Determination of ascorbic acid by HILIC approach - a comparison of HILIC stationary phases.

This thesis was focused on the determination of ascorbic acid (AA) using Hydrophilic Interaction Chromatography (HILIC) approach and the comparison of HILIC stationary phases. Determination was carried out using three types of detection. To optimize the method ultraviolet (UV) detection was used. For the determination of AA and DHA (dehydroascorbic acid) on different stationary phases aerosol based detectors Charged Aerosol Detector (CAD) and Nano Quantity Analyte Detector (NQAD) were used. During method optimization suitable conditions for the determination of AA and DHA have been defined. Aqueous part of the mobile phase was formed by water, acetic acid or ammonium acetate buffer of increasing strength and different pH. Organic part was formed by acetonitrile. During the optimization the influence of gradual changes of water and organic part has been monitored. Compared stationary phases were Atlantis T3 and those specially developed for the determination by HILIC method - Luna HILIC and ZIC-HILIC. In the case of Luna HILIC and Atlantis T3 both analytes were not separated from the dead retention time. On ZIC-HILIC column separation of substances occurred, but peaks were too broad. The Application of DNR orders in clinical practise focused on the issue Non providing aid Do Not Resuscitate orders, as a special type of advanced directives, constitutes an impressive framework full of dilemmas, both between the patient's right to autonomy, self-determination and the physician's duty to provide proper medical care and between the patient's right to autonomy and physician's tendency to apply his/her professional medical opinion. As well this issue has not only the medical and the legal aspect but it possess ethical, sociological and even economical ones, indeed. Despite the fact there are some provisions on the regulation of advance directives at the moment available, neither the position of the patient nor the position of the doctor is actually solved. Furthermore, the legal state of advance directives forces the doctors in order to protect themselves to ignore on purpose the knowledge that there are some advance directives being made by the full competent patient. The consequences of not providing aid are rush ones, with a great possibility of imposing criminal liability for commission of one of the offences against person. AGREEMENT ON FUTURE AGREEMENT ACCORDING TO PROVISIONS OF CZECH COMMERCIAL CODE The aim of this thesis was to describe the current statutory provisions relating to an agreement on future agreement according to the Act. No. 513/1991 Coll., the Commercial Code of the Czech Republic, as amended, to distinguish its provisions in comparison with the provisions on agreement on future agreement according to Act no. 40/1964 Coll., the Civil Code, as amended, and to point to certain areas of legislation which are not sufficiently clear and to provide suggestions for dealing with them. The method used was description with the use of available specialist literature and case law (bearing in mind case law is not a source of law in the Czech Republic per se, and form only a supportive role), and in cases where legislation and case law could not provide a sufficient answer, author gave his own opinion on the given matter with his reasoning for it. The agreement on future agreement according to provisions of Czech Commercial Code must, in order to be valid, be made in writing, contain obligation of at least one party to conclude future agreement that must be described at least generally. Barbora Szotáková, Ph.D. Supervisor specialist: RNDr. Věra Králová, Ph.D. Title of diploma thesis: Influence of albendazole on cell proliferation in vitro Albendazole (ABZ) is a benzimidazole anthelmintic widely used for the treatment of parasitic diseases in man and animals. The mechanism of action is based on inhibition of microtubule polymerization by binding of ABZ to the β-subunit of tubulin in parasites. As a result, it subsequently leads to the disruption of vital functions of the parasite and to its death. After elucidation of this mechanism, ABZ was tested as a potential anticancer agent. It has been shown that it is able to inhibit proliferation of some cancer cell lines. In this study, we examined the effect of ABZ on cell proliferation in vitro. For the purpose of our experiment we used cell lines derived from colorectal carcinoma - SW 480 and SW 620 together with a cell line NCM 460 isolated from normal colonic mucosa. The cells were treated with ABZ in the concentration range of 0.01 - 10 µM for 24, 48 and 72 hours. Cell viability was determined using WST-1 assay and Neutral Red Uptake Test (NRU). Water supply without a residual disinfectant

The diploma thesis focuses on the water distribution system functioning without chlorine-based disinfectant. This thesis contains research of documented cases of examples of drinking water distribution systems both abroad and in the Czech Republic, which do not use disinfectants. Moreover, the thesis pay attention to chlorine and its effects on bacteria. The thesis deals with the first assessment of water supply structure, which determines whether the water supply structure is suitable for operating without disinfectant. The thesis describes the process of transition to the disinfectant-free operation and determines the undesired events, which may occur with the ending of the use of disinfectant. As a part of the research, the thesis examines the water supply system in village Kateřinice, to determine if it is suitable for operation without disinfectant.

Determination of floodplain in Kyjovka river at km 71 to km 74

The diploma thesis discusses the current status of watercourse Kyjovka and a proposal for appropriate protection measures of the territory. The selected section starts at river-km 71,338 and ends at river-km 74,443. The selected part of the watercourse runs through the town of Koryčany. This thesis examines the selected section of Kyjovka split into two subsections. That allows for detailed presentation of several proposed appropriate measures to be applied at critical spots in the selected subsections. The calculation of the watercourse model and its capacity with the water level progresses for selected N(y.o.) flow rates were processed in HEC-RAS5.0.1. program. Proposed flood protection measures are incorporated in the drawings.

Pressure loss valves in heating systems

The theoretical part of the thesis deals with the hydraulics of heating systems. The aim of the project is a design of the heating and hot water preparation system for an apartment building. The building has 6 above-ground floors and 2 underground floors. Two different options of heating systems were designed, comparison and results are presented in the thesis. In the first case the heat source during the winter season and for water heating, where chosen two gas condensing boilers, situated in separated room on the second underground floor. In the second case, each apartment has its own gas condensing boiler. The experimental part of the project deals with measurements of pressure losses in water armatures used in heating systems. The aim of this part is the determination of coefficients of added resistance for uncontrolled elements, which are normally presented as tabular value. In thesis are presented results for a ball valve, backflow preventer, filter, for slide valve and thermostatic valve. Measurements for thermostatic valve where done with different settings and results are compared with parameters listed in the datasheet.

The study of selected sewer network in an urbanized catchment

This paper deals the case study of the chosen sewer system in the village Starý Mateřov. The study solves passportization of sewer, what is the base for creating technical condition mainly for three selected sections. According to the results from the technical conditions of the three selected sections were suggested suitable options of redevelopment. The options are compared in financial way. The last chapter suggests two possibilities how to deal with the sewage. The first option consists of the transferring waste water to the waste water treatment plant in Pardubice - Semtín. The second option suggests building a new waste water treatment plant in Starý Mateřov. The options are compared in financial way.

Optimal digital terrain model for erosion analysis

The thesis deals with the question of using available sources of elevation data for the evaluation of topographic factor in estimating the amount of water erosion in the area. Work compares the results with consideration to the origin of the data, the accuracy and resolution of the digital terrain model, which is interpolated from the source data. The calculation of topographic factor in ArcGIS will be confronted with the results of the Universal Soil Loss Equation (USLE). The frequency of compliance will tell us which data source and digital terrain model resolution is the most suitable for determining the topographic factor.

Vývoj metody na stanovení steroidních látek ve vodách

The theoretical part of the Master´s thesis describes steroids, their features, and the methods for their determination in waters. In experimental part three steroid hormones, namely 1,4-androstadiene-3,17-dione (ADD), 1,4-androstadiene-3-one-17(beta)-ol (boldenone) and 17(beta)-estradiol were selected. The compounds were concentrated from water by LLE and SPE extraction and determined by gas chromatography coupled to mass spectrometry. Several experiments were conducted to assess the recovery of specific extraction for studied compound.
