of stability and predictability of tax systems in the Czech republic and Ireland
Fortuník, Jan ; Pavel, Jan (advisor) ; Vítek, Leoš (referee)
The aim of this bachelor thesis is to find out whether the Czech tax system is way less stable and less predictable than a tax system of a western European country, particularly Ireland. This is done by comparing the number of changes that took place in the tax systems of these two countries during years 2004 to 2014 and the amount of time available for tax payers to adapt to those changes. Three taxes were chosen for the purpose of this analysis: personal income tax, corporate income tax and value added tax. The result show that the Czech tax system is less stable and predictable than the Irish tax system, but the difference between these two tax systems is not as significant as assumed.
